A Brucella abortus soluble antigen (BASA) preparation and fractions obtained thereof, by column chromatography, were compared in terms of their ability to induce specific lymphocyte stimulation responses (LSR) in lymphocytes from cattle infected with B. abortus. Endotoxin and protein contents and the fractions were determined. The LSR induced by BASA and the fractions were compared in terms of correctly identifying samples from infected and non-infected cattle. The sensitivity and specificity for each preparation were determined and these two attributes were then correlated with endotoxin and protein content. The results suggest that lymphocyte stimulation had greater association with relative protein content than endotoxin content of the antigen preparations.  相似文献

The interactions of 2 capsular serotype A and 4 serotype D strains of Pasteurella multocida with rabbit polymorphonuclear neutrophils (PMN) were compared in vitro, using a PMN phagocytic and bactericidal assay. Bacteria and rabbit PMN were incubated for 15 minutes. The suspensions were subjected to differential centrifugation and the percentage of phagocytosis (cell association) was determined from the number of viable noncell-associated bacteria. The cell pellets and the associated bacteria were resuspended and PMN bactericidal activity was calculated from the number of remaining viable cell-associated bacteria at 45 and 75 minutes after the start of the assay. Test bacteria were not opsonized or were opsonized with immune serum containing active complement. One type A strain was ingested and killed by PMN in the presence and absence of opsonins. The 5 remaining strains were resistant to PMN killing, but only the type A strain resisted phagocytosis. Resistance of the type A strain was attributed to the hyaluronic acid capsule, since pretreatment of the bacteria with hyaluronidase rendered opsonized bacteria susceptible to ingestion and killing. The pattern of resistance of the 4 type D strains was different from that of the resistant type A strain. Both opsonized and nonopsonized type D bacteria became cell associated, but none were killed by PMN. The mechanism of resistance of these 4 strains to PMN bactericidal activity is currently unknown.  相似文献

Antidotal effects of the 2 antioxidants butylated hydroxyanisole (BHA) and ethoxyquin (EQ) were evaluated in bitterweed (Hymenoxys odorata DC) toxicosis in sheep. Bitteerweed contains a toxic sesquiterpene lactone, hymenoxon, the toxicity of which is reduced by cysteine. Both BHA and EQ are known to induce hepatic glutathione production in rodents. Treatment of sheep with EQ (2.5 g/sheep/day for 9 days before poisoning) gave significant protection from toxic doses of bitterweed, but the protective effect of BHA was insignificant. Of 6 sheep given EQ in the feed, 5 survived 7 doses of bitterweed (4 g/kg/day or higher for 7 days), whereas 5 of 7 controls and 4 of 7 sheep given feed with added BHA died. The added EQ in the feed decreased the serum alkaline phosphatase activity and total protein, albumin, and calcium concentrations. Seemingly, EQ is the first protective agent with field application potential for bitterweed toxicity.  相似文献

An autogenous, multivalent, adjuvanted bacterin for epididymitis was tested in flocks of 700 to 800 Suffolk X white-faced lambs. Long-term feeding of low dosages of antibiotics also was tested in the Suffolk flock. Both methods appeared to reduce the incidence of the disease. The incidence of clinical epididymitis and the number of positive culture results from clinically affected rams were significantly reduced.  相似文献

