Seasonal variations in solar elevation, canopy phenology and leaf pigmentation result in a dynamic variation in the radiation regime within a deciduous forest. Measurements of several insolation components taken within an east Tennessee oak-hickory forest during eight combinations of season and phenological conditions of the canopy are presented. Insolation measurements were made with instruments mounted on a moving tram system at seven levels within, and at one level above the canopy.The attenuation of solar radiation is least during the leafless phenoseason since only woody biomass is present to intercept and absorb it. Net radiation is attenuated to a greater extent than shortwave and photosynthetically active radiation, which are attenuated to a similar degree, since the sparse woody biomass does not effectively trap the outgoing reflected and radiated radiation. Solar elevation did not influence the attenuation of insolation within the leafless forest under the conditions studied.The attenuation of insolation within the leafing, spring canopy increases progressively with increasing leaf area. The attenuation among the components, however, varies. Shortwave attenuation progresses at a faster rate with expanding leaf area than photosynthetically active radiation since newly expanded leaves are low in chlorophyll. The penetration of light into the leafing canopy is independent of solar elevations less than 40° and increases linearly with greater solar elevations.The radiation regime within the fully-leafed canopy is rather static. For daily mean values, no effects of leaf age or seasonal variation in solar elevation are evident. Penetration of light into the canopy, however, increases when solar elevation exceeds 65°.The attenuation of insolation components diminishes during the autumnal senescence—abscission phenoseason, in response to changes in leaf pigment and loss of foliage. Attenuation of insolation is greater in the autumn than during spring periods with similar leaf area since the sun is lower in the sky in the autumn.The depletion of photosynthetically active radiation with depth is minimal during the leafless, early leafing and autumnal phenoseasons. Moderate depletion occurs during the late leafing period and maximal depletion occurs during the fully-leafed phenoseason.  相似文献

Kansas State University implemented a Web-based program to assess students' competency to perform technical skills during clinical rotations throughout the fourth year of the veterinary curriculum. The classes of 2009 and 2010 recorded a minimum number of procedures (104 and 103, respectively) from a menu of more than 220 recommended procedures. Procedures were categorized by species (small animal, equine, food animal) and disciplines (imaging, anesthesia, diagnostic medicine/necropsy). Ophthalmology was added as a fourth discipline for the class of 2010. Students recorded procedures into the Web-based system, including information about the patient, procedure performed, supervisor, and a self-assessment of performance. Faculty, staff, and house officers evaluated the procedures electronically by confirming that they witnessed the procedure and providing qualitative and written feedback. The class of 2009 recorded 18,492 procedures (M=171/student) and the class of 2010 recorded 16,935 procedures (M=158/student). Two students from each class (2009 and 2010) did not complete the minimum required skills during clinical rotations and returned to perform procedures immediately before (n=3) or immediately after (n=1) graduation to receive their diploma. The Web-based system captured a large number of assessments of technical competency performed in the clinical setting. The system provided students with formative feedback throughout the clinical year, ensured equitable distribution of procedural opportunities across the student body, and required minimal additional resources.  相似文献

A quantitative polymerase chain reaction (qPCR) assay was developed for the detection and quantification of Edwardsiella ictaluri in channel catfish Ictalurus punctatus pond water using modifications to a published E. ictaluri-specific qPCR assay and previously established protocols for the molecular detection of myxozoan parasites in catfish ponds. Genomic DNA equivalents indicative of the number of bacteria in a sample were determined and standard curves correlating to bacterial numbers were established. The assay was found to be highly repeatable and reproducible, with a linear dynamic range of five orders of magnitude. There was no interference of the assay from the presence of large quantities of nontarget DNA. Known quantities of bacteria were added to sample volumes of 40 or 500 mL of pond water collected from several different ponds. The minimum level of detection was approximately 100 cell equivalents (CE) in 40 (2.5 CE/mL) or 500 mL of pond water (0.2 CE/mL). Sample volumes of 40 mL yielded the most consistent results, which were not significantly different from those obtained from broth culture alone. Cell equivalents determined by qPCR in 40-mL pond water samples spiked with known quantities of bacteria were within one order of magnitude of the actual number of cells added. Repetitive element-based polymerase chain reaction analysis of archived isolates demonstrated the genetic homogeneity of E. ictaluri, and consistent amplification of these isolates by qPCR analysis demonstrated the stability of the PCR target. The assay described here provides a reliable method for the detection and quantification of E. ictaluri in pond water and will be an invaluable tool in epidemiological studies. Additionally, the assay provides a way to evaluate the effects that vaccination, antibiotic treatments, and restricted feeding practices have on E. ictaluri populations during an outbreak. Information obtained with these tools will aid in optimizing disease management practices designed to maximize productivity while minimizing losses.  相似文献

