ObjectiveTo investigate the effects of methadone on the minimum alveolar concentration of isoflurane (ISOMAC) in dogs.Study designProspective, randomized cross-over experimental study.AnimalsSix adult mongrel dogs, four males and two females, weighing 22.8 ± 6.6 kg.MethodsAnimals were anesthetized with isoflurane and mechanically ventilated on three separate days, at least 1 week apart. Core temperature was maintained between 37.5 and 38.5 °C during ISOMAC determinations. On each study day, ISOMAC was determined using electrical stimulation of the antebrachium (50 V, 50 Hz, 10 mseconds) at 2.5 and 5 hours after intravenous injection of physiological saline (control) or one of two doses of methadone (0.5 or 1.0 mg kg?1).ResultsMean (±SD) ISOMAC in the control treatment was 1.19 ± 0.15% and 1.18 ± 0.15% at 2.5 and 5 hours, respectively. The 1.0 mg kg?1 dose of methadone reduced ISOMAC by 48% (2.5 hours) and by 30% (5 hours), whereas the 0.5 mg kg?1 dose caused smaller reductions in ISOMAC (35% and 15% reductions at 2.5 and 5 hours, respectively). Both doses of methadone decreased heart rate (HR), but the 1.0 mg kg?1 dose was associated with greater negative chronotropic actions (HR 37% lower than control) and mild metabolic acidosis at 2.5 hours. Mean arterial pressure increased in the MET1.0 treatment (13% higher than control) at 2.5 hours.Conclusions and clinical relevanceMethadone reduces ISOMAC in a dose-related fashion and this effect is lessened over time. Although the isoflurane sparing effect of the 0.5 mg kg?1 dose of methadone was smaller in comparison to the 1.0 mg kg?1 dose, the lower dose is recommended for clinical use because it results in less evidence of cardiovascular impairment.  相似文献

With the development of the textile industry, there has been a demand for dye removal from contaminated effluents. In recent years, attention has been directed toward various natural solid materials that are capable of removing pollutants from contaminated water at low cost. One such material is sugarcane bagasse. The aim of the present study was to evaluate adsorption of the dye Acid Violet Alizarin N with different concentrations of sugarcane bagasse and granulometry in agitated systems at different pH. The most promising data (achieved with pH 2.5) was analyzed with both Freundlich and Langmuir isotherms equations. The model that better fits dye adsorption interaction into sugarcane bagasse is Freundlich equation, and thus the multilayer model. Moreover, a smaller bagasse granulometry led to greater dye adsorption. The best treatment was achieved with a granulometry value lower than 0.21 mm at pH 2.50, in which the total removal was estimated at a concentration of 16.25 mg mL−1. Hence, sugarcane bagasse proves to be very attractive for dye removal from textile effluents.  相似文献

Electrochemical processes in industrial effluents have been studied as a means to obtain higher efficiency in wastewater treatment. Heterogeneous photocatalysis appears as a low-cost alternative through the use of lower wattage lamps and thermal TiO2 films. Photocatalysis became a clean process for water treatment due to hydroxyl radicals generated on semiconductor surface. Such radicals are able to degrade several organic compounds. This study used different electrodes and analytical methods for degradation of phenol molecules to reduce treatment costs, improve efficiency, and identify compounds formed during the decomposition of phenolic molecules. Thermal growth of TiO2 film was observed on the titanium electrode in rutile form. Application of an electrical potential on the Ti/TiO2 working electrode increases efficiency in reducing concentration of phenol after photocatalytic treatment. Still, high energy radiation (UVC) showed best degradation rates in photolytic process. Different compounds formed during the degradation of phenol were also identified in the UVC?CPE treatment.  相似文献

Neosporosis is of alarming economic concern in the cattle industry. The effectiveness of diagnostic tests for detecting specific antibodies against Neospora caninum is hampered by potential cross-reaction with other coccidia. Use of a single specific antigen might improve test specificity. An indirect enzyme-linked immunosorbent assay (ELISA) was developed using the truncated protein NcSRS2 expressed in Escherichia coli. The ELISA results were compared with those of the indirect fluorescence antibody test (IFAT). Receiver Operating Characteristic (ROC) and Tests in the Absence of a Gold Standard (TAGS) analysis revealed an assay having 96% specificity and 95% sensitivity when applied to 145 positive and 352 negative sera from two distinct cattle populations. Using OD ≤ 0.095 as the cut-off point, the assay's negative and positive predictive values ranged from 98.8% to 50.8% and from 58.8% to 99.1%, respectively, depending on neosporosis prevalence in a given area. The novel ELISA-NcSRS2 format described in the present report constitutes a specific and sensitive method for detecting N. caninum in cattle.  相似文献

