During the 1990s, the Government of Peru began to aggressivelyprivatize agriculture. The government stopped loaning money to farmers' cooperatives and closed the government rice-buying company. The government even rented out most of its researchstations and many senior scientists lost their jobs. As part of this trend, the government eliminated its seed certification agency. Instead, private seed certification committees were set up with USAID funding and technical advise from a US university. The committees were supposed to become self-financing (bycertifying seed grown by small seed producers) and each committee was supposed to encourage the development of a group of small seed-producing firms, clustered around the seedcertification agency. The amazing thing is that many of the seed committees actually accomplished these goals. The agronomists who staffed the committees stood by their jobs,even after US funding ended, even though the committees' income was (at best) modest, and occasionally under the threat of violence from the extreme left. Some seed certificationcommittees failed and others did not. Some of the problems with Peruvian agricultural liberalization can be seen in regard to the seed programs of maize, rice, potatoes, and beans. Cell cycle-dependent histone genes are transcribed at a basal level throughout the cell cycle, with a three- to fivefold increase during early S phase. Protein-DNA interactions in the 5' promoter region of a cell cycle-regulated human H4 histone gene have been analyzed at single-nucleotide resolution in vivo. This region contains two sites, with four potential protein-binding domains, at which the DNA is protected from reaction with dimethyl sulfate in cells and from digestion with deoxyribonuclease I in nuclei. These protein-DNA interactions persist during all phases of the cell cycle and dissociate with 0.16 to 0.2M sodium chloride. 相似文献

Of the four types of biological control, (1) natural, (2) conservation, (3) augmentation, and (4) importation), ethical concerns have been raised almost exclusively about only one type: importation. These concerns rest largely on fears of extinction of animal species. Importation biological control is a cost-effective alternative to chemical control for basic food crops of resource-poor farmers. Regarding the other types of biological control, natural biological control is not consciously manipulated by humans. Augmentation has some technical concerns, but is generally an environmentally-sound, viable alternative to chemicals and offers local employment. Folk experiments in agriculture are often inspired by new ideas blended with old ones, motivated by economic and environmental change. They tend to save labor or capital. These notions are illustrated with nine short case studies from Nicaragua and El Salvador. The new ideas that catalyze folk experiments may be provided by development agencies, but paradoxically, the folk experiments are so common that the agencies that inspire them usually pay little attention to them. Some folk experiments are original, but others simply copy innovations that farmers have seen somewhere else. Unlike formal scientific research, in which results are consistently written, folk experiments are rarely “inscribed,” because the results are for use by individual farmers and need not be shared with an audience. 相似文献

Protein O-mannosylation is an essential and evolutionarily conserved post-translational modification among eukaryotes. This form of protein modification is also described in Mycobacterium tuberculosis; however, the mechanism of mannoprotein assembly remains unclear. Evaluation of differentially translocated chimeric proteins and mass spectrometry to monitor glycosylation demonstrated that specific translocation processes were required for protein O-mannosylation in M. tuberculosis. Additionally, Rv1002c, a M. tuberculosis membrane protein homolog of eukaryotic protein mannosyltransferases, was shown to catalyze the initial step of protein mannosylation. Genetic sequence alignment is the basis of many evolutionary and comparative studies, and errors in alignments lead to errors in the interpretation of evolutionary information in genomes. Traditional multiple sequence alignment methods disregard the phylogenetic implications of gap patterns that they create and infer systematically biased alignments with excess deletions and substitutions, too few insertions, and implausible insertion-deletion-event histories. We present a method that prevents these systematic errors by recognizing insertions and deletions as distinct evolutionary events. We show theoretically and practically that this improves the quality of sequence alignments and downstream analyses over a wide range of realistic alignment problems. These results suggest that insertions and sequence turnover are more common than is currently thought and challenge the conventional picture of sequence evolution and mechanisms of functional and structural changes. 相似文献

The 1918 "Spanish" influenza pandemic represents the largest recorded outbreak of any infectious disease. The crystal structure of the uncleaved precursor of the major surface antigen of the extinct 1918 virus was determined at 3.0 angstrom resolution after reassembly of the hemagglutinin gene from viral RNA fragments preserved in 1918 formalin-fixed lung tissues. A narrow avian-like receptor-binding site, two previously unobserved histidine patches, and a less exposed surface loop at the cleavage site that activates viral membrane fusion reveal structural features primarily found in avian viruses, which may have contributed to the extraordinarily high infectivity and mortality rates observed during 1918. 相似文献

We have observed both superluminal and ultraslow light propagation in an alexandrite crystal at room temperature. Group velocities as slow as 91 meters per second to as fast as -800 meters per second were measured and attributed to the influence of coherent population oscillations involving chromium ions in either mirror or inversion sites within the crystal lattice. Namely, ions in mirror sites are inversely saturable and cause superluminal light propagation, whereas ions in inversion sites experience conventional saturable absorption and produce slow light. This technique for producing large group indices is considerably easier than the existing methods to implement and is therefore suitable for diverse applications. 相似文献

Mounting a protective immune response is critically dependent on the orchestrated movement of cells within lymphoid organs. We report here the visualization, using major histocompatability complex class I tetramers, of the CD8-positive (CD8) T cell response in the spleens of mice to Listeria monocytogenes infection. A multistage pathway was revealed that included initial activation at the borders of the B and T cell zones followed by cluster formation with antigenpresenting cells leading to CD8 T cell exit to the red pulp via bridging channels. Strikingly, many memory CD8 T cells localized to the B cell zones and, when challenged, underwent rapid migration to the T cell zones where proliferation occurred, followed by egress via bridging channels in parallel with the primary response. Thus, the ability to track endogenous immune responses has uncovered both distinct and overlapping mechanisms and anatomical locations driving primary and secondary immune responses. 相似文献
