Yolk coelomitis is a major cause of death in captive sexually mature female Fiji Island banded iguanas (Brachylophus fasciatus) maintained by the Zoological Society of San Diego. The medical records, breeding histories, and pathology archival materials from this group were reviewed to study this health problem. From 1987 through 2004, deaths of nine of 21 adult females were due to yolk coelomitis. Most iguanas had a history of reproduction-related problems, which included reproductive failure, episodes of lethargy associated with ovarian activity, folliculostasis, ovostasis, and behavioral abnormalities. Most affected iguanas either were found dead or presented moribund and subsequently died or were euthanized. Clinical signs were nonspecific and included lethargy, cutaneous discoloration, and coelomic effusion. Yolk leakage in most cases was associated with the presence of large vitellogenic follicles undergoing atresia and resulted in coelomitis characterized by florid mesothelial proliferation. 相似文献

Beneficial effects of aluminum (Al) on plant growth have been reported for plant species adapted to acid soils. However, mechanisms underlying the stimulatory effect of Al have not been fully elucidated. The aim of this study was to determine the possible contribution of photosynthesis, antioxidative defense, and the metabolism of both nitrogen and phenolics to the Al‐induced growth stimulation in tea (Camellia sinensis [L.] Kuntze) plants. In hydroponics, shoot growth achieved its maximum at 50 μM Al suply (24 μM Al3+ activity). A more than threefold increase of root biomass was observed for plants supplied with 300 μM Al (125 μM Al3+ activity). Total root length was positively related to root Al concentrations (r = 0.98). Chlorophyll a and carotenoid concentrations and net assimilation rates were considerably enhanced by Al supply in the young but not in the old leaves. Activity of nitrate reductase was not influenced by Al. Higher concentrations of soluble nitrogen compounds (nitrate, nitrite, amino acids) and reduction of protein concentrations suggest Al‐induced protein degradation. This occurred concomitantly with enhanced net CO2‐assimilation rates and carbohydrate concentrations. Aluminum treatments activated antioxidant defense enzymes and increased free proline content. Lowering of malondialdehyde concentrations by Al supply indicates that membrane integrity was not impaired by Al. Leaves and roots of Al‐treated plants had considerably lower phenolic and lignin concentrations in the cell walls, but a higher proportion of soluble phenolics. In conclusion, Al‐induced growth stimulation in tea plants was mediated by higher photosynthesis rate and increased antioxidant defense. Semi-natural grasslands in Sweden are threatened by land-use change and lack of management with attendant risk to their biodiversity. We present a model to explore the effects of grazing frequency and intensity on plant species persistence, and the relative effects of grassland size and pattern. We used a landscape modelling platform, LAMOS (LAndscape MOdelling Shell), to design a landscape model of vegetation dynamics incorporating the effects of local succession, dispersal and grazing disturbance. Five plant functional groups (PFG), representing various combinations of persistence and dispersal character, light requirements and disturbance responses, were defined to model species dynamics. Based on old cadastral maps three different landscapes were designed representing specific time-layers, i.e., a historical (17th to 18th century), a pre-modern (1940s) and a present-day landscape. Simulations showed that a threshold was crossed when grasslands decreased in area to about 10–30% of the modelled area, and as a consequence the biomass of grassland-specific PFGs was strongly reduced. These competition sensitive groups did not persist in the model even with intense grazing in the present-day landscape, where grasslands occupy 11% of the total area. However, all grassland species would have been able to persist in the historical landscape, where grasslands occupied 59% of the total area, even without grazing. Our results suggest that continuous but low-intensity grazing is more positive for grassland PFGs than discontinuous but highly intensive grazing. This effect was particularly strong when the frequency and/or intensity of grazing dropped below a threshold of 20%. Simulations using three landscape maps designed to explore effects of further fragmentation and habitat loss showed that the spatial pattern of remaining grasslands is important for the persistence of grassland-specific PFG. The model presented here is an advance towards more realistic grazing models to explore the effects of prescribed grazing and landscape fragmentation on the persistence species or plant functional groups.This revised version was published online in May 2005 with corrections to the Cover Date. 相似文献

Genome mining of pigmented Pseudoalteromonas has revealed a large potential for the production of bioactive compounds and hydrolytic enzymes. The purpose of the present study was to explore this bioactivity potential in a potent antibiotic and enzyme producer, Pseudoalteromonas rubra strain S4059. Proteomic analyses (data are available via ProteomeXchange with identifier PXD023249) indicated that a highly efficient chitin degradation machinery was present in the red-pigmented P. rubra S4059 when grown on chitin. Four GH18 chitinases and two GH20 hexosaminidases were significantly upregulated under these conditions. GH19 chitinases, which are not common in bacteria, are consistently found in pigmented Pseudoalteromonas, and in S4059, GH19 was only detected when the bacterium was grown on chitin. To explore the possible role of GH19 in pigmented Pseudoalteromonas, we developed a protocol for genetic manipulation of S4059 and deleted the GH19 chitinase, and compared phenotypes of the mutant and wild type. However, none of the chitin degrading ability, secondary metabolite profile, or biofilm-forming capacity was affected by GH19 deletion. In conclusion, we developed a genetic manipulation protocol that can be used to unravel the bioactive potential of pigmented pseudoalteromonads. An efficient chitinolytic enzyme cocktail was identified in S4059, suggesting that this strain could be a candidate with industrial potential. 相似文献

