Ethiopia has a large and diverse population of small ruminants, which contribute substantially to the livelihood and income of the rural poor and the country at large. However, the sector faces a number of challenges. Productivity per animal and flock offtake are both very low. Reasons attributed for the apparent low productivity are as follows: absence of appropriate breeding programmes, lack of technical capacity, inadequate and poor‐quality feeds, diseases leading to high lamb mortality, and underdeveloped markets in terms of infrastructure and information. Historically, sheep and goats have received little policy or investment attention. Genetic improvement of small ruminants could contribute to bridging the productivity gap. In the past, the government of Ethiopia has placed much emphasis on importing exotic genetics and cross‐breeding with local stock as a strategy for genetic improvement. However, this has not led to a significant productivity improvement and the programmes have generally been unsustainable. Currently, there is a change in approach and a recognition of the need to focus genetic improvement efforts on the local genetic resources that are well adapted to the diverse agro‐ecologies and production environments in the country. Community‐based breeding programmes (CBBPs), which focus on indigenous stock and consider farmers’ needs, views, decisions and active participation, from inception through to implementation, have been identified as programmes of choice. The Ethiopian government and the private sector need to invest in strategic areas around CBBPs to make the programme work for the poor and be sustainable in low‐input systems. 相似文献
This experiment was conducted to investigate effect of dried Sesbania sesban leaves supplementation on milk yield, feed intake and digestibility of Holstein Frisian X Zebu (Arado) crossbred dairy cows. Twelve cows at midlactation (155.83?±?4.49 days of lactation), second parity, and 442.21?±?3.40 kg average live body weight were randomly assigned to one of four dietary treatments according to a randomized complete block design. Cows were blocked according to their daily milk yield into three blocks of four animals each. Cows were fed a basal diet (control) or a basal diet supplemented with 1.25 kg/day dried Sesbania sesban leaves, 2 kg/day dried Sesbania sesban leaves and 2.75 kg/day dried Sesbania sesban leaves on a dry matter basis for 8 weeks. Total dry matter intake, nutrient intake, milk yield, dry matter digestibility, and nutrient digestibility showed significant variation among treatments. Cows supplemented with the highest level of Sesbania sesban (2.75 kg/day) had higher total dry matter and nutrient intake. Similarly, cows supplemented with 2 and 2.75 kg/day had higher milk yield than the nonsupplemented cows (up to 11.3 and 16.2%, respectively). Digestibility was lower for the nonsupplemented cows compared to cows supplemented with 2 and 2.75 kg/day dried Sesbania sesban leaves but statistically similar to the cows supplemented with 1.25 kg/day dried Sesbania sesban leaves. Supplementation with 2.75 kg/day Sesbania sesban resulted in higher organic matter digestibility (OMD) compared to the control. Crude protein digestibility (CPD), neutral detergent fiber digestibility (NDFD), and acid detergent fiber digestibility (ADFD) were significantly affected by Sesbania sesban supplementation. The nonsupplemented cows had lower CPD, NDFD, and ADFD. These results indicate that dried Sesbania sesban leaves supplementation to dairy increases total DM intake, digestibility, and milk yield.
The objective of this study was to measure the effect of feeding two total mixed rations (TMRs), differing in their roughage content and in vitro dry matter digestibility, on the respiratory rate, body temperature, eating behavior and energy balance of lactating cows. The partitioning of metabolizable energy intake (MEI) between heat production (HP) and retained energy (RE) of cows held under heat load conditions was measured. Forty-two lactating cows were divided into two similar sub-groups, each of 21 animals, and were fed either a control (CON) ration containing 18% roughage neutral detergent fiber (NDF) or an experimental (EXP) TMR that contained 12% roughage NDF and used soy hulls as partial wheat silage replacer. The in vitro DM digestibility of the CON and EXP TMRs was 75.3 and 78.6%, respectively (P < 0.05). The EXP diet reduced rectal temperature and respiratory rate of the cows while increasing their number of meals per day by 32.7% as compared with the CON fed cows, and these meals were shorter in duration and were eaten faster. The EXP diet increased total DM intake from 19.6 to 21.5 kg/cow/d, milk yield from 32.3 to 34.6 kg, and yield of energy corrected milk from 30.9 to 32.2 kg, as compared with the CON group. Cows fed the EXP TMR had increased RE in milk and body tissue, as compared with the CON group, but the diets had no effect on the measured HP that was maintained similar (121 vs. 127 MJ/cow/d) in the two groups. 1. The avian eggshell is a biomineralised composite ceramic consisting of calcium carbonate embedded in an organic matrix. Matrix components are supposed to be involved in the control of mineralisation, crystallographic texture and biomechanical properties of eggshell. 2. The structure and eggshell matrix composition of various domesticated bird species were compared to gain insight into the universality of the eggshell mineralisation process. 3. The SDS-PAGE profiles of soluble eggshell matrix were specific within groups of birds (a: laying hen, breeder hen, quail, pheasant and possibly turkey; b: guinea fowl; c: duck and goose) but some of the protein bands were common to all groups. 4. Analogies between species were confirmed by Western blotting using hen protein antibodies. Ovocleidin-17 (OC-17) and ovalbumin were revealed in all species (except quail for OC-17). Lysozyme was present only in hen eggshell. Another egg white protein: ovotransferrin showed a positive signal in hens, turkey and quail. Osteopontin was observed in laying and breeder hens and quail. 