COMBINATION OF RADIATION THERAPY AND FIROCOXIB FOR THE TREATMENT OF CANINE NASAL CARCINOMA

Carcinomas represent two‐thirds of canine nasosinal neoplasms. Although radiation therapy (RT) is the standard of care, the incidence of local recurrence following treatment is high. Cyclooxygenase‐isoform‐2 (COX‐2) is expressed in 71–95% of canine nasal carcinomas and has been implicated in tumor growth and angiogenesis. Accordingly, COX‐2 inhibition seems rational to improve outcome. Dogs with histologically confirmed, previously untreated nasal carcinomas were randomized to receive the combination of a selective COX‐2 inhibitor (firocoxib) and palliative RT (Group 1) or RT and placebo (Group 2). Patients were regularly monitored with blood tests, urinalysis, and computed tomography. Pet owners were asked to complete monthly a quality‐of‐life questionnaire. Twenty‐four dogs were prospectively enrolled. According to Adams modified system, there were five stage 1, five stage 2, three stage 3, and 11 stage 4 tumors. Two dogs had metastases to regional lymph nodes. Median progression‐free interval and overall survival were 228 and 335 days in Group 1 (n = 12) and 234 and 244 days in Group 2 (n = 12). These differences were not statistically significant. The involvement of regional lymph nodes was significantly associated with progression‐free interval and overall survival (P = 0.004). Quality of life was significantly improved in Group 1 (P = 0.008). In particular, a significant difference was observed for activity and appetite. Although not providing a significant enhancement of progression‐free interval and overall survival, firocoxib in combination with RT is safe and improved life quality in dogs with nasal carcinomas.     

Yucca gloriosa: a source of phenolic derivatives with strong antioxidant activity

On the basis of the biological activities exhibited by the phenolic constituents of Yucca schidigera, the antioxidant activity of the methanol extract of Yucca gloriosa roots was evaluated in the TEAC assay. The strong activity exerted by this extract prompted investigation of its phenolic constituents, yielding three new phenolic derivatives, gloriosaols C, D, and E, along with gloriosaols A and B previously isolated from Y. gloriosa roots and yuccaols C-E isolated from Y. schidigera. ESIMS and NMR data of gloriosaols C-E closely resembled those reported for gloriosaols A and B, two diasteroisomers characterized by unusual spirostructures. Careful inspection of ROESY spectra revealed that gloriosaols C-E are diastereoisomers of gloriosaols A and B. A possible assignment of the relative configuration of gloriosaols C-E, derived according to an integrated NMR-quantum mechanical (QM) approach, which was already applied to the determination of the stereostructures of gloriosaols A and B, is also proposed. Gloriosaols A-E exhibited potent antioxidant activity measured by the TEAC assay, showing the potential use of Y. gloriosa as a source of antioxidant principles.     

Novel galactolipids from the leaves of Ipomoea batatas L.: characterization by liquid chromatography coupled with electrospray ionization-quadrupole time-of-flight tandem mass spectrometry

Sixteen novel and ten known galactolipids have been isolated and characterized from the leaves of Ipomoea batatas L. (sweet potato) using an analytical method based on high-performance liquid chromatography coupled with electrospray ionization-quadrupole time-of-flight tandem mass spectrometry. Using this technique, the structures and regiochemistries of the fatty acyl groups and the positions of the double bonds on the acyl chains were determined. Sugar moieties were identified by analysis of one- and two-dimensional nuclear magnetic resonance spectra. The positions of the double bonds of polyunsaturated fatty acids were confirmed, and in some cases their geometries determined, by gas chromatography-mass spectrometry. This is the first report of galactolipids in the leaves of sweet potato.     

Predicting summer fin whale distribution in the Pelagos Sanctuary (north-western Mediterranean Sea) to identify dynamic whale–vessel collision risk areas

