When the rate of ruminal epithelial cell proliferation was measured on the basis of 3H-thymidine incorporation into the cellular DNA, butyrate dose-dependently reduced 3H-thymidine incorporation. In contrast, glucagon at 10 and 100 pg/ml had a slight stimulatory effect on the incorporation, but only in the absence of butyrate.  相似文献

Mitotic indices of the ruminal epithelium of two castrated male sheep fed at 11:00 h were determined at 00:00, 04:00, 08:00, 12:00, 16:00 and 20:00 h. The schedules were repeated in four trials done in December 1975, February, March and June 1976 respectively. The indices ranged from 0.13 to 0.92 per cent, both animals showing a similar tendency in these fluctuations. The mean rate of cell division was low in the morning, high in the afternoon and in the evening, and declined at about midnight. Another pair of male castrated sheep were fed at 07:00, and rumen samples were biopsied six times in three days. The mean rate of epithelial cell division in the rumen was low at 08:00, but high at noon, suggesting that the rate possibly was affected by the food intake of the experimental animals.  相似文献

Differentiation of the histochemical characteristics of the olfactory receptor cells (ORC) was examined by immunohistochemistry for protein gene product 9.5 (PGP 9.5) and calretinin (CR) and lectin histochemistry for Phaseolus vulgaris agglutinin-L (PHA-L) in the developing olfactory epithelium (OE) of the barfin flounder. PGP 9.5 immunoreactivity was diffuse and CR immunoreactivity was restricted at day 7, but these immunoreactivities became intense in the OE toward day 91. Crypt cells were first identified at day 56. PHA-L staining was faint at day 28, but became intense toward day 91. These findings suggest that PGP 9.5-immunopositive cells, CR-immunopositive cells, crypt cells and PHA-L-reactive cells differentiate independently in the developing OE and constitute subsets of the ORC in the OE.  相似文献

The present study was undertaken to investigate whether withdrawal of estrogen and progesterone (EP-withdrawal) stimulates prostaglandin F2alpha (PGF2alpha) production through oxygen radical (ROS)-induced NF-kappaB activation in human endometrial stromal cells (ESC). To study the EP-withdrawal, ESC that had been treated with estradiol (E, 10(-8) M) and medroxyprogesterone acetate (MPA, 10(-6) M) for 12 days were then incubated with or without E+MPA for a further 11 days. PGF2alpha concentrations in the medium and cyclooxygenase-2 (COX-2) mRNA levels were significantly increased after EP-withdrawal, while they were unchanged by the continuous treatment with E+MPA. When ESC were incubated with N-acetyl-L-cysteine (Nac, 50 mM), an antioxidant, during EP-withdrawal, Nac blocked the increases in PGF2alpha production and COX-2 mRNA expression caused by EP-withdrawal. Next, we examined whether ROS generated in response to EP-withdrawal acted through NF-kappaB activation. Electrophoretic mobility shift assay revealed that EP-withdrawal caused marked increases in NF-kappaB DNA binding activity, which was completely suppressed by Nac. Furthermore, when ESC were incubated with MG132 (3 microM), which inhibits NF-kappaB activation, during EP-withdrawal, MG132 blocked the increases in PGF2alpha production and COX-2 mRNA expression caused by EP-withdrawal. In conclusion, EP-withdrawal stimulates COX-2 expression and PGF2alpha production through ROS-induced NF-kappaB activation, suggesting a possible mechanism for menstruation.  相似文献

We describe here the detection by fluorescence of a new photosensitizer, PAD-S31, in tumours in dogs and cats and the effect of photodynamic therapy (PDT) by using PAD-S31 for skin tumours in two dogs and one cat. PAD-S31 is a hydrophilic photosensitizer that has two peaks at absorption wavelengths 406 and 665 nm in distilled water. In a preliminary experiment in mice transplanted with SCCVII and colon 26, PAD-S31 was retained in tumour tissues rather than in other organs. The tumours resected from dogs and cats after intravenous administration of PAD-S31 at a dose of 15 mg/kg emitted strong red fluorescence under light illumination of 402 nm wavelength. Animals given PAD-S31 showed no cutaneous photosensitivity under room light illumination. Irradiation at laser light 670 nm wavelength (fluence rate 150 mW/cm2 and total light dosage 150 J/cm2) on cutaneous mast cell tumours in dogs ( n=2 ) and a cutaneous basal cell tumour in a cat induced complete remission. These results suggest PAD-S31 could be a promising photosensitizer for use in a small animal veterinary practice.  相似文献

