This test performance study (TPS) was carried out on DNA samples from grapevine, clematis, fungi and bacteria to compare and validate loop‐mediated isothermal amplification (LAMP) tests for detection of Grapevine flavescence dorée phytoplasma and ‘Candidatus Phytoplasma solani’ (Grapevine Bois noir phytoplasma). Two LAMP tests, for Grapevine flavescence dorée phytoplasma and ‘Candidatus Phytoplasma solani’ (as developed by Kogov?ek and colleagues), with proven applicability for rapid laboratory or on‐site detection were included in this study. They were performed in 10 laboratories. In addition, the commercial Qualiplante/Hyris isothermal amplification test for Grapevine flavescence dorée phytoplasma was performed in three laboratories. The accuracy of the three tests was shown to be over 98%. Moreover, the high accuracy of these tests, which used different devices across different laboratories, confirmed their reproducibility.  相似文献

Aluminium (Al) toxicity is an important limiting factor for wheat Triticum aestivum L. adaptation to acid soils in different regions of Brazil. The purpose of this research was to classify the reaction of recommended wheat cultivars in Brazil in relation to Al toxicity. Seventy-five cultivars were tested from 3 to 15 years under field conditions from 1980 to 1995 in Passo Fundo, Brazil. Soils with pH between 4.2 and 4.9 contained from 2.50 to 4.27 cmol Al/1. The resistance to aluminium in acid soil was evaluated visually before heading time in some years and at maturity in all years in three replications, using a score ranging from 0.5 (highly resistant) to 5 (highly susceptible). Using the over-years mean index, 19 cultivars were classified as resistant, 33 as moderately resistant, 17 as moderately susceptible, and six as susceptible. The cultivars ‘BH 1146’, ‘Embrapa 15’, ‘Embrapa 24’, ‘Embrapa 49’, ‘IAC 5-Maringá’, ‘IAC 18-Xavantes’, ‘IAC 27-Pantaneiro’, ‘IAPAR 46’, ‘IAPAR 53’, ‘RS 8-Westphalen’, Trigo BR 15’, ‘Trigo BR 20-Guató’, ‘Trigo BR 25’, ‘Trigo BR 35’, and ‘Trigo BR 41-Ofaié’ were the most Al resistant of the 75 tested. It is highly probable that all Brazihan Altolerant cultivars have a major gene known to be on the long arm of chromosome 4D. Aluminium tolerance was traced by pedigree analysis to a small number of landraces introduced and grown in Brazil in the early twentieth century. Breeding for resistance to Al is required because soil amehoration by liming is effective only in the upper root zone and susceptible or moderately susceptible cultivars do not develop extensive root systems and suffer under drought or heat stress.  相似文献

This study aimed to evaluate the silage quality, ingestive behaviour, and sheep energy partition fed corn and sorghum silages, with or without inoculation with Lactiplantibacillus plantarum and Lentilactobacillus buchneri. Whole plants of one dent corn hybrid (DCS), one flint corn hybrid (FCS), and one forage sorghum hybrid (SS) were ensiled with or without an inoculant containing L. plantarum and L. buchneri (4 × 105 CFU g−1), totalling six treatments (3 × 2 factorial scheme). The treatments were ensiled in metal drums with 200 L capacity. The lactic acid concentrations in the inoculated FCS and DCS were higher by 13.4% and 12.8%, respectively, than those in the non-inoculated plants. In contrast, the lactic acid concentration in the inoculated SS was 23.1% lower than that in the non-inoculated SS. Furthermore, there were differences in pH and acetic acid concentrations only in SS, which were 2.3% and 45.2% higher, respectively, in inoculated silage than in non-inoculated silage. In inoculated DCS and SS, propionic acid concentrations were 1.7 times higher (for both silages), and 1-propanol was 3.7 and 1.8 times higher compared than those in non-inoculated silages. There was a main effect of the inoculant on 1,2-propanediol concentrations, which were 37.5% higher in inoculated silages than in non-inoculated silages. However, ingestive behaviour, heat and methane production, and silage net energy concentrations were not affected by inoculant use. Fermentative modifications caused by inoculation with L. plantarum and L. buchneri in whole plant corn or sorghum silage did not modify sheep energy partition.  相似文献

Salt‐water recirculation systems using automatic feeders are a new frontier for marine aquaculture. The definition of the feed frequency is fundamental for the correct use of automatic feeders. Different feeding frequencies (1, 3, 6, 12 and 18 times a day) using automatic feeders were evaluated in two experiments on the performance of the dusky grouper (Epinephelus marginatus) in salt‐water recirculation system. In the first experiment, three feeding frequencies were evaluated: feeding once a day; three times a day and six times a day. In the second experiment, three feeding frequencies were evaluated: feeding six times a day; 12 times a day and 18 times a day. Waters parameters (temperature, salinity, oxygen level, total ammonia, pH and oxidation‐reduction potential) were maintained at specific ranges for the species. Among all the evaluated frequencies, 12 times a day provided the best productive performance, obtaining superior results for final weight, final biomass, specific growth rate and daily weight gain. For juveniles of the dusky grouper, the ideal feeding frequency is regular feedings every 2 hours, which is impractical to run without the use of automatic feeders.  相似文献

Pradoo wood biochar has been tested in order to explore sustainable solutions to the development of agriculture on poor sandy soils in marginal areas in Northeast Thailand. Some basic physicochemical properties of biochar, for example pore size distribution, cation exchange capacity (CEC), specific surface area (SSA), and water and nutrient adsorption, were determined and compared to soil properties in order to determine appropriate biochar application to soil. Pradoo wood biochar showed important adsorption properties with high SSA, CEC and nutrient adsorption. The water retention properties were also improved on the dry end of the water retention curve. Phosphorous and ammonium adsorption–desorption isotherms were established and their respective affinity for the biochar surface was quantified, by the means of a retention index and thermodynamical parameters. We found that despite excellent retention properties, biochar needs to be added in large amounts (between 10 and 70 kg m−2) to soil to be able to modify noticeably the resulting soil properties.  相似文献

Brazil is the world's largest producer of common beans (Phaseolus vulgaris L.). Drought stress harms the morphological and agronomic traits of beans. This study evaluates the reaction to water deficit in five genotypes of black beans. The experiment was conducted in the IDR-IAPAR-EMATER in Londrina-PR, Brazil. A split-plot design was used, with three replications. The genotypes were included in the subplots and the treatments with or without water deficit in the plots. Water deficit was induced on the pre-flowering stage and maintained for 20 days in the plots submitted to drought stress. For the growth analysis, plants were collected at 35, 54 and 70 days after emergence. At the stage of physiological ripeness, several morphological and yield traits were evaluated. The genotypes IPR Uirapuru and BRS Esplendor can be considered tolerant and used as a tolerant source to water deficit in common bean germplasm banks. The line LP 08-90 has morphological and agronomic adaptations efficient to overcome water deficit's effects, presenting a higher grain yield in both crop conditions, which indicates the success of black beans breeding to deal with water deficit.  相似文献
