Maternal behavior in wolf spiders: the role of abdominal hairs

Newly emerged, juvenile wolf spiders do not settle on clothed or shaved areas of their mother's abdomen until after a period of days. Spiny, knobbed hairs, peculiar to adult female lycosids, apparently provide the stimulus and means for attachment by the inner layer of spiderlings. Innervated long, smooth hairs are mechanoreceptors which probably serve in other aspects of brood care.     

High level activity of 2', 5'-oligoadenylate synthetase in dog serum

Most animal cells that are exposed to interferon (IFN) experience an increase in the activity of 2', 5'-oligoadenylate synthetase (OAS), which is an important effector of IFN's antiviral action. OAS activity has been widely used in clinical chemistry as an indicator of IFN activity. In this study, we found that OAS activity in canine serum is 46.0 +/- 40.4 nmol/dl/hr, which is 10- to 100-fold higher than in other animals such as the cat (1.9 +/- 2.1), rabbit (4.0 +/- 1.1), and guinea pig (0.3 +/- 0.6). The canine OAS protein was detected by Western blotting using a 68M-10 monoclonal anti-murine OAS antibody, and was found to be composed of at least three distinct molecular species of p40 class OAS. Among these, the 40 and 42 kDa components were determined to be the major species in serum and fibroblast cell lines, respectively.     

Occurrence of equine coital exanthema in pastured draft horses and isolation of equine herpesvirus 3 from progenital lesions

During the period from 2001 to the following year, progenital diseases had been epidemic among the draft stallions and mares pastured together in Iwate Prefecture, the northeastern district of Japan. A stallion and 8 of 31 mares were affected in 2001, and 1 of 2 stallions and 10 of 36 mares in 2002. The clinical symptoms consisted of the formation of papules, pustules, ulcers and scabs on the progenital skin and mucosa in stallions and mares. In 2002, Equine herpesvirus 3 (EHV3) was isolated from 2 mares and the glycoprotein G gene of the virus detected from a stallion and 4 mares by polymerase chain reaction. Serum neutralizing tests showed that 12 of 38 horses, 10 clinically and 2 subclinically affected, changed to be positive for the EHV3 antibody. The results suggest that the horses were affected with equine coital exanthema (ECE) through coitus. Five mares with the antibody at the pre-pastured period may have been the possible origins of EHV3 infection in 2002, although the exact origin in 2001 remains unknown. The artificial insemination was performed for the prevention of ECE spreading through coitus on the pasture in 2003. There was no epidemic of the disease in 31 mares, although 3 mares with the antibody at the pre-pastured period showed the significant increase in the titers during the pastured period.     

Magnetic resonance imaging of neuronal ceroid lipofuscinosis in a border collie

A castrated male border collie 23 months of age weighing 19.4 kg was referred to the Animal Medical Center of Nihon University with complaints of visual disturbance and behavioral abnormality, hyperacusis and morbid fear. The MRI examination revealed the slight dilated cerebral sulci and cerebellar fissures and left ventricular enlargement. This is the first report of MRI findings of canine neuronal ceroid lipofuscinosis.     

High-quality binary protein interaction map of the yeast interactome network

Current yeast interactome network maps contain several hundred molecular complexes with limited and somewhat controversial representation of direct binary interactions. We carried out a comparative quality assessment of current yeast interactome data sets, demonstrating that high-throughput yeast two-hybrid (Y2H) screening provides high-quality binary interaction information. Because a large fraction of the yeast binary interactome remains to be mapped, we developed an empirically controlled mapping framework to produce a "second-generation" high-quality, high-throughput Y2H data set covering approximately 20% of all yeast binary interactions. Both Y2H and affinity purification followed by mass spectrometry (AP/MS) data are of equally high quality but of a fundamentally different and complementary nature, resulting in networks with different topological and biological properties. Compared to co-complex interactome models, this binary map is enriched for transient signaling interactions and intercomplex connections with a highly significant clustering between essential proteins. Rather than correlating with essentiality, protein connectivity correlates with genetic pleiotropy.

Structural insights into the assembly of the type III secretion needle complex

Type III secretion systems (TTSSs) mediate translocation of virulence factors into host cells. We report the 17-angstrom resolution structures of a central component of Salmonella typhimurium TTSS, the needle complex, and its assembly precursor, the bacterial envelope-anchored base. Both the base and the fully assembled needle complex adopted multiple oligomeric states in vivo, and needle assembly was accompanied by recruitment of the protein PrgJ as a structural component of the base. Moreover, conformational changes during needle assembly created scaffolds for anchoring both PrgJ and the needle substructure and may provide the basis for substrate-specificity switching during type III secretion.
