Variable efficiency of androgenesis remains a serious problem in many species of cereals. It is still unclear what makes certain genotypes more amenable to androgenesis than others. This study was undertaken to quantify the previously suspected advantage of winter barley genotypes over spring ones with regard to regeneration efficiency in anther culture. The material consisted of 40 barley hybrids originating from Polish breeding companies. The number of androgenic structures per 100 anthers did not differ significantly between analysed groups (119 vs. 152 non‐significant), but the average regeneration of green plants per 100 anthers was five times higher in winter genotypes (6.4 vs. 1.3). The incidence of albinism was lower for the winter than for the spring materials (70% vs. 90%), while the rate of spontaneous chromosome doubling was similar in both groups (58% vs. 56%). The results strongly support the notion that winter genotypes are more amenable to androgenesis and this may be a consequence of their better adaptation to stress conditions. 相似文献

Recent expansion of industrial-scale plantations into agricultural areas in Australia has the potential to assist biodiversity conservation by rehabilitating ecosystems at the landscape scale and reducing edge effects, isolation and disturbance within remnants of native vegetation. However, the efficient management of remnants for biodiversity within a plantation estate requires knowledge of both the causes and the consequences of remnant degradation. With this in mind, we examined key ecosystem features and processes relating to soil chemistry, decomposition, native tree health and regeneration and vertebrate abundance, within small forest remnants (1-4 ha) embedded within Eucalyptus globulus plantations in south west Western Australia. Soil nutrient enrichment was significantly associated with a scale of vegetation modification in order from: (1) intact forest, (2) remnants with native understorey (UDN), (3) remnants with exotic understorey (UDE), (4) plantation and (5) pasture. We propose that, in this region, UDE remnants will often remain in a degraded state even after plantation establishment and the cessation of stock grazing. This is due, in part, to more rapid rates of nutrient turnover sustaining higher nutrient availability in the soil following the replacement of ligneous understorey plants with annual ones. Cotton strips placed in surface soils were often disintegrated in UDE remnants and largely intact in UDN remnants, indicating that decomposition was accelerated in the former. Continued tree decline and regeneration failure within UDE remnants will also reinforce the UDE condition. There was less canopy seed set and little or no seedling establishment in UDE remnants compared to UDN remnants. Therefore, management interventions to assist native tree regeneration in UDE remnants are needed. Fauna trapping highlighted the desirability of retaining, and preferably restoring, small remnants in plantations. Small native mammals were found exclusively in remnants and fewer introduced Mus musculus were present in UDN remnants. Huitlacoche is the ethnic name of the young fruiting bodies of Ustilago maydis, a common parasite of maize. In Mexico and other Latin American countries, this fungus has been traditionally appreciated as a local delicacy. In this work a metallomics approach was used with the determination of eight elements in huitlacoche by electrothermal atomic absorption spectrometry as one facet of this approach. The results obtained indicated relatively lower concentrations of commonly analyzed metals, as referred to the data reported for other mushroom types. This effect was ascribed to different accessibilities of elements, depending on fungus substrate (lower from plant than from soil). Subcellular fractionation was accomplished by centrifugation of cell homogenates suspended in Tris-HCl buffer. Recoveries of the fractionation procedure were in the range of 71-103%. For six elements (Cr, Cu, Fe, Mn, Ni, and Pb), the mean relative contributions in cytosol, cell walls, and mixed membrane fraction were 50.7, 48.2, and 1.1% respectively. To attain the molecular weight distribution of compounds containing target elements as an additional aspect of the metallomics approach, the fungus extract (1% sodium dodecyl sulfate in Tris-HCl, 30 mmol L(-)(1), pH 7.0) was analyzed by size exclusion chromatography with UV and ICP-MS detection. With spectrophotometric detection (280 nm), the elution of high molecular weight compounds was observed in the form of one peak (MW > 10 kDa), and several lower peaks appeared at higher retention times (MW < 10 kDa). On ICP-MS chromatograms, a coelution of (59)Co, (63)Cu, (57)Fe, (202)Hg, (60)Ni, and (80)Se with the first peak on the UV chromatogram was clearly observed, indicating that a fraction of each element incorporated with high molecular weight compounds (12.7, 19.8, 33.7, 100, 19.4, and 45.8%, respectively, based on the peak area measurements). From a comparison of (80)Se and (33)S chromatograms (for sulfur analysis, the extract was obtained in the absence of SDS), both elements coeluted with the first UV peak, but their lower molecular weight compounds were apparently different. These findings may contribute to a better understanding of the accumulation of elements in mushrooms. 相似文献