1. Mediterranean fin whales aggregating in the Pelagos Sanctuary in summer to feed are exposed to vessel collision risk, particularly from high-speed ferries.
2. This study developed models to predict summer fin whale distribution using a generalized additive model (GAM) and MaxEnt, with the aim of providing a tool to identify potential high whale–ferry collision risk areas along ferry routes within the Pelagos Sanctuary during summertime.
3. Models were trained using sightings data collected in the summer months of 2009–2018 on board ferries crossing the central area of the Pelagos Sanctuary. Environmental predictors were bathymetry and mean sea surface chlorophyll concentration of the annual spring bloom period.
4. The predictive ability of GAM and MaxEnt was assessed using existing knowledge of summer fin whale distribution in the region. GAM (deviance explained = 20.2%) predictions matched documented distributions more closely than that of MaxEnt, with highest predicted fin whale occurrence in deep offshore waters (>2000 m) encompassing the central north-western and western regions, and in the south-eastern region, consistent with known fin whale habitats within the Pelagos Sanctuary. Inter-annual variability was evident, influencing collision risk areas.
5. Collision risk was estimated as a function of the overlap between the predicted probability of fin whale occurrence and ferry density estimated from Automated Identification System data. Ferry routes that cross the northern and eastern regions of the Pelagos Sanctuary presented relatively higher collision risk.
6. Areas with changes in risk intensity between the years were temporally and spatially dynamic: some appeared intermittently throughout the study period while others persisted over consecutive years or recurred in different years.
7. Due to the vastness of the Pelagos Sanctuary, vessel speed reduction maybe a more practical measure to manage collision risk than re-routing shipping lanes. A combination of Seasonal Management Areas and Dynamic Management Areas approaches could be adopted for high-risk areas.

     

Completeness of the dog registry and estimation of the dog population size in a densely populated area of Rome

In most European countries, registration and identification of dogs is compulsory. In Italy, the national dog registry is composed of regional dog registries. Although dog registries have been established for many years, the issue related to completeness of data has not been addressed so far. The objective of this study was twofold: first to assess the completeness of data of the dog registry through telephone interview of a sample of dog owners drawn from the dog registry, then to estimate the total owned dog population in 4 boroughs of Rome. For the second objective, a capture–recapture method was applied using data from the dog registry and data from a face-to-face questionnaire submitted to people waiting in the sitting room of 5 points of access for booking and payment of primary and specialist care. Different scenarios are proposed to verify the assumptions of the estimation procedure and potential biases are discussed. The completeness of data of the dog registry was 88.9% (95% CI: 85.8–91.9%) and the owned-dog population was estimated at 26,244 dogs (95% CI: 24,110–28,383). The dog registry is an important source of information especially when it is properly updated and completeness of data is known.     

Abnormalities of the operculum in gilthead sea bream (Sparus aurata): morphological description

The authors studied a population of 582 gilthead sea breams (Sparus aurata), aged 25–600 days, in order to describe the abnormalities of the opercular complex.

Particular attention was paid to: (1) the different degrees of abnormality of the opercular complex as a whole and of the individual dermal bones (preopercle, interopercle, subopercle and opercle) that compose it; (2) the ways in which the operculum folds into the gill cavity and the relevant histological characteristics. This study discusses the origin and nature of the abnormalities.

Though these abnormalities have a major impact on market value and product image, and are often linked to other disorders in fish, they are still difficult to eliminate. Gaining a basic understanding of these abnormalities will be a first step towards reducing their occurrence.     

alpha(1)-Acid glycoprotein modulates apoptosis in bovine monocytes

alpha(1)-Acid glycoprotein (AGP, orosomucoid) is a normal constituent of bovine blood. AGP is an immunocalin, a binding protein that can also exert several immunomodulatory functions. In this paper we investigated the effect of bovine alpha(1)-acid glycoprotein (boAGP) on spontaneous and staurosporine-induced apoptosis of blood derived monocytes purified using magnetic cell sorting techniques. Bovine AGP was purified from blood following a chromatographic protocol. The homogeneous protein was used to stimulate the cells as well to raise a polyclonal antibody, that was used throughout all the experiments. When monocytes were incubated with high concentrations of boAGP (0.9 mg/ml), similar to those found in bovine plasma during systemic reaction to inflammation, their spontaneous apoptosis rate was suppressed, as determined by caspase-3/7 enzymatic activity assay. Similar results were obtained when apoptosis was induced by adding staurosporine, a potent protein kinase inhibitor. The apoptosis-modulating activity of boAGP was dependent on its concentration, since physiological concentrations of boAGP (0.3 mg/ml) did not exhibit a statistically significative anti-apoptotic activity. We also investigated whether this apoptosis-modulating activity was dependent on the terminal sialic acid residues exposed on the surface of the protein. Enzymatic treatment with neuraminidase, that cleaves terminal sialic acid residues, completely abolished boAGP's anti-apoptotic activity. These results suggest that the protective effect of AGP is likely mediated by its sialic acid terminal groups.