Meteorological scenarios concerning the data required for engineering applications of pollutant transport modeling in the low layers of troposphere are defined through a simple methodology. This involves only data at soil surface and substantially relies on the determination of atmospheric stability carried out through fictitious vertical profiles of air temperature considered as representative of the actual vertical profiles in a study area. This assumption is supported by comparisons of fictitious vertical profiles, obtained by measurements of air temperature at soil surface but at different heights above sea level, and the temperature vertical profiles observed by the radio-sounding station closer to the study area. The fictitious profiles are first used to derive the usual classes of atmospheric stability as unstable atmosphere, neutral atmosphere, stable atmosphere, thermal inversion at the surface and thermal inversion in the upper layers. Then, each scenario is determined through the classes of atmospheric stability observed at 06.00 GMT and 15.00 GMT together with the experimental data of air temperature, relative humidity, wind speed, wind direction, and cloud cover, all averaged in the period of investigation of a few years. An area of Central Italy, where the meteorological measurements for a period of 7 years were available, has been selected for this study.  相似文献

Abstract

The foliar application of silicon (Si) and salicylic acid (SA) can be beneficial to plants. However, there are doubts about the interaction of Si and SA in the physiology and yield of legume crop. The objective of this study was to evaluate the effects of foliar application of Si and SA on the photosynthetic variables and yield of soybean and bean grains. The experiments were composed by four treatments: potassium silicate and stabilized sodium at a concentration of 3.6?g?L?1 Si in the absence and presence of SA (210?mg?L?1), only SA, and without Si or SA. The interaction of Si with foliar SA enhanced photosynthesis only in soybean; it did not affect the physiological variables of bean and did not alter the yield of the two crops. The results indicated that foliar Si application at high concentrations, independently of SA application, increased the physiological variables of the soybean without affecting the yield; however, the combined application of Si and SA had an adverse effect on the physiology and yield of bean.  相似文献

ABSTRACT

Citrus cultures have a fundamental importance to the Brazilian economy; certain aspects such as plant nutrition, yield, and fruit quality are vital for the citrus industry sustainability. The present study evaluated the nutritional status of manganese in adult Pear orange trees using different lime rates topically applied to the soil. The direct evaluation of lime rates effects on leaf manganese (Mn) levels revealed a decrease of the nutrient correlated to its increased, as well as passage of time between application and measurement. Foliar sampling 30 months after surface lime application evidenced a high correlation of foliar manganese levels with soil base saturation of 10–20 cm. Leaf manganese levels which showed a great probability of high productivity were between 33 and 70 mg kg?1.  相似文献

Low-molecular-weight glutenin subunits (LMW-GS) are polymeric protein components of wheat endosperm and like all seed storage proteins, are digested to provide nutrients for the embryo during seed germination and seedling growth. Due to their structural characteristics, they exhibit features important for the technological properties of wheat flour. Their ability to form inter-molecular disulphide bonds with each other and/or with high-molecular-weight glutenin subunits (HMW-GS), is important for the formation of the glutenin polymers, which are among the biggest macromolecules present in nature, and determine the processing properties of wheat dough. Explanation of the structural basis for these correlations continues to intrigue researchers and, while earlier emphasis had been on HMW-GS, considerable attention is now being focused on the LMW-GS.LMW-GS are a highly polymorphic protein complex, including proteins with gliadin-type sequences. Difficulty in separating single components, arising from the complexity of the group, has limited the characterisation of the individual proteins and the establishment of clear-cut relationships with quality parameters.Here we review results concerning different aspects of LMW-GS, including their structural characteristics, genetic control, and relationships with quality parameters. In addition, we emphasise the distinction between the components with sequences unique to the LMW-GS fraction and those behaving like glutenin subunits (incorporated into polymers), but with sequences corresponding to gliadins.  相似文献

In this study we demonstrate that the demosponge Suberites domuncula harbors a Mn(II)-oxidizing bacterium, a Bacillus strain, termed BAC-SubDo-03. Our studies showed that Mn(II) stimulates bacterial growth and induces sporulation. Moreover, we show that these bacteria immobilize manganese on their cell surface. Comparison of the 16S rDNA sequence allowed the grouping of BAC-SubDo-03 to the Mn-precipitating bacteria. Analysis of the spore cell wall revealed that it contains an Mn(II)-oxidizing enzyme. Co-incubation studies of BAC-SubDo-03 with 100 μM MnCl(2) and >1 μM of CuCl(2) showed an increase in their Mn(II)-oxidizing capacity. In order to prove that a multicopper oxidase-like enzyme(s) (MCO) exists in the cell wall of the S. domuncula-associated BAC-SubDo-03 Bacillus strain, the gene encoding this enzyme was cloned (mnxG-SubDo-03). Sequence alignment of the deduced MCO protein (MnxG-SubDo-03) revealed that the sponge bacterium clusters together with known Mn(II)-oxidizing bacteria. The expression of the mnxG-SubDo-03 gene is under strong control of extracellular Mn(II). Based on these findings, we assume that BAC-SubDo-03 might serve as a Mn reserve in the sponge providing the animal with the capacity to detoxify Mn in the environment. Applying the in vitro primmorph cell culture system we could demonstrate that sponge cells, that were co-incubated with BAC-SubDo-03 in the presence of Mn(II), show an increased proliferation potential.  相似文献