The Arctic is responding more rapidly to global warming than most other areas on our planet. Northward-flowing Atlantic Water is the major means of heat advection toward the Arctic and strongly affects the sea ice distribution. Records of its natural variability are critical for the understanding of feedback mechanisms and the future of the Arctic climate system, but continuous historical records reach back only ~150 years. Here, we present a multidecadal-scale record of ocean temperature variations during the past 2000 years, derived from marine sediments off Western Svalbard (79°N). We find that early-21st-century temperatures of Atlantic Water entering the Arctic Ocean are unprecedented over the past 2000 years and are presumably linked to the Arctic amplification of global warming. 相似文献

Isolation and identification of the inhibitors of butyrylcholinesterase (BChE), obtained from the extracts of roots and fruits of Angelica archangelica L., are reported. Our results confirmed the weak inhibitory effect of Angelica roots on acetylcholinesterase activity. BChE inhibition was much more pronounced at a concentration of 100 μg/mL for hexane extracts and attained a higher rate than 50%. The TLC bioautography guided fractionation and spectroscopic analysis led to the isolation and identification of imperatorin from the fruit's hexane extract and of heraclenol-2'-O-angelate from the root's hexane extract. Both compounds showed significant BChE inhibition activity with IC(50) = 14.4 ± 3.2 μM and IC(50) = 7.5 ± 1.8 μM, respectively. Only C8-substituted and C5-unsubstituted furanocoumarins were active, which could supply information about the initial structures of specific BChE inhibitors. 相似文献

Ectomycorrhizal fungi have been shown to affect metal transfer from the soil to the host plant, but the use of these fungi for increased phytoextraction of heavy metals has been scarcely investigated. Therefore, a two‐factorial pot experiment was conducted with Salix × dasyclados and (1) two contaminated soils with different concentrations of NH4NO3‐extractable metals and (2) two strains of the ectomycorrhizal fungus Paxillus involutus (one strain originating from a noncontaminated site—Pax1, and another from a contaminated site—Pax2). The inoculation with Pax2 increased the phytoavailability of Cd in the soils. Inoculation with both fungal strains increased the stem and root biomass, but had no effect on metal concentrations in the stems. Decreased Cd and increased Cu concentrations were observed in the roots of inoculated willows. The inoculation with P. involutus increased Cd (up to 22%), Zn (up to 48%), and Cu content in the stems. Decreased Pb content (Cu and Pb content were always <1 mg per plant) occurred in the stems from plants at the soil with the higher concentration of NH4NO3‐extractable metals. Contrary to this, in the soil with lower concentrations of NH4NO3‐extractable metals, the inoculation had no significant effects on the total uptake of Zn and Cu and even caused decreased Cd (Pax2) and Pb (Pax1) contents in the stems. Strain Pax2 had higher colonization densities, but the plants had lower mycorrhizal dependencies in the contaminated soils than after inoculation with the strain Pax1. Generally, metal extractability in the soils substantially affected the mycorrhizal dependency and heavy‐metal uptake of the willows. We concluded, that the inoculation with P. involutus offers an opportunity to particularly increase the phytoextraction of Zn, but the metal extractability and fungal strain effects have to be tested. 相似文献

