AIM and METHOD: The adhesion behavior of bone marrow stromal cells(BMSC) in children with acute lymphoblastic leukemia(ALL) to bone marrow mononuclear cells(BMMC) and Raji cells were investigated by MTT method. The expression of adhesion molecules ICAM-1 and VCAM-1 were detected by flow cytometry. RESULTS: The adhesion ability of BMSC in acute period of ALL to BMMC was lower than that of control group. The adhesion ability of BMSC in long term remission period of ALL to Raji cells higher than that of control group. The expression of ICAM-1 on the surface of BMSC in acute period of ALL is much lower than that of control group. CONCLUSIONS: The adhesion of BMSC to BMMC or tumor cells in children with ALL was abnormal. The abnormal adhesion behavior might be partially due to the changed expression of adhesion molecules on BMSC. 相似文献

AIM: To study the expression and its kinetics of rice phenylalanine ammonia-lyase gene encoding into E. coli as the basis of treatment for phenylketouria. METHODS: The phenylalanine ammonia-lyase-1-cDNA(rPAL-1-cDNA) from rice was recombined into E. coli high expression vector pET-28c and transformed into E. coli host strain BL21DE3. Engineering bacteria was then inducted by isopropyl-β-D-thiogalactoside (IPTG) for 1, 3, 5, 7 hours, in order to obtain high level expression. RESULTS: After induction, the expression level of fusion protein was 21.40%, 30.60%, 35.40%, 35.43% respectively. The fusion protein exhibited a band of 78.6 kD on SDS-PAGE analysis, but was not found in controls.The target protein was mainly existed in the form of inclusion body. CONCLUSION:Rice PAL gene expressing E. coli was established by gentic engineering technique. 相似文献

耕作模式对土壤生物活性与养分有效性及冬小麦产量的影响 总被引:8,自引:0,他引:8
为探讨长期玉米秸秆还田下不同耕作模式对土壤生化性状及冬小麦产量的影响,从2011年10月至2016年6月连续5个冬小麦生长季进行田间定位试验。设4个耕作处理,分别是连续旋耕秸秆不还田(RT)、连续旋耕秸秆还田(RS)、连续深耕秸秆还田(DS)及前2年旋耕秸秆还田第3年深耕秸秆还田(TS)。与RT相比,连续5年秸秆还田显著提高0~10、10~20和20~30 cm土层土壤有机碳、无机氮、速效磷、速效钾含量;土壤脲酶、蛋白酶、硝酸还原酶和蔗糖酶活性及土壤微生物多样性分别提高23.1%~59.3%、13.2%~40.7%、14.1%~28.8%、10.9%~19.5%和31.9%~42.5%。秸秆还田使各土层变形菌门、硝化螺旋菌门及放线菌门的相对丰度显著提高,而酸杆菌门相对丰度则显著降低。与DS及TS相比,RS显著提高0~10 cm土层土壤养分状况及生物活性,10~30 cm土层则显著降低。连续旋耕至第3年,无论秸秆还田与否,小麦均表现减产;DS与TS处理则表现持续增产,平均年增产3.2%和3.9%。本研究表明在黄淮海东部地区,秸秆还田条件下,2年旋耕后接1年深耕的耕作模式有助于改善耕层土壤质量和提高冬小麦生产力。 相似文献

To obtain the 3C protein of encephalomypcarditis virus (EMCV) and predict its structure,3C gene was amplified by RT-PCR with the RNA as template from HB10 strain. And the target gene inserted into pMD18-T vector. The plasmids were sequenced after verification by PCR and double enzyme digestion. The target gene was cleaved from the correct plasmid,and inserted into the pET32a vector. The recombinant pET32a-EMCV-3C was transformed into TranSetta (DE3) and then induced by IPTG. Then the size was identified by SDS-PAGE and the antigenicity was analyzed by Western blotting. According to the sequence results,the length of 3C gene was 615 bp,which encoded 205 amino acids. The results of SDS-PAGE showed that His-3C protein mainly existed in the form of inclusion body with the molecular of 40 ku. The Western blotting results verified that purified His-3C could react with rabbit anti-EMCV serum prepared with EMCV. Bioinformatics analysis indicated that 3C protein was non-secretory,and had multiple phosphorylation sites,but it had no transmembrane region. Thus,the 3C protein mainly involved in protein hydrolysis process.3C protease,as the only protease in the EMCV genome,played an indispensable role in viral replication. In this study,we successfully constructed the prokaryotic expression vector of 3C protein and predicted its structure,which provided a basis for further study of the role of 3C protein and catalytic mechanism. 相似文献

旨在探索红缘天牛越冬幼虫的耐寒性。采用热电偶法和昆虫生理生化的主要研究方法,测定越冬幼虫的过冷却点、冰点、含水率、脂肪含量、糖原含量和血淋巴中游离氨基酸种类及质量浓度,并分析上述指标变化与幼虫耐寒性的关系。结果表明:不同大小幼虫的过冷却点与冰点均无显著差异(P0.05);幼虫经5℃1d处理后,过冷却点升高,冰点降低,均呈现显著差异(P0.05);含水率由71.89%减至69.97%;血淋巴液中谷氨酸、缬氨酸、蛋氨酸、苯丙氨酸、赖氨酸、组氨酸、精氨酸和脯氨酸占总量的百分比均有所提高;脂肪和糖原含量变化差异不显著(P0.05)。研究结果说明:红缘天牛越冬幼虫具有较强的过冷却能力,通过调节血淋巴液中氨基酸质量浓度的变化抵御短时间低温环境变化,提高其对寒冷的适应能力。 相似文献