Immunoaffinity-purified bovine respiratory syncytial virus (BRSV) fusion (F) protein elicited anti-BRSV-specific antibody responses in BRSV-seronegative calves. After primary vaccination, all calves seroconverted to BRSV as determined by the virus neutralization (VN) test and developed anti-F protein antibodies detectable by protein immunoblot analyses. Subsequent vaccinations induced greater than twofold increase in VN titer in 3 of 9 (33%) calves, and 1 calf became VN-negative, but still had nonneutralizing antibody detectable by protein immunoblot analysis. This calf remained seronegative after challenge exposure. Two groups of calves were vaccinated IM with immunoaffinity-purified BRSV F protein. Each dose was 2 ml containing 20 micrograms of purified F protein. Freund's adjuvants were used for all vaccinations, with Freund's complete adjuvant used for the primary vaccination and Freund's incomplete adjuvant for subsequent vaccinations. The vaccine was administered to both groups at weeks 0 and 3; the first group received a third vaccination at weeks 21. Group-1 and -2 vaccinated calves and non-vaccinated contact controls were intranasally aerosol challenge-exposed with low cell culture-passage BRSV on weeks 22 and 9, respectively. Eight of 9 vaccinated calves did not develop a humoral anamnestic response following challenge exposure, as demonstrated by VN test and protein immunoblot analyses. 1. The energy expenditure (H) and physical activity of laying hens were measured under lighting regimes of 14L:10D (standard), 2L:10D:2L:10D (interrupted) and (15 X (13 min L:47 min D]: 9D (fragmented). 2. Neither of the intermittent regimes produced a significant change in total daily energy expenditure, although large alterations occurred in the distribution of H between the lighting phases. The absence of change in total H resulted from the combined effects of greater H in the light in the intermittent regimes, greater H in darkness during the interrupted day than at night and slightly greater H at night in the intermittent regimes. 3. Physical activity count, like H, was redistributed over time but, unlike H, was also significantly reduced in total; changes in the energy cost of unit activity, however, were such that no reduction occurred in the total amount of H attributable to activity. 4. In the standard groups, 90% of total activity costs were incurred in the light; in the fragmented-day and interrupted-day groups, respectively, 7% and 55% of total activity costs were incurred in the light periods and 85% and 30% in daytime darkness. 5. In terms of both H and activity, there was clear differentiation between night darkness and subjective-day darkness. 6. Even when the energy cost of activity was excluded, significant differences remained between H at night, H in the light and H in daytime darkness.  相似文献

Two experiments were conducted to evaluate the effects of dietary CP level on rate, efficiency and composition of gain of growing beef bulls. In Exp. 1, 59 bulls (333 +/- 15.8 kg) were used. Eleven bulls were slaughtered on d 0 to provide an estimate of initial carcass composition (9-10-11 rib section chemical analyses), and remaining bulls were assigned to treatment diets containing 10, 12 or 14% dietary CP. Bulls fed the 10% CP diet grew slower (P less than .05) than bulls fed the 12 or 14% CP diets, although dry matter intake and feed-to-gain ratio did not differ. Bulls fed the 12% CP diet had fatter carcasses (P less than .05) than bulls fed the 10 or 14% CP diets and had greater daily fat accretion than bulls fed the 10% CP diet. In Exp. 2, 60 bulls (318 +/- 9.0 kg) were used. Bulls were assigned to initial slaughter (n = 6) or to one of three dietary treatments, 10, 12 or 14% CP, and were slaughtered after feeding for 66, 136 or 202 d (n = 6 . treatment -1 . slaughter time -1). Bulls fed 10% CP diets had lower (P less than .05) rates of carcass protein accretion during d 0 to 136 and d 0 to 202. Carcass fat gain was similar among treatments over the entire experiment, although bulls fed the 14% CP diet gained more fat during d 0 to 136 than bulls fed the other treatments.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Concentrations of prostaglandin F2 alpha (PGF2 alpha) and thromboxane B2 (TXB2) were evaluated in the milk of cows with naturally occurring (n = 3) and experimentally induced (n = 5) acute coliform mastitis. These arachidonic acid metabolites were measured by radioimmunoassay in unextracted milk. Experimental infections were induced by inoculating 600 to 1,200 colony-forming units of Escherichia coli into 1 mammary quarter per experimental cow. In the experimental cows, milk was collected before inoculation and at 12, 24, 36, 48, 60, and 72 hours after inoculation. Somatic cell concentrations, bovine serum albumin, and concentrations of PGF2 alpha and TXB2 were determined in milk collected at each sampling. Mild-to-moderate increases in milk PGF2 alpha and TXB2 concentrations were observed in cows with naturally occurring mastitis. the increases corresponded to the clinical severity and course of mastitis. In the experimental cows, increases in milk PGF2 alpha and TXB2 concentrations were observed, but the increases were not significant, using a statistical model that included factors of treatment, cows, hours after inoculation, cows-by-treatment and hours-by-treatment interactions, and random error (residual). Results of the present study indicated a large biological variability in milk arachidonic acid metabolite concentrations in cows with acute coliform mastitis, and that arachidonic acid metabolites may be important in the pathophysiologic process of acute coliform mastitis.  相似文献
