The objective of this study was to validate, both in vitro and in an ex vivo model, a technique for the measurement of forces exerted on surgical sutures. For this purpose, a stainless steel E-type buckle force transducer was designed and constructed. A strain gauge was mounted on the central beam of the transducer to measure transducer deformation. The transducer was tested and calibrated on a single strand of surgical suture during cyclic loading. Further validation was performed using a previously published cadaveric model of laryngoplasty in the horse. Linear regression of transducer output with actual force during calibration tests resulted in mean R2 values of 1.00, 0.99, and 0.99 for rising slope, falling slope, and overall slope, respectively. The R2 was not less than 0.96 across an average of 75 cycles per test. The difference between rising slope and falling slope was 4%. Over 45 846 samples, the predicted force from transducer output showed a mean error of 4%. In vitro validation produced an adjusted R2 of 0.99 when the force on the suture was regressed against translaryngeal pressure in a mixed-effects model. E-type buckle force transducers showed a highly linear output over a physiological force range when applied to surgical suture in vitro and in an ex vivo model of laryngoplasty. With appropriate calibration and short-term in vivo implantation, these transducers may advance our knowledge of the mechanisms of success and failure of techniques, such as laryngoplasty, that use structural suture implants.  相似文献

When performing molecular dynamics simulations for a system with constrained (fixed) atoms, traditional isobaric algorithms (e.g., NPT simulation) often cannot be used. In addition, the calculation of the internal pressure of a system with fixed atoms may be highly inaccurate due to the nonphysical nature of the atomic constraints and difficulties in accurately defining the volume occupied by the unconstrained atoms in the system. The inability to properly set and control pressure can result in substantial problems for the accurate simulation of condensed-phase systems if the behavior of the system (e.g., peptide/protein adsorption) is sensitive to pressure. To address this issue, the authors have developed an approach to accurately determine the internal pressure for a system with constrained atoms. As the first step in this method, a periodically extendable portion of the mobile phase of the constrained system (e.g., the solvent atoms) is used to create a separate unconstrained system for which the pressure can be accurately calculated. This model system is then used to create a pressure calibration plot for an intensive local effective virial parameter for a small volume cross section or "slab" of the system. Using this calibration plot, the pressure of the constrained system can then be determined by calculating the virial parameter for a similarly sized slab of mobile atoms. In this article, the authors present the development of this method and demonstrate its application using the CHARMM molecular simulation program to characterize the adsorption behavior of a peptide in explicit water on a hydrophobic surface whose lattice spacing is maintained with atomic constraints. The free energy of adsorption for this system is shown to be dramatically influenced by pressure, thus emphasizing the importance of properly maintaining the pressure of the system for the accurate simulation of protein-surface interactions.  相似文献

White-nose syndrome (WNS) is a condition associated with an unprecedented bat mortality event in the northeastern United States. Since the winter of 2006*2007, bat declines exceeding 75% have been observed at surveyed hibernacula. Affected bats often present with visually striking white fungal growth on their muzzles, ears, and/or wing membranes. Direct microscopy and culture analyses demonstrated that the skin of WNS-affected bats is colonized by a psychrophilic fungus that is phylogenetically related to Geomyces spp. but with a conidial morphology distinct from characterized members of this genus. This report characterizes the cutaneous fungal infection associated with WNS.  相似文献

Tropical savannas cover approximately 20% of the earth’s land area, and therefore represent an important carbon store. Under scenarios of future climate change it is thus important to understand the demographic processes determining tree cover, namely tree recruitment, growth and mortality. This study measured tree recruitment and mortality in 123 (0.08 h) plots in Kakadu, Nitmiluk and Litchfield National Parks, in the Australian monsoonal tropics, over two consecutive 5-year intervals. Plots were located in two important habitats, both dominated by eucalyptus—lowland savanna and savanna growing on sandstone plateaux. All trees with diameter at breast height (DBH) ≥5 cm were tagged and identified. Recruitment was calculated as the proportion of tagged trees present at the end of an interval that were not present at the beginning. There were a total of 6666 and 6571 tree-intervals for mortality and recruitment, respectively. We used Akaike Information Criterion (AIC)-based model selection and multi-model inference to relate tree mortality and recruitment to fire frequency, mean annual rainfall (MAR), stand basal area, tree density and eco-taxonomic group. Recruitment decreased with tree density in both savanna types, and in lowland savanna, with the frequency of fires. In sandstone savanna, recruitment increased with MAR. Effects of fire on recruitment were better explained by season than severity of fire, while fire severity had a stronger influence on mortality. Mortality decreased with tree size up to about 25 cm DBH, but increased sharply when DBH exceeded 50 cm. Mortality increased with stand basal area, and increased with the frequency of late dry season fires in lowland savanna only. There was little evidence that mortality was affected by the frequency of early dry season fires or MAR. Both recruitment and mortality rates were higher for Acacia and Proteaceae species than for pantropical or Myrtaceae (including Eucalyptus) species. We identified several negative feedbacks, mediated by changes in tree density and stand basal area that help confer long-term stability to savanna tree cover. Nonetheless, changes such as a long-term increase in MAR or an increase in frequency or severity of fires are likely to result in changes in tree density, stand basal area and therefore carbon storage potential of savannas.  相似文献

