Factors limiting the maintenance of recombinant ruminal bacterium in the rumen were evaluated in vitro , using batch culture prepared from rumen fluid of sheep. Butyrivibrio fibrisolvens expressing a foreign xylanase gene ( B. fibrisolvens NO4) was used as a tested recombinant that was selectable on an erythromycin-containing agar medium. The recombinant tended to reduce its level slowly in the rumen fluid of sheep on a high hay diet, while its initial decrease was more apparent in the rumen fluid of sheep on a high concentrate diet. Incubation with cell-free ruminal fluid revealed a significant decrease of inoculated recombinant, suggesting the presence of antibacterial factors limiting maintenance of the recombinant. In particular, during the first 12 h of incubation this inhibition was more notable in culture prepared from rumen fluid of sheep given the high concentrate diet. Autoclaving the cell-free rumen fluid inactivated the inhibition. Numbers of the recombinant for inoculation did not influence the final level of survived recombinant, that is, the initial depression was larger as more recombinant was inoculated. Subculturing with xylan before inoculation and/or direct addition of xylan to the batch culture did not improve survival of the recombinant. From these results it is suggested that the level of survived recombinant is limited to 102–4 /mL of in vitro batch culture with restricted energy supply and that initial depression of the recombinant is mainly caused by the heat-sensitive antibacterial factors not associating with microbial cells in the rumen. 相似文献

In the wheat (Triticum aestivum L.) cultivar ‘Zenkoujikomugi’, a single nucleotide polymorphism (SNP) in the promoter of MOTHER OF FT AND TFL1 on chromosome 3A (MFT-3A) causes an increase in the level of gene expression, resulting in strong grain dormancy. We used a DNA marker to detect the ‘Zenkoujikomugi’-type (Zen-type) SNP and examined the genotype of MFT-3A in Japanese wheat varieties, and we found that 169 of 324 varieties carry the Zen-type SNP. In Japanese commercial varieties, the frequency of the Zen-type SNP was remarkably high in the southern part of Japan, but low in the northern part. To examine the relationship between MFT-3A genotype and grain dormancy, we performed a germination assay in three wheat-growing seasons. On average, the varieties carrying the Zen-type SNP showed stronger grain dormancy than the varieties carrying the non-Zen-type SNP. Among commercial cultivars, ‘Iwainodaichi’ (Kyushu), ‘Junreikomugi’ (Kinki-Chugoku-Shikoku), ‘Kinuhime’ (Kanto-Tokai), ‘Nebarigoshi’ (Tohoku-Hokuriku), and ‘Kitamoe’ (Hokkaido) showed the strongest grain dormancy in each geographical group, and all these varieties, except for ‘Kitamoe’, were found to carry the Zen-type SNP. Greenhouse-grown tree ripe (TR) and mature green (MG) mangoes (cv. Irwin) were exposed to high electric field treatment before 20 and 30 days of storage at 5 degrees C. MG fruits were allowed to ripen at room temperature after low-temperature storage. Fruit physical quality attributes, ascorbic acid, carotene, quercetin, total phenols, and antioxidant capacity were estimated before and after the storage period. Antioxidant capacity of fruit juice was estimated using the ferric reducing antioxidant power (FRAP) assay. Fruit firmness decreased significantly during storage. Titratable acidity decreased 20 days after storage. Total soluble solids did not change during storage. Antioxidant capacity of fruits remained unchanged up to 20 days of storage period and decreased thereafter. Total phenol and carotenes increased during storage. Antioxidant capacity of fruits was significantly correlated only to ascorbic acids. Peel color and carotenes were higher in TR fruits, whereas titratable acidity and firmness were higher in MG fruits. There was no significant difference in other parameters between the stages of picking. Electric field pretreatment affected the respiration and antioxidant capacity of TR fruits and did not have any significant affect on other parameters. TR mangoes of cv. Irwin are more suitable for low-temperature storage and can be successfully stored for up to 20 days at 5 degrees C without any significant losses in functional properties and quality attributes. 相似文献

The major taste active component, glutamate (Glu), improves the taste of meat. In this study, we investigated the effect of a short‐term high‐protein (HCP) diet on the intramuscular free Glu content to improve the taste of meat. Furthermore, we elucidated how the muscle free Glu content was controlled by the HCP diet. Chicks (14 days old) were fed the control diet or HCP diet for 10 days. Plasma and muscle free amino acid concentrations, and activity and messenger RNA (mRNA) expression of muscle enzymes related to Glu metabolism were determined. Muscle free Glu content was increased (P < 0.01) by 51%. Activity and mRNA expression of glutaminase (GA), which is one of the major Glu‐related enzymes, were significantly decreased (P < 0.05) in the HCP group because of feedback inhibition. The mRNA expression of lysine α‐ketoglutarate reductase (LKR), which is the enzyme involved in lysine (Lys) degradation and Glu production, was significantly increased (P < 0.001) in the HCP group. These results suggest that short‐term dietary HCP feeding is an effective treatment for improving the taste of meat. Lulu is a dwarf cattle population bred in the Mustang district of western and central Nepal. This area is located around the habitat boundary between Bos taurus and Bos indicus. The peculiarities of Lulu are their small size (weight range in the adult female: 68–153 kg) and rearing in high mountain areas at 2800 m to 4000 m in altitude. There were 5770 head of Lulu cattle in the Mustang district in 1998, 4333 females and 1437 males. The morphological appearance of Lulu is Bos taurus. However, one of the five Lulu studied in Kagbeni, Mustang had a Bos indicus mitochondrial DNA type based on the D‐loop sequence, while the other four were Bos taurus. It is suggested that there are hybrids of Lulu with Bos indicus maternal lineage in a mostly taurine‐breed genetic background. Steps must be taken to preserve the unique Lulu. 相似文献
