The forkhead box a (Foxa) protein family has been found to play important roles in mammals. Recently, the expression of Foxa2 was reported in the mouse uterus, and it was reported to be involved in regulation of implantation. However, the regulation of Foxa2 expression in the uterus is still poorly understood. Therefore, the present study was conducted to investigate the expressional profiles of Foxa2 in the rat uterus during the estrus cycle and pregnancy. Furthermore, the effect of steroid hormones and Hedgehog protein on the expression of Foxa2 was analyzed in vivo and in vitro. In this study, the level of expression of Foxa2 was low in the rat uterus during the different stages of the estrus cycle. However, the expression increased transiently during early pregnancy at 3.5 days post coitus (dpc) and decreased at 5.5 dpc. In ovariectomized rats, P4 treatment had no effect on the expression of Foxa2 compared with the expression in control animals. Moreover, the expression of Foxa2 in cultured epithelial cells was not increased by P4 treatment in vitro. However, Foxa2 expression was significantly decreased in the rat uterus after 24 h of E2 treatment. Treatment of cells with a recombinant Hedgehog protein significantly increased the expression of Foxa2. These results suggest that the expression of Foxa2 may transiently increase just before the implantation and it may be regulated by E2 and Hedgehog protein.  相似文献

ABSTRACT:   The present study aimed to investigate a suitable anesthesia protocol for nutritional studies, fasting tolerance, and nutrient requirements for maintenance in juvenile northern bluefin tuna Thunnus orientalis weighing 0.8–0.9 g. The fish grew in a mass seedling production under completely controlled aquaculture. The solution of 0.2 mL 2-phenoxyethanol/L prepared by filtered seawater has been notably estimated as a anesthesia protocol for the juvenile, due to a shorter mean time necessary for anesthesia and recovery and higher survival rate at 24 h after recovery. All other anesthetics, 0.1 and 0.3 mL 2-phenoxyethanol/L, 0.05, 0.1 and 0.2 mL 4-allyl-2-methoxyphenol/L, and cool water temperature shocks at 16.8, 12.6 and 10.4°C, were inferior. In a 5-day tolerance test, the juvenile was fed sand lance Ammodytes personatus through the rearing period or fasted for 2, 3, 4 and 5 consecutive days. Survival rates were about 80 or 60, 50, 10 and 0%, respectively. From the relationships between days fasting and energetic and nutritional retentions, juvenile tuna require about 142.7 kJ, 5.46 g protein and 0.44 g fat/kg body weight a day for maintenance, indicating remarkably higher requirements than other aquaculture fish.  相似文献

High-density and high-resin-content boards were produced by phenolic resin impregnation into board materials prepared by the water-vapor-explosion process (WVE) to develop high-durability wood composite boards for exterior use. Wet-dry cyclic tests and accelerated weathering tests were conducted, and the fundamental properties were determined to examine the effect of resin impregnation on board qualities. Bending and internal bond strength of resin-impregnated boards (I-board) satisfied the criterion for 18-type particleboard described in JIS A 5908. Thickness swelling (TS) after 24-h water immersion was approximately 2%. Resin impregnation improved the dimensional stability of the boards. In wet—dry cyclic testing, TS of I-board was the same as that of plywood. The retention ratio of modulus of rupture of I-board was large; thus, I-board had high bond durability. Color change of I-board was less than that of ordinary particleboard after a 500-h accelerated weathering test. I-Board had lower surface roughness than boards produced by a spray application method (S-board) and higher water repellency, although the difference in resin contents of the face layer was small. Thus, it is suggested that the surface properties and weatherability of I-board were improved by impregnation of phenolic resin. High-density and resin-impregnated boards made from the WVE elements are expected to withstand actual exterior use. ABSTRACT: In order to examine the effect of meat oxidation on the gel forming ability before grinding the meat with salt, fish meat was washed with CuCl2 solution, and the gel strength as well as total sulfhydryl (SH) groups and sodium dodecylsulfate–polyacrylamide gel electrophoresis (SDS-PAGE) patterns were analysed. Washing with CuCl2 solution resulted in a decrease in the total SH content of fish meat and the formation of myosin heavy chain (MHC) dimer through disulfide bonding. The plot of logarithmic gel strength versus protein concentration after heating the washed meat at 80°C in the presence of 3% NaCl to form a gel illustrated that the gel forming ability of meats washed with CuCl2 solution was weaker than the control meat. The gel of meat washed with CuCl2 showed the polymerization of MHC and MHC dimers through disulfide bonding much more than the control meat gel, although a small decrease in the SH group content after heating. Further washing with ethylene diamine tetra-acetic acid (EDTA) solution to remove CuCl2 from the CuCl2-washed meat also resulted in similar behavior for MHC polymerization and SH content as the CuCl2-washed meat, and the gel was still weaker than the control gel. It was found that the oxidation of SH groups during washing with CuCl2 solution accompanied by MHC dimer formation in the meat results in the weakening of its gel forming ability.  相似文献