A reduction in inflammatory cell apoptosis is an important concept in the maintenance of inflammation and a potential target for the resolution of inflammation in many inflammatory diseases. Dysregulation of apoptosis has been implicated in a range of diseases, including tumors, neurodegenerative disorders and autoimmunity, and may also be implicated in allergic asthma. In horses, recurrent airway obstruction (RAO) is an asthma-like condition that is characterized increased survival neutrophil bronchial. Tamoxifen is a synthetic, non-steroidal, anti-estrogen agent that is widely used for treating all stages of breast cancer and has been approved for the prevention of breast cancer in high-risk women. The observed efficacy of tamoxifen has been attributed to both growth arrest and the induction of apoptosis. Therefore, the aim of our study was to evaluate the ability of tamoxifen to induce apoptosis in vitro in granulocytic cells from peripheral blood and in mononuclear cells from bronchoalveolar lavage fluid (BALF) in horses. Flow cytometry using commercial AnnexinV-FITC and propidium iodide was used to quantify early and late apoptotic leukocytes, respectively. The results showed a significant increase in early apoptosis in peripheral blood and bronchial granulocytic cells treated with tamoxifen. The rate of early apoptosis of mononuclear cells from blood and BALF when incubated with tamoxifen was significantly lower compared with granulocytic cells. We did not observe a direct effect of tamoxifen on late apoptosis in any of the in vitro assays in the cell types used here. These results indicate that the apoptotic mechanisms under these experimental conditions would affect only blood and BALF granulocytic cells, particularly in early apoptosis. Finally, further in vitro and in vivo studies are needed to better understand apoptotic mechanisms because tamoxifen could be used to treat chronic, inflammatory pathologies associated with granulocytes and allergic diseases, such as asthma or equine RAO. 相似文献

Background
Dichelobacter nodosus is the causative agent of footrot in sheep. The survival of the bacterium in soil is of importance for the epidemiology of the disease. The investigation evaluates the survival of D. nodosus in soil with and without added hoof powder stored under different temperatures.Results
An experimental setup was used with bacteriological culture and real-time polymerase chain reaction (PCR), and the results indicate that the bacteria can survive in soil for longer time than previously expected. The survival time was found to be dependent on temperature and the addition of hoof powder to the soil, with the longest survival time estimated to be 24 days in soil samples with hoof powder stored at 5°C.Conclusion
Our findings indicate that the survival time of D. nodosus and its ability to infect susceptible sheep on pasture under different climatic conditions should be studied further. 相似文献999.

Background
During epididymal transit, functional and structural modifications leading to full maturation enable male gametes to reach, recognize and fertilize the oocytes. In dogs, little is known on the modifications of spermatozoa during the passage in the epididymis. The aim of this study was to describe the motility, morphology and acrosomal patterns of canine spermatozoa retrieved from the epididymis caput, corpus and cauda.Results
After the dilution required for the collection of epididymal content, sperm motility was significantly higher (P <0.0001) in the cauda compared to corpus and caput.Proportions of spermatozoa with normal morphology were significantly higher in corpus (P =0.02) and cauda (P <0.0001) compared to caput. Overall morphological abnormalities of the head and neck/midpiece were similar in the three different epididymal regions. A significantly increased prevalence of tail defects, mainly represented by single bent tails, was observed in the corpus compared to caput (P <0.0001) and cauda (P =0.006).Numbers of immature sperm with cytoplasmic droplets decreased from the proximal to the distal region of the epididymis. Particularly, proximal cytoplasmic droplets were more frequently found in spermatozoa collected from the caput epididymis than in the corpus (P <0.0001) and in the cauda (P <0.0001), whereas the occurrence of distal cytoplasmic droplets was higher in the corpus than in the caput (P =0.0003) and in the cauda (P <0.05).Significantly higher proportions of spermatozoa with intact acrosomes were retrieved from the cauda epididymis than from the caput (P =0.03) and the corpus (P =0.008). This difference was mainly due to a lower proportion of spermatozoa with abnormal acrosomes (mainly swollen acrosomes) rather than with absent acrosomes.Conclusions
Canine spermatozoa undergo several modifications in the epididymis. The acquisition of progressive motility, migration of the cytoplasmic droplet and acrosomal reshaping lead to mature spermatozoa which are then stored in the cauda epididymis. From this site, spermatozoa can be retrieved and used in assisted reproductive techniques as a valuable tool for propagating genetic traits of high value individuals that dies accidentally or undergoes orchiectomy for medical purposes. Thirty-two anesthetic episodes used a combination of tiletamine-zolezepam (50 mg/ml each), ketamine (80 mg/ml), and xylazine (20 mg/ml) at various dosages for routine diagnostic and minor surgical procedures in 13 captive cheetahs (Acinonyx jubatus). The mean dosage (0.023 +/- 0.003 ml/kg) provided rapid induction with a single i.m. injection along with safe predictable working time, good muscle relaxation, and analgesia.