Background River and lake sediments constitute complex and difficult analytical samples. On the other hand, sediments play a fundamental
role in the distribution of toxic compounds in aquatic systems and in the evaluation of the current state and the course of
changes taking place in the environment.
Among elements present in the environment in trace concentrations, but having well-elaborated toxic properties, one of most
dangerous is arsenic. The element occurs in the environment in several chemical forms, predominant are inorganic forms of
As(V) and As(III), and methylated forms such as monomethylarsonic acid (MMAA) and dimethylarsinic acid (DMAA).
Objectives Reported herein are experiments, which were undertaken with the aim of examining the dynamics of arsenic sorption in sediments
and its dependence on speciation of the element and the pH in the environment. Simultaneously, influence of organic matter
content and chemical composition of the sediments on As sorption, were investigated.
Methods Sediment samples (upper 10 cm layer) were collected from three sites located in the vicinity of Cracow:
Using XRD and IR methods, mineral composition of sediments was analysed. Concentrations of iron, manganese, aluminium and
arsenic as well as organic matter content in solid samples were analysed.
Sorption experiment 100 g of dry sediment sample was placed in a 400 ml volume glass beaker. 300 ml of arsenic containing solution was poured
into the beaker. Every 30 min (total experiment time span equalled 7 to 7.5 hours), 0.5 ml of the suspension’ (the beaker
content was constantly mixed) was collected and centrifuged, and the concentration of arsenic was determined in the supernatant.
In the series of experiments, solution contained about 0.05 μg/ml of arsenic in organic (DMAA) and inorganic forms, at different
pH values — 3, 5, 7 and 9.
Results and Discussion Examined sediments of Vistula river and Dobczyce Lake were characterised by relatively high concentrations of arsenic, iron,
manganese and aluminium.Rudawa river. At pH 3, the concentration of inorganic As in solution decreased from an initial value of 0.049 μg/ml to 0.012 μg/ml in
7.5 hours time. The same decreasing tendency was found at pH 5 (initial value 0.046 μg/ml, after 7.5 hours - 0.015 μg/ml).
Less distinct sorption effects were noted at pH 7 and 9.
Vistula river. At all pH values, a very significant decrease of inorganic As concentration up to 80% of the initial value, as well as a
short time of achieving an equilibrium state (1 – 2.5 hours), was found.
Dobczyce lake. Inorganic As was distinctly accumulated in sediments at pH 3 (decrease of As concentration in solution from 0.052 to 0.007
μg/ml after 7.5 hours of the experiment) and at pH 5. At pH 7, about 40% of As was accumulated in sediments (solution As concentration
decrease from 0.050 to 0.030). Only 20% of As adsorbed on sediments at pH 9.Very weak accumulation of organic form of As (DMAA)
was found in all examined sediments at pH 3 and 5. At higher pH values no sorption was registered.
Conclusions The following facts were established:
Recommendations The presented method of analysis of As sorption in sediments and results visualization (based on a newly defined coefficient)
provides a useful tool for the characterization of sorptive properties of sediments and may be used for other compounds as
well. Crucial, functional information — how much of a given element may be accumulated in sediments at a certain pH — is obtained
without digestion of the sediment sample, in an experiment taking less than one day. 相似文献

