The Atacama Desert, located in northern Chile, is one of the driest deserts on the Earth. However, in some years, short and sporadic rainfall in the southern end of this desert has increased the soil moisture that produces ephemeral “desert bloom”. Our goal was to assess the composition of the bacterial community and determine variations in the ammonia-oxidizing bacteria guild diversity from soils collected during the course of the “desert bloom” event. The bacterial composition from this arid soil was determined by cloning and sequencing the 16S rRNA gene. A relatively high diversity of clones belonging to 14 bacterial groups was detected. The ammonia-oxidizers showed a significantly higher diversity of amoA gene clones after the “desert bloom” than during or at the beginning of this event. All sequences obtained were related to Nitrosospira genera and environmental clones. These results suggest that the diversity of ammonia-oxidizing bacteria in this arid soil can be affected by the occurrence of “desert bloom”.  相似文献

Application of crop residues to soil and reduced or no tillage are current management practices in order to achieve better water management, increase soil fertility, crop production and soil erosion control. This study was carried out to quantify the effect of wheat straw mulching in a no tilled Fluvisol under semi-arid conditions in SW Spain and to determine the optimum rate in terms of cost and soil protection. After a 3-years experiment, mulching application significantly improved physical and chemical properties of the studied soil with respect to control, and the intensity of changes was related to mulching rate. The organic matter content was generally increased, although no benefit was found beyond 10 Mg ha1 year1. Bulk density, porosity and aggregate stability were also improved with increasing mulching rates, which confirmed the interactions of these properties. Low mulching rates did not have a significant effect on water properties with respect to control, although the available water capacity increased greatly under high mulching rates. After simulated rainfall experiments (65 mm h1 intensity), it was found that the mulch layer contributed to increase the roughness and the interception of raindrops, delaying runoff generation and enhancing the infiltration of rain water during storms. Mulching contributed to a reduction in runoff generation and soil losses compared to bare soil, and negligible runoff flow or sediment yield were determined under just 5 Mg ha1 year1 mulching rate. It was observed that during simulations, the erosive response quickly decreases with time after prolonged storms (30 min) due to the exhaustion of available erodible particles. These results suggest that the erosive consequences of intermediate intensity 5-years-recurrent storms in the studied area could be strongly diminished by using just 5 Mg ha1 year1 mulching rates.  相似文献

The distribution and variation with soil depth of water repellency has been studied in fire-affected sand dunes under three different vegetation types (pine forest, shrubland and sparse herbaceous vegetation) in SW Spain. The persistence and intensity of water repellency at the exposed surface of soil was measured using the water drop penetration time test and the contact angle method, respectively, in surface samples (0–3 cm) collected at burned and unburned areas. The variation of water repellency with depth in burned areas was studied in soil profiles every 5 cm between 0 and 40 cm depth. None or slight soil water repellency was observed at unburned soil sites, whereas burned soil sites showed a high degree of repellency, especially under pines and shrubland. The spatial pattern of fire-induced soil water repellency was found to be associated to vegetation types, although it was modulated by soil acidity and the soil organic carbon content. Soil water repellency was generally higher at the soil surface, and decreased with depth. Dense pine forests and shrublands showed strong and/or severe water repellency in depth, but it was rare and limited to the first five centimeters under sparse herbaceous vegetation. The heterogeneity of moisture patterns under dense pine forests or shrublands showed the existence of wetting and water repellent three-dimensional soil patches.  相似文献

Herpes simplex virus type-1 (HSV-1) amplicon vectors are versatile and useful tools for transferring genes into cells that are capable of stimulating a specific immune response to their expressed antigens. In this work, two HSV-1-derived amplicon vectors were generated. One of these expressed the full-length glycoprotein D (gD) of bovine herpesvirus 1 while the second expressed the truncated form of gD (gDtr) which lacked the trans-membrane region. After evaluating gD expression in the infected cells, the ability of both vectors to induce a specific gD immune response was tested in BALB/c mice that were intramuscularly immunized. Specific serum antibody responses were detected in mice inoculated with both vectors, and the response against truncated gD was higher than the response against full-length gD. These results reinforce previous findings that HSV-1 amplicon vectors can potentially deliver antigens to animals and highlight the prospective use of these vectors for treating infectious bovine rhinotracheitis disease.  相似文献

The objective of this study was to develop with thermoplastic extrusion amaranth starch derivatives and to characterize and evaluate their functionality as encapsulating agents of Bifidobacterium breve ATCC 15700 and Lactobacillus casei ATCC 334 during spray drying. The survival of both probiotics during storage at different water activities and at two storage temperatures, their viability in a food model system, and their tolerance to a simulated gastrointestinal tract were determined. Native amaranth starch was chemically modified to obtain phosphorylated, acetylated, and succinylated starch. Starch derivatives were reduced in viscosity, and the solubility in water was increased. In general, the modified amaranth starches and control corn starch did not provide good protection to both probiotics during storage at 25°C. However, there was excellent viability during storage at 4°C for both probiotics. Microcapsules showed a uniform coverage of the cells. Storage for 35 days at 25°C of blends of oat with succinylated amaranth microcapsules with probiotics had a lower reduction. Also, this succinylated amaranth starch containing probiotics showed a higher resistance to simulated gastrointestinal conditions. The results with food model systems supported the applicability of the modified starches.  相似文献