5. Different proteoglycans were localised to discrete regions within the eggshell. Dermatan sulphate was observed within the matrix of the calcified shell of all species except quail which contained chondroitin-6-sulfate. Keratan sulphate was observed in mammillary bodies of breeder and laying hen, quail, pheasant and turkey while chondroitin sulphate was also present in guinea fowl and duck. 6. The general structural organisation of the different avian eggshells was similar but specific differences were observed in the ultrastructure of the mammillary layer. Species of the same taxonomic family could be grouped according to their structural analogies: breeder hen, turkey and pheasant resembled that of the domestic fowl. Guinea fowl was unique. Goose and duck were quite similar with large and confluent mammillary bodies. 7. Some matrix components are therefore common to eggshells of various species but more information is needed to relate differences in matrix composition between taxonomic groups with differences in ultrastructure. 相似文献
Fish growth hormones (GHs) play an important role in regulating growth, metabolism, reproduction, osmoregulation, and immunity and have thus garnered attention for their application in aquaculture. Zebrafish GH (zGH) cDNA or rainbow trout GH (rtGH) cDNA was cloned into the pMon3401 vector, expressed in MON105-competent Escherichia coli and purified to homogeneity. Their biological activity was evidenced by their ability to interact with ovine GH receptor extracellular domain and stimulate GH receptor-mediated proliferation in FDC-P1-3B9 cells stably transfected with rabbit GH receptor. The relative affinity of zGH and rtGH, estimated by IC50, was about 38-fold and 512-fold lower, respectively, than ovine GH. This is likely the reason for the low biological activity in cells with rabbit GH receptor, ~?36-fold lower for zGH and ~?107-fold lower for rtGH than for human GH. This was not due to improper refolding, as evidenced by circular dichroism analysis. Predicting the activity of fish GHs is problematic as there is no one single optimal in vitro bioassay; heterologous assays may be ambiguous, and only homologous assays are suitable for measuring activity. 相似文献
Late blight is an important constraint to potato production and genotype resistance is an effective disease control mesure. Ten late blight resistant potato genotypes (R-gene free) were assessed for yield performance and stability at early (90 days) and late harvest (120 days) at two locations in Kenya during two years. Significant differences (P ≤ 0.05) in area under disease progress curves (AUDPC) were detected among potato genotypes. Resistant genotypes free of R-genes had significantly (P ≤ 0.05) higher yield at late than early harvest, perhaps due to increased tuber bulking period. The rank of genotypes for AUDPC, late blight resistance, and tuber yield varied across seasons and locations (environment). Additive main effects and multiplicative interaction (AMMI) analysis of tuber yield and late blight resistance resulted in significant (P ≤ 0.05) effects of genotypes (G) and environments (E). The proportion of genotypic variance was larger than the environmental variance and the G × E interactions. For tuber yield, the G, E, and G × E interactions accounted for 42.9, 39.6 and 17.5%; and 53.4, 29.7, and 16.9% at early and late harvests, respectively. For AUDPC, G, E, and G × E accounted for 80.2, 5.0, and 14.8%; as well as 82.3, 4.6, and 13% for early and late harvests, respectively. The resistance of potato genotypes without R-genes varied. The study was conducted at Sinana Agricultural Research Center, Ethiopia to assess the supplementation of graded levels of
vetch (Vicia dasycarpa `lana’) and lucerne (Medicago sativa,’ Hunter river’) hay on feed intake, digestibility and body weight (BW) change of Arsi-Bale sheep fed urea treated barley
straw (UTBS). A 7 day- digestibility and a 90 day- feed intake trials were conducted using 28 and 35 sheep, respectively.
The experimental design was a randomized complete block design with seven dietary treatments that consisted of feeding UTBS
(T1) as the control treatment, UTBS plus 150, 250 and 350 g dry matter (DM) per day of vetch for T2, T3, T4, respectively
and UTBS plus 150, 250 and 350 g DM per day of lucerne for T5, T6 and T7, respectively. Intake of UTBS was not affected (P > 0.05)
by inclusion of lucerne hay at 25–35% of daily DM intake. The supplements increased daily intake of total DM, organic matter
(OM), neutral detergent fiber (NDF), acid detergent fiber (ADF) and metabolizable energy (ME) (P < 0.001) as well as apparent
digestibility of DM, OM (P < 0.001), NDF (P < 0.01), ADF, crude protein (CP) (P < 0.05) and daily BW gain (P < 0.001). Supplementation
with lucerne than vetch hay promoted higher (P < 0.001) CP and ME intakes and daily BW gain. Feeding with the UTBS without
supplementation was enough to meet the maintenance requirements of the sheep and allow small BW gain. The results of the study
showed that urea treatment of barley straw in conjunction with supplementation of lucerne or vetch hay could serve as a useful

Measuring disease resistance accurately and reproducibly is a key requirement for the introgression of partial resistance genes into breeding lines. Here, a qPCR protocol is used to measure fungal biomass, using the wheat- Stagonospora nodorum pathosystem as a model. Seven cultivars of differing reported resistance levels were used. Fungal biomass taken at 220°C thermal days after inoculation accurately predicts the final grain weight loss. It is concluded that a test based on qPCR methods is specific, quantitative, rapid and objective. Such tests could provide useful and economic tools in the development of robustly resistant crop cultivars. 相似文献