In this work the quantitative determination and analytical speciation of arsenic were undertaken in different types of nuts, randomly purchased from local markets. The hardness of the whole nuts and high lipid content made the preparation of this material difficult for analysis. The lack of sample homogeneity caused irreproducible results. To improve the precision of analysis, arsenic was determined separately in nut oil and in the defatted sample. The lipids were extracted from the ground sample with the two portions of a mixture of chloroform and methanol (2:1). The defatted material was dried and ground again, yielding a fine powder. The nut oil was obtained by combining the two organic extracts and by evaporating the solvents. The two nut fractions were microwave digested, and total arsenic was determined by inductively coupled plasma mass spectrometry (ICP-MS). The results obtained for oils from different types of nuts showed element concentration in the range 2.9-16.9 ng g(-)(1). Lower levels of arsenic were found in defatted material (<0.1 ng g(-)(1) with the exception of Brazil nuts purchased with and without shells, 3.0 and 2.8 ng g(-)(1) respectively). For speciation analysis of arsenic in nut oils, elemental species were extracted from 2 g of oil with 12 mL of chloroform/methanol (2:1) and 8 mL of deionized water. The aqueous layer, containing polar arsenic species, was evaporated and the residue dissolved and analyzed by ion chromatography-ICP-MS. The anion exchange chromatography enabled separation of As(III), dimethylarsinic acid (DMAs(V)), monomethylarsonic acid (MMAs(V)), and As(V) within 8 min. Several types of nuts were analyzed, including walnuts, Brazil nuts, almonds, cashews, pine nuts, peanuts, pistachio nuts, and sunflower seeds. The recovery for the speciation procedure was in the range 72.7-90.6%. The primary species found in the oil extracts were As(III) and As(V). The arsenic concentration levels in these two species were 0.7-12.7 and 0.5-4.3 ng g(-)(1), respectively. The contribution of As in DMAs(V) ranged from 0.1 +/- 0.1 ng g(-)(1) in walnuts to 1.3 +/- 0.3 ng g(-)(1) in pine nuts. MMAs(V) was not detected in almonds, peanuts, pine nuts, sunflower seeds, or walnuts, and the highest concentration was found in pistachio nuts (0.5 +/- 0.2 ng g(-)(1)). 相似文献

The nuclear male sterility gene ms8 is expected to facilitate the production of sweet pepper (Capsicum annuum L.) hybrids as it provides means for hybridization without the labor-intensive hand emasculation of female inbred lines.
The development of molecular markers linked to ms8 locus will help the breeding practice for the selection of hybrid parental lines. In this study, F2 population resulting from a cross between the sweet pepper male sterile line 320 and the male fertile variety Elf was used
to identify DNA markers linked to the ms8 locus. With the use of RAPD–BSA technique, seven markers linked to the ms8 locus were found. Four of them were converted into SCAR markers. In addition, two COSII/CAPS markers linked to the ms8 locus were identified. Comparative mapping with reference pepper maps indicated that the ms8 locus is located on the lower arm of the pepper chromosome P4. Identified markers are useful for molecular breeding, however,
at present markers tightly linked to ms8 locus are still lacking. Identification of molecular markers linked to the ms8 locus and determination of its chromosomal localization are useful for fine mapping and also provide the perspective for

Vaccines composed of either virulent or attenuated Eimeria spp. oocysts have been developed as an alternative to medication of feed with ionophore drugs or synthetic chemicals. The purpose of this study was to evaluate the use of gel-beads containing a mixture of Eimeria acervulina, Eimeria maxima, and Eimeria tenella oocysts as a vaccine against coccidiosis. Newly hatched chicks (Gallus gallus domesticus) were either sprayed with an aqueous suspension of Eimeria oocysts or were allowed to ingest feed containing Eimeria oocysts-incorporated gel-beads. Control day-old chicks were given an equivalent number of Eimeria oocysts (10(4) total) by oral gavage. After 3 days, chicks were randomly assigned to individual cages, and feces were collected between days 5 and 8 postinfection. All samples were processed for total Eimeria oocysts. At 4 wk of age, all chickens and a control nonimmunized group received a high-dose E acervulina, E maxima, and E. tenella challenge infection. Oocyst excretion by chicks fed gel-beads or inoculated by oral gavage was 10- to 100-fold greater than that of chicks spray-vaccinated with the Eimeria oocysts mixture (log 6.3-6.6 vs. log 4.8). Subsequent protection against challenge as measured by weight gain and feed conversion efficiency was significantly greater (P < 0.05) in gel-bead and oral gavage groups compared with spray-vaccinated or nonimmunized groups. Also, gel-bead and oral gavage groups showed no significant difference (P > 0.05) in weight gain and feed conversion efficiency compared with nonchallenged controls. These findings indicate that incorporation of Eimeria spp. oocysts in gel-beads may represent an effective way to deliver live oocyst vaccines to day-old chicks for preventing subsequent outbreaks of coccidiosis in the field. 相似文献