A field study was conducted in order to study the effects of different wildfire severities on [1] soil organic matter content, [2] soil water repellency, and [3] aggregate stability; [4] the distribution of soil water repellency in aggregate sieve fractions (1–2, 0.5–1, 0.25–0.5 and < 0.25 mm) was also studied. Five similar burned sites and two long-unburned control sites were selected under mixed fir and pine forests in volcanic highlands from Michoacán, Mexico. Soil water repellency was observed in soil samples from all sites, although changes were influenced by fire severity. Sites affected by low severity fires did not show important changes in burned soils in comparison with controls, while high severity fires caused different responses: water repellency was increased or destroyed probably due to temperatures below or above 200–250 °C during burning. The degree of wettability/repellency from the fine earth fraction of burned soils seems to be conditioned by < 0.5 mm aggregates, more than coarser aggregates which always showed a higher degree of wettability. It is suggested that destruction of organic matter during burning occurs principally in coarse aggregates, where combustion can be more intense. Aggregate stability (measured using pre-wetted aggregates between 4 and 4.8 mm) did not change under low severity burning but it was considerably reduced in the case of a high fire severity. Losses of organic matter and destruction of water repellency seem to be the reasons for that reduction in this type of soil in contrast to previous studies, where aggregate stability increased after burning. Changes in both properties (water repellency and aggregate stability) are expected to induce modifications in runoff and soil loss rates at the hillslope scale.  相似文献

Pollution of water bodies with heavy metal ions is a major worldwide environmental problem. The objective of this study was to elucidate the mechanism in which metallic ions are adsorbed and reduced to metallic nanoparticles onto plant materials using microwave radiation. In this research, we have fabricated metallic silver and lead nanoparticles from their corresponding ions using the aquatic plants Azolla filiculoides and Pistia stratiotes (since identical results are obtained for both plants, the emphasis will be on the Azolla) under microwave radiation. Our data show that metallic silver and metallic lead nanoparticles were completely removed from the polluted solution and were embedded in the A. filiculoides surface after 5 min of microwave reaction. It was also found that, for both metals, reduction of the metallic ions was accomplished by the plant matrix without the need of an external reducing agent. Most of the particles had a spherical shape within the 10?C50 nm size range. Mass balance data clearly indicate that most of the silver particles were found on the surface of the plant and not in the clean water. Pectin and ??-glucuronic acid did not reduce the silver or lead ions under microwave radiation. We therefore hypothesize that perhaps the proteins or sugar alcohols in the plant matrix were serving as the reducing agents. We believe that this technique in which adsorption and reduction are combined using microwave radiation can be applied for removing and recycling metallic ions from contaminated water and industrial wastewater.  相似文献

ObjectiveTo compare the post-operative analgesic effects of butorphanol or firocoxib in dogs undergoing ovariohysterectomy.Study designProspective, randomized, blinded, clinical trial.AnimalsTwenty-five dogs >1 year of age.MethodsDogs received acepromazine intramuscularly (IM), 0.05 mg kg?1 and either butorphanol IM, 0.2 mg kg?1 (BG, n = 12) or firocoxib orally (PO), 5 mg kg?1 (FG, n = 13), approximately 30 minutes before induction of anesthesia with propofol. Anesthesia was maintained with isoflurane. Ovariohysterectomy was performed by the same surgeon. Pain scores using the dynamic and interactive visual analog scale (DIVAS) were performed before and at 1, 2, 3, 4, 6, 8 and 20 hours after the end of surgery by one observer, blinded to the treatment. Rescue analgesia was provided with morphine (0.5 mg kg?1) IM and firocoxib, 5 mg kg?1 (BG only) PO if DIVAS > 50. Groups were compared using paired t-tests and Fisher’s exact test (p < 0.05). Data are presented as mean ± SD.ResultsThe BG required significantly less propofol (BG: 2.6 ± 0.59 mg kg?1; FG: 5.39 ± 0.7 mg kg?1) (p < 0.05) but the anesthesia time was longer (BG: 14 ± 6, FG: 10 ± 4 minutes). There were no differences for body weight (BG: 7.9 ± 5.0, FG: 11.5 ± 4.6 kg), sedation scores, and surgery and extubation times (BG: 10 ± 2, 8 ± 5 minutes; FG: 9 ± 3, 8 ± 4 minutes, respectively) (p > 0.05). The FG had significantly lower pain scores than the BG at 1, 2 and 3 hours following surgery (p < 0.05). Rescue analgesia was administered to 11/12 (92%) and 2/13 (15%) dogs in the BG and FG, respectively (p < 0.05).Conclusion and clinical relevanceFirocoxib produced better post-operative analgesia than butorphanol. Firocoxib may be used as part of a multimodal analgesia protocol but may not be effective as a sole analgesic.  相似文献