In this study, we examined the existence and structure of areolae and the steroidogenesis of areolar trophoblast cells in the Antarctic minke whale placenta morphologically and immunohistochemically. Placentas were collected from the 15th, 16th and 18th Japanese Whale Research Program under Special Permit in the Antarctic (JARPA) and 1st JARPA II organized by the Institute of Cetacean Research in Tokyo, Japan. The opening and cavity of fetal areolae formed by taller columnar trophoblast cells (areolar trophoblast cells) with long microvilli and a bright cytoplasm, as compared with the trophoblast cells of the chorionic villi interdigitating with the endometrial crypts, were recognized in observations of serial sections. The opening of the areolar cavity was hidden by chorionic villi with areolar trophoblast cells. Furthermore, a closed pouch-like structure lined by tall columnar cells similar to areolar trophoblast cells within the stroma of chorionic villi was noticed and continued to the areolar cavity, with the opening seen on serial sections. In a surface investigation of the chorion and endometrium by SEM, maternal (endometrial) areolae irregularly surrounded by endometrial folds were obvious. Moreover, we distinguished areolar trophoblast cells with long microvilli attached with many blebs from trophoblast cells. In our immunohistochemical observations, a steroidogenic enzyme, cytochrome P450 side chain cleavage enzyme (P450scc), was detected with strong immunoreactivity in trophoblast cells. However, areolar trophoblast cells showed weak or no immunoreactivity for P450scc.  相似文献

The immune response during the onset of coliform mastitis in vaccinated cows was investigated by measuring lactoferrin (LF), interleukin-8 ( IL-8), and interleukin-1β (IL-1β) concentrations and somatic cell counts in 28 milk samples at the onset of acute coliform mastitis (ACM) and 73 milk samples at the onset of peracute coliform mastitis (PCM). Vaccinated ACM, unvaccinated ACM, and vaccinated PCM showed significantly higher values for LF and IL-1β levels than unvaccinated PCM (p < .01). The IL-8 concentration was lower in vaccinated PCM than in unvaccinated PCM (p < .05). There was no significant difference in somatic cell counts for each parameter. There were no significant differences in the parameters between vaccinated and unvaccinated ACM cows, or vaccinated ACM and PCM cows. From the above results, it is suggested that mastitis vaccination improved the early immune response, particularly at the onset of PCM, and played a large role in host defense against the initial infection.  相似文献

The effects of low pH on upstream migratory behavior in land-locked sockeye salmon Oncorhynchus nerka were examined using 2-way flow-through channels. Homing mature salmon captured at the mouth of their native stream were used. When neutral (pH 6.7) hatchery water of the same origin as native stream water was allowed to flow through both channels, fish showed active swimming into both channels during a 48 hrs period. When water pH in one channel was lowered by the addition of sulfuric acid, salmon completely ceased upstream swimming behavior into areas of pH lower than 6.0, and swam up into only neutral areas. In the lower pond of the 2-way channels, fish also avoided acidic areas even when average pH was 6.2. Since it has been reported that extremely slight changes in pH such as in the pH 6 range inhibits spawning behavior in this species, it is suggested that land-locked sockeye salmon have a sensitive ability to perceive even slight acidity, and avoid environments which may be deleterious to their offspring.  相似文献

Summary Total chloroplast DNA (cpDNA) from Solanum incanum, a wild relative of eggplant, was used to probe total DNA of Solanum melongena (eggplant). The DNA fragments detected were the same as observed using purified chloroplast DNA. Chloroplast DNAs were also analysed for nine species of Solanum that are cross-compatible with eggplant: S. aethiopicum, S. anguivi, S. gilo, S. incanum, S. indicum, S. integrifolium, S. macrocarpon, S. olivare and S. panduriforme.Restriction fragments generated by eight enzymes were recorded as present or absent, and a matrix for all fragment positions, species and enzymes was used for cluster analysis. In the resulting dendrogram, the species tested formed three distinct groups: (1) S. aethiopicum, S. anguivi, S. gilo, S. indicum, S. integrifolium and S. olivare, (2) S. incanum, S. melongena and S. panduriforme, (3) S. macrocarpon. Six species of the first group belonging to section Oliganthes appears more closely related to the second group members belonging to section Melongena than does S. macrocarpon, which also belongs to section Melongena. Within the second group, S. panduriforme is slightly more like eggplant than is S. incanum.  相似文献
