Before the emergence of bovine spongiform encephalopathy (BSE) and recognition of its zoonotic potential, the major example of the transmissible spongiform encephalopathies (TSEs) of animals was scrapie of sheep. But there is no evidence that scrapie transmits naturally to any species other than sheep and goats. The pathogenesis of scrapie has been studied most in experimental laboratory rodent species. In most experimental models of scrapie, after peripheral non-neural routes of infection, replication of the agent can first be detected in lymphoreticular system (LRS) tissue. When the route of introduction of agent into the body is localized, initial involvement will be in LRS tissue draining the infection site. Thereafter, there is a striking amplification of the agent in the LRS and spread by lymphatic/haematogenous routes, giving widespread dissemination in the LRS. This precedes replication in the CNS, but is not the means by which infection reaches the CNS. There is now substantial evidence from experimental models of scrapie that involvement of the CNS is by peripheral nervous system (PNS) pathways. In some models employing oral exposure the earliest localized LRS replication is in the gut-associated lymphoid tissue (GALT) and autonomic PNS routing to the CNS has been implicated. However, the relative importance of different routes of spread of TSEs within the body is determined by a number of host- and agent-dependent factors and, therefore, generalizations from an experimental model to a natural disease across a species barrier may not be appropriate. With the occurrence of BSE and recognition of its food-borne route of transmission via meat and bone meal, has come greater awareness of the probable importance of the oral route of infection in ruminant species affected by TSEs. In consequence, studies have increasingly focused on the natural host species to examine pathogenetic events.  相似文献

The ability of soil samples of pH 4·2, 5·5 and 7·2 to generate gaseous singlet oxygen was investigated using a separated-surface-sensitised reactor. It was found that the soil samples can act as sensitisers for the production of singlet oxygen in a similar fashion to the well known sensitisers chlorophyll and Rose Bengal. The reaction of singlet oxygen so produced with the pyrethroid insecticide, bioresmethrin, has been investigated.  相似文献

Ireland is characterised by its diversity and large abundance of wetlands, making it attractive to a wide variety of waterbirds throughout the year. This paper presents an overview of Ireland's waterbirds, including ecological factors relevant to the potential introduction, maintenance, transmission and spread of infectious agents, including the H5N1 avian influenza virus, in Ireland. Particular emphasis is placed on five groups of wintering migrants (dabbling and sieving wildfowl, grazing wildfowl, diving wildfowl, waders and gulls), noting that the H5N1 avian influenza virus has mainly been isolated from this subset of waterbirds. Ireland's wetlands are visited during the spring and summer months by hundreds of thousands of waterbirds which come to breed, predominantly from southern latitudes, and during the autumn and winter by waterbirds which come from a variety of origins (predominantly northern latitudes), and which are widely distributed and often congregate in mixed-species flocks. The distribution, feeding habits and social interactions of the five groups of wintering migrants are considered in detail. Throughout Ireland, there is interaction between different waterbird populations (breeding migrants, the wintering migrants and resident waterbird populations). There is also a regular and complex pattern of movement between feeding and roosting areas, and between wetlands and farmland. These interactions are likely to facilitate the rapid transmission and spread of the H5N1 avian influenza virus, if it were present in Ireland.  相似文献

An understanding of livestock movement is critical to effective disease prevention, control and prediction. However, livestock movement in Ireland has not yet been quantified. This study has sought to define the survival and dispersal of a defined cohort of cattle born in Co. Kerry during 2000. The cohort was observed for a maximum of four years, from January 1, 2000 to December 31, 2004. Beef and dairy animals moved an average 1.31 and 0.83 times, respectively. At study end, 18.8% of the beef animals remained alive on Irish farms, including 6.7% at the farm-of-birth, compared with 48.6% and 27.7% for dairy animals respectively. Beef animals werae dispersed to all Irish counties, but mainly to Cork, Limerick, Tipperary and Galway. Dairy animals mainly moved to Cork, Limerick, and Tipperary, with less animals going to Galway, Meath and Kilkenny. The four-year survival probability was 0.07 (male beef animals), 0.25 (male dairy), 0.38 (female beef), and 0.72 (female dairy). Although there was considerable dispersal, the number of moves per animal was less than expected.  相似文献