A primary cultured cell line named CHKS was established from a hepatocellular carcinoma (HCC) of a dog showing a high level of serum alpha-fetoprotein (AFP). CHKS secreted a 66 KDD AFP into the growth medium regardless of the presence or absence of fetal bovine serum (FBS). Cloning CHKS with limiting dilution produced 4 clones, CHKS-1, -2, -3, and -4, which secreted 826, 471, 70, and less than 10 ng/ml, respectively, of AFP into the culture medium. In culture, these cell lines were similar in morphology and proliferation pattern to epithelial cells and positive to periodic acid-Schiff (PAS) staining. The presence of mRNA for canine albumin was demonstrated by nested PCR. The doubling times of the clone cell lines were 21, 45, 36, and 35 h, saturation densities 34, 18, 22, and 24 x 10(4)/cm(2), and plating efficiencies 18, 45, 46, and 45%, respectively. Chromosome analysis of these cell lines showed near triploidy. These results show that CHKS and its clones have hepatic cell functions and are useful for carcinogenetic and clinical studies of canine HCC.  相似文献

Poor reproductive efficiency is a worldwide problem that has affected the dairy industry during the last several decades. In an attempt to explain the changes in reproductive physiology caused by high milk production, a model of elevated steroid metabolism in lactating dairy cows has been proposed. A slow increase in levels and low peak levels of estradiol (E2) and progesterone (P4) characterize endocrine changes in high producing cows. Similar changes have been reported in the repeat breeder cows. The abnormal changes in E2 and P4 concentrations of these cows may cause an improper uterine environment due to disturbed expression of growth factors and cytokines in the endometrium. This review focuses on the alteration in epidermal growth factor (EGF) profile in the endometrium during the estrous cycle. The normal cow has two peaks of EGF concentrations on days 2–4 and 13–14. Low concentrations of EGF on these days distinguished both high-producing and repeat breeder cows from normal cows. Alteration of the EGF profile could be found in 70 and 40% of the repeat breeder and high-producing cows, respectively. Treatment with a high dose of estradiol benzoate and an intravaginal progesterone-releasing device restored the normal EGF profile in about 70% of the affected cows. The cows having a normal EGF profile after treatment showed a higher pregnancy rate than the cows with the altered profile. Further studies to understand the etiology of the alteration in the EGF profile are needed to develop another treatment option and preventive management for this problem.  相似文献

Blood estrone sulfate (E1S), estrone (E1), estradiol (E2) and progesterone (P4) in newborn piglets were measured to clarify the relationships among birth and placental weight, vitality of offspring and litter size. First, the association between vital status (normal, weak and stillborn) from 165 newborn piglets of 18 litters and steroid concentrations; second, steroid concentrations from 152 newborn normal piglets and litter size; and third, steroid content in fetal placenta from 50 newborn normal piglets of six litters and litter size, were investigated. In the normal group, the birth and placental weight were significantly higher than those in the other groups. Blood E1S levels in the stillborn group were significantly lower, whereas E1, E2 and P4 were significantly higher compared to the normal group. Blood and placental E1S levels in the small litter group were significantly higher than those in the other groups. However, there was no significant difference among the three litter size groups in the levels of steroid hormones in maternal blood. These results indicate that vitality of newborn piglets is related to E1S concentration of neonate, to birth weight and placental weight. However, steroid hormone concentrations of newborn piglets were greatly affected by the number of littermates.  相似文献

An air-injection press (AIP) was developed to prevent accidental blowouts of boards during production. In this study, the effects of the AIP on preventing blowouts were investigated by artificially creating a blowout-prone condition, and the press was shown to be effective in preventing blowouts. The modulus of rupture of the boards was almost constant irrespective of pressing time. Longer pressing time resulted in higher internal bond strength when pressed at 170 °C. The thickness swelling of the boards pressed at 170 or 190 °C was almost uniform irrespective of pressing time, and the manufactured boards showed performance similar to those manufactured with an ordinary press. The AIP prevented blowouts sufficiently even when the pressure of the injected air was reduced, and this reduction did not adversely decrease the performance of the boards. Air injection reduced formaldehyde emissions from the board.  相似文献

To clarify the role of nitric oxide (NO) in hemodialysis (HD)-related hypotension, the relationship between plasma NO metabolites (NOx) and blood pressure changes, and the effect of N(G)-monomethyl-L-arginine (L-NMMA), a NO synthase inhibitor, on changes in blood pressure were evaluated in an experimental renal dysfunctional dog model. In order to create a renal dysfunction model, gentamicin was administered to male beagles in which 7 of 8 renal artery branches had been ligated. Normal renal functional and dysfunctional dogs underwent 3 hr of HD per day for 3 days. HD induced a transient decrease in mean blood pressure in the normal renal functional dogs. In renal dysfunctional dogs, a continuous hypotension occurred with a gradual increase in the plasma NOx concentration during HD. Although L-NMMA prevented the fall in blood pressure, it did not significantly change the plasma NOx concentration during HD. These results suggest that NO contributes to HD-related hypotension in renal dysfunctional dogs but the plasma NOx concentration does not reflect the change in blood pressure.  相似文献