Post‐dispersal predation is a potentially significant modifier of the distribution of recruiting plants and an often unmeasured determinant of the effectiveness of a frugivore's dispersal service. In the wet tropical forests of Australia and New Guinea, the cassowary provides a large volume, long distance dispersal service incorporating beneficial gut processing; however, the resultant clumped deposition might expose seeds to elevated mortality. We examined the contribution of post‐dispersal seed predation to cassowary dispersal effectiveness by monitoring the fate of 11 species in southern cassowary (Casuarius casuarius johnsonii Linnaeus) droppings over a period of 1 year. Across all species, the rate of predation and removal was relatively slow. After 1 month, 70% of seeds remained intact and outwardly viable, while the number fell to 38% after 1 year. The proportion of seeds remaining intact in droppings varied considerably between species: soft‐seeded and large‐seeded species were more likely to escape removal and predation. Importantly, across all species, seeds in droppings were no more likely to be predated than those left undispersed under the parent tree. We speculate that seed predating and scatter‐hoarding rodents are responsible for the vast majority of predation and removal from droppings and that the few seeds which undergo secondary dispersal survive to germination. Our findings reinforce the conclusion that the cassowary is an important seed disperser; however, dispersal effectiveness for particular plant species can be reduced by massive post‐dispersal seed mortality.  相似文献

The transition from anoestrus to oestrus in mares is controlled by photoperiod. The present study examined whether additional daylength would accelerate the mares' response to gonadotrophin-releasing-hormone (GnRH). Nine anoestrous mares were placed under ambient or artificial long lighting on 7th January. The four month experimental period was divided into a three-day sequence which was repeated at 21 day intervals. Ovaries were palpated rectally on Day 1; saline was injected (1 ml intravenously [iv]) on Day 2; GnRH was administered (0.59 microgram/kg bodyweight iv) on Day 3. Blood was taken at -60, 0, 15, 30, 60 and 120 mins relative to saline or GnRH treatment. Serum luteinising hormone (LH) was determined by a homologous equine radioimmunoassay (RIA). Several criteria were employed to define a positive response to GnRH and the results were analysed by Fisher's exact probability test. Treatment with artificial light allowed a response to GnRH within six weeks whereas the mares in ambient lighting took 12 weeks to respond to GnRH. The advancement in the time of response to GnRH under the long photoperiod could be related to changes in pituitary LH content, accelerated follicular activity or alterations in other brain-pituitary hormone levels.  相似文献

Enteric septicemia of catfish (ESC), caused by Edwardsiella ictaluri, is the most problematic bacterial disease affecting catfish aquaculture in the southeastern United States. Efforts to develop an effective ESC vaccine have had limited industrial success. In commercial settings, ESC vaccines are typically administered by immersion when fry are transferred from the hatchery to rearing ponds. While this approach is a practical method of mass delivery, this strategy administers vaccines to very young fish, which lack a fully developed immune system. To circumvent this limitation, an oral vaccination strategy was evaluated as a means of immunizing catfish at the fingerling stage of production, when fish possess a more complete immune arsenal. A virulent E. ictaluri isolate (S97-773) was attenuated by successive passage on media containing increasing concentrations of rifamycin. In laboratory trials, cultured vaccine was diluted and mixed with feed (100 mL diluted vaccine/454 g feed). This mixture was then fed to Channel Catfish Ictalurus punctatus fingerlings. Two separate dilutions of cultured vaccine (1:10 and 1:100) were used to create the vaccine–feed mixture, equating to estimated doses of 5 × 107 and 5 × 106 CFU/g of feed, respectively. After 30 d, catfish were exposed by immersion (1 × 106 CFU/mL) to the virulent parental strain of E. ictaluri. The target dose (1:100 dilution, ~5 × 106 CFU/g of feed) offered exceptional protection (relative percent survival = 82.6–100%). In addition, negligible deaths occurred in fish vaccinated at 10 times the target dose (1:10 dilution, ~5 × 107 CFU/g of feed). In pond trials, antibody production increased 18-fold in orally vaccinated fish. When compared with nonvaccinated controls, vaccination significantly improved survival, feed fed, feed conversion, biomass produced, and total harvest. This research demonstrates Channel Catfish can be successfully immunized in a commercial setting against E. ictaluri with a single dose of an orally delivered, live attenuated, E. ictaluri vaccine.

Diatoms are dominant photosynthetic organisms in the world’s oceans and represent a major food source for zooplankton and benthic filter-feeders. However, their beneficial role in sustaining marine food webs has been challenged after the discovery that they produce secondary metabolites, such as polyunsaturated aldehydes (PUAs), which negatively affect the reproductive success of many invertebrates. Here, we report the effects of two common diatom PUAs, heptadienal and octadienal, which have never been tested before at the molecular level, using the sea urchin, Paracentrotus lividus, as a model organism. We show that both PUAs are able to induce teratogenesis (i.e., malformations), as already reported for decadienal, the better-studied PUA of this group. Moreover, post-recovery experiments show that embryos can recover after treatment with all three PUAs, indicating that negative effects depend both on PUA concentrations and the exposure time of the embryos to these metabolites. We also identify the time range during which PUAs exert the greatest effect on sea urchin embryogenesis. Finally, we report the expression levels of thirty one genes (having a key role in a broad range of functional responses, such as stress, development, differentiation, skeletogenesis and detoxification processes) in order to identify the common targets affected by PUAs and their correlation with morphological abnormalities. This study opens new perspectives for understanding how marine organisms afford protection from environmental toxicants through an integrated network of genes.  相似文献