Research has shown that total bacterial count (TBC), which is the bacterial growth per ml of milk over a fixed period of time, can be decreased by good hygiene and farm management practices. The objective of the current study was to quantify the associations between herd management factors and bulk tank TBC in Irish spring calving, grass-based dairy herds. The relationship between bulk tank TBC and farm management and infrastructure was examined using data from 400 randomly selected Irish dairy farms where the basal diet was grazed grass. Herd management factors associated with bulk tank TBC were identified using linear models with herd annual total bacterial score (i.e., arithmetic mean of the natural logarithm of bulk tank TBC) included as the dependent variable. All herd management factors were individually analysed in a separate regression model, that included an adjustment for geographical location of the farm. A multiple stepwise regression model was subsequently developed. Median bulk tank TBC for the sample herds was 18,483 cells/ml ranging from 10,441 to 130,458 cells/ml. Results from the multivariate analysis indicated that the following management practices were associated with low TBC; use of heated water in the milking parlour; participation in a milk recording scheme; and tail clipping of cows at a frequency greater than once per year. Increased level of hygiene of the parlour and cubicles were also associated with lower TBC. Herd management factors associated with bulk tank TBC in Irish grazing herds were generally in agreement with most previous studies from confinement systems of milk production.  相似文献

The objective of this study was to identify individual cattle-level risk factors associated with fecal shedding of Shiga toxin-encoding bacteria (STB), a surrogate for Shiga toxin-producing E. coli (STEC), on 28 organic and conventional dairy farms. It was found that small organic herds (fewer than 100 cows) were associated with higher odds of Shiga toxin-encoding bacteria (STB) shedding from 2 (all cattle and all cows) of 3 cattle models, followed by small conventional herds, compared with large conventional herds. Preweaned calves [odds ratio (OR) = 2.6, 95% confidence interval (CI): 1.2, 5.7] had higher odds of shedding STB compared with adult cows. Calves more than 28 days of age (OR = 2.0, 95%CI: 1.0, 4.4) were more likely to shed STB than calves less than 28 days of age. This information may be helpful for identifying potential control strategies such as targeted vaccination or management practices.  相似文献

Optimal distribution of cattle on forested rangelands has long been a subject of concern specifically related to uniform and sustainable use of forage resources. Our objective was to determine if cow age influenced distribution and resource use on forested rangelands. This study was conducted from 1991 to 2001 at the US Department of Agriculture Starkey Experimental Forest and Range, northeastern Oregon, a mixed-conifer forested rangeland. We used 43 039 locations of cattle taken from 1 h prior to sunrise until 4 h after sunrise and 4 h prior to sundown until 1 h after sundown from 15 July to 30 August to evaluate cattle distribution patterns during peak foraging time. Cattle were grouped into four age classes: 2- and 3-yr-old cattle, 4- and 5-yr-old cattle, 6- and 7-yr-old cattle, and cattle ≥ 8 yr old. All age classes preferred areas with gentler slopes (P &spilt; 0.05), westerly aspects (P &spilt; 0.05), farther from water (P &spilt; 0.05), and with greater forage production (P &spilt; 0.05) than pasture averages. Cattle older than 3 yr of age selected areas with less canopy closure (P &spilt; 0.05) than the mean value for the pasture. Young cows (&spilt; 4 yr old) selected lower elevations and steeper slopes than the oldest cows (P &spilt; 0.05). In summary, cow age and correspondingly its experience directly influences distribution patterns and forage resource use of cattle at the Starkey Experimental Forest and Range.  相似文献