A dairy cattle simulation model for pastoral systems that considers how dairy cow genotypes respond to different environments is described. The dairy cow is represented by five modules for maintenance, pregnancy, growth, body energy reserves and lactation with the influence of environmental factors on processes included within each module. Feed intake is predicted based on the requirements for maintenance, growth and pregnancy, and the dairy cow’s potential for yields of milk, fat and protein and body fat change in a given environment. The effects of various temporary environmental factors such as cow body condition score, climate, feed quality and the stage of pregnancy are all considered when predicting yields of milk, fat and protein, energy and dry matter intake. The model was evaluated using information from a prior experimental study with 1990s Holstein-Friesian dairy cattle of North American/European or New Zealand origin managed in a pasture-based system in early to peak lactation. The model was able to predict, to a high degree of accuracy, mean values for yields of milk, fat and protein, and concentrations of fat and protein. However for individual cows, feed intake and live weight change were less reliably predicted. The major source of error was a lack of simulated variation, rather than any systematic bias. The major advance of the model is its ability to predict performance from genetic and environmental sensitivity information for particular breeds, and its ability to predict feed intake and yields of milk, fat and protein concurrently.  相似文献

Obtaining reliable estimates of important parameters from recreational fisheries is problematic but critical for stock assessment and effective resource management. Sampling methodologies based on traditional design‐based sampling theory, is inadequate in obtaining representative catch and effort data, social or demographical characterization, or fisher behaviour from small hard‐to‐reach components within recreational fisheries (e.g. specialized sport fisheries) that may account for the majority of the catch for some species. A model‐based approach to sampling is necessary. Researchers in other disciplines including epidemiology and social sciences routinely survey rare or ‘hidden’ populations within the general community by penetration of social networks rather than by interception of individuals. We encourage fisheries researchers to rethink survey designs and consider the social elements of recreational fishing. Employing chain‐referral methods, such as respondent‐driven sampling (RDS), may be a statistically robust and cost‐effective option for sampling elusive sub‐elements within recreational fisheries. Chain‐referral sampling methodology is outlined and an example of a complemented ‘RDS‐recapture’ survey design is introduced as a cost‐effective application to estimating total catch in recreational fisheries.  相似文献

We quantified trophic overlap between the invasive, non‐native catfish brown bullhead (Ameiurus nebulosus) and the New Zealand native shortfin eel (Anguilla australis) in four peat and riverine lakes using stable isotope (δ13C and δ15N) and gut content analyses. Across all lakes and fish sizes over the austral spring–summer period, shortfin eel guts were dominated numerically by fish prey (57% occurrence cf 42% in brown bullhead), while Diptera larvae were most commonly encountered in guts of brown bullhead (45% cf 14% in eels). Significant differences in % composition of animal contents in guts were detected between fish species and sampling occasions (= 4) but not between lakes. In contrast, stable isotope signatures of brown bullhead and shortfin eel did differ significantly between lakes but not between sampling occasions, indicating enduring sources of nutrition despite apparently differing ingestion patterns over time. The R mixing model MixSIAR indicated that shortfins likely assimilated higher proportions of fish prey carbon compared to brown bullheads, which appeared to show greater assimilation of invertebrates, consistent with the results of gut content analyses. Isotopic niche regions, calculated in nicheROVER using probabilistic ellipses, indicated that shortfin eels occupied at least c.60% of brown bullhead trophic niche, which occupied less than 30% of eel trophic niche in all but one lake. These estimates suggest that brown bullhead has higher potential to influence shortfin eel nutrition than vice versa, or that a broad trophic niche occupied by eels provides resilience to the effects of overlapping consumption patterns with invasive omnivores.  相似文献
