Current theory expects that fungi, on the one hand, are spatially ubiquitous but, on the other, are more susceptible than bacteria to disturbance such as land use change due to dispersal limitations. This study examined the relative importance of location and land use effects in determining soil fungal community composition in south-eastern Australia. We use terminal restriction fragment length polymorphism (T-RFLP; primer pair ITS1-F–ITS4) and multivariate statistical methods (NMDS ordinations, ANOSIM tests) to compare relative similarities of soil fungal communities from nine sites encompassing three locations (ca 50–200 km apart) and four land uses (native eucalypt forest, Pinus radiata plantation, Eucalyptus globulus plantation, and unimproved pasture). Location effects were generally weak (e.g. ANOSIM test statistic R  0.49) and were, in part, attributed to minor differences in soil texture. By contrast, we found clear and consistent evidence of land use effects on soil fungal community composition (R  0.95). That is, soils from sites of the same land use grouped together in NMDS ordinations of fungal composition despite geographic separations of up to ca 175 km (native eucalypt forests) and 215 km (P. radiata plantations). In addition, different land uses from the same location were clearly separate in NMDS ordinations, despite, in one case, being just 180 m apart and having similar land use histories (i.e. P. radiata versus E. globulus plantation both established on pasture in the previous decade). Given negligible management of all sites beyond the early establishment phase, we attribute much of the land use effects to changes in dominant plant species based on consistent evidence elsewhere of strong specificity in pine and eucalypt mycorrhizal associations. In addition, weak to moderate correlations between soil fungal community composition and soil chemical variables (e.g. Spearman rank correlation coefficients for individual variables of 0.08–0.32), indicated a minor contributing role of vegetation-mediated changes in litter and soil chemistry. Our data provide evidence of considerable plasticity in soil fungal community composition over time spans as short as 6–11 years. This suggests that – at least within geographic zones characterised by more-or-less contiguous forest cover – soil fungal community composition depends most on availability of suitable habitat because dispersal propagules are readily available for colonisation after land use change.  相似文献

To evaluate the utility of random-effects linear modeling for herd-level evaluation of trace mineral status, we performed a retrospective analysis of the results for trace mineral testing of bovine liver samples submitted to the Michigan State University Veterinary Diagnostic Laboratory between 2011 and 2017. Our aim was to examine random-effects models for their potential utility in improving interpretation with minimal sample numbers. The database consisted of 1,658 animals distributed among 121 herds. Minerals were assayed by inductively coupled plasma–mass spectroscopy, and included cobalt, copper, iron, molybdenum, manganese, selenium, and zinc. Intraclass correlation coefficients for each mineral were significantly different (p < 0.001) from zero and ranged from 0.38 for manganese to 0.82 for selenium, indicating that the strength of herd effects, which are presumably related to diet, vary greatly by mineral. Analysis of the distribution and standard errors of best linear unbiased predictor (BLUP) values suggested that testing 5–10 animals per herd could place herds within 10 percentile units across the population of herds with 70–95% confidence, the confidence level varying among minerals. Herd means were generally similar to BLUPs, suggesting that means could be reasonably compared to BLUPs with respect to the distributions reported here. However, caution in interpreting means relative to BLUPs should be exercised when animal numbers are small, the standard errors of the means are large, and/or the values are near the extremes of the distribution.  相似文献

A factorial experiment (3 x 4 x 2 x 3) was conducted in programmable incubators to investigate interaction between the effects of rainfall amount, rainfall distribution and evaporation rate on development of Haemonchus contortus to L3. Sheep faeces containing H. contortus eggs were incubated on sterilised soil under variable temperatures typical of summer in the Northern Tablelands of NSW, Australia. Simulated rainfall was applied in 1 of 3 amounts (12, 24 or 32 mm) and 4 distributions (a single event on the day after deposition, or the same total amount split in 2, 3 or 4 equal events over 2, 3 or 4 days, respectively). Samples were incubated at either a Low or High rate of evaporation (Low: 2.1-3.4 mm/day and High: 3.8-6.1 mm/day), and faeces and soil were destructively sampled at 4, 7 and 14 days post-deposition. Recovery of L3 from the soil (extra-pellet L3) increased over time (up to 0.52% at day 14) and with each increment of rainfall (12 mm: <0.01%; 24 mm: 0.10%; 32 mm: 0.45%) but was reduced under the High evaporation rate (0.01%) compared with the Low evaporation rate (0.31%). All rainfall amounts yielded significantly different recoveries of L3 under Low evaporation rates but there was no difference between the 12 and 24 mm treatments under the High evaporation rate. The distribution of simulated rainfall did not significantly affect recovery of infective larvae. Faecal moisture content was positively associated with L3 recovery, as was the ratio of cumulative precipitation and cumulative evaporation (P/E), particularly when measured in the first 4 days post-deposition. The results show that evaporation rate plays a significant role in regulating the influence of rainfall amount on the success of L3 transmission.  相似文献

Fever is a non-specific host defense mechanism that comprises part of the innate immune response. Innate immune function is thought to be an important adaptive immunological response to infection because it occurs across a broad diversity of phyla. Some reptiles can mount a febrile response, despite the fact that their internal body temperatures (T(b)s) are, to some extent, controlled by the environmental temperatures in which they live. This study was undertaken to determine if LPS would induce fever in green anole lizards (Anolis carolinensis). Lizards were maintained in thermal gradients (22-45 degrees C) with a 12-h diurnal cycle. anoles were injected with LPS, pyrogen-free saline, or left untreated, and their T(b)s were recorded every 15min using internal cloacal probes. All lizards showed a diurnal periodicity in T(b) characterized by decreased temperatures during the scotophase (dark hours) and higher temperatures during the photophase (light phase). Anoles injected with LPS exhibited a hypothermic response, relative to untreated and saline-injected animals. The response varied from 2.1 to 4.6 degrees C lower than control lizards. The hypothermic response was initiated within 12-24h of LPS injection, and continued for 3 days after treatment. However, the anapyrexic response was observed primarily during scotophases, with photophase hypothermia observed only on the first day after LPS injection.  相似文献

T‐cell lymphomas (TCL) are a diverse group of neoplasms with variable diagnostic features, pathophysiologies, therapeutic responses and clinical outcomes. In dogs, TCL includes indolent and aggressive tumours such as T‐zone lymphoma (TZL) and peripheral T‐cell lymphoma (PTCL), respectively. Delineation of molecular subtypes and investigation into underlying pathophysiologies of aggressive TCLs remains inadequate. We investigate the correlations between flow cytometry and histopathology of 73 cases of nodal TCL. The majority of cases (82.2%) were characterized as CD4+ TCL by flow cytometry. Fewer cases were classified as CD8+ TCL (6.8%) or CD4?CD8? TCL (11.0%). All cases, regardless of immunophenotype, exhibited conserved histologic features consistent with the WHO classification of PTCL. Histologic subsets of PTCL corresponding to immunophenotypic features were not identified. Neoplastic cell size determined by flow cytometry correlated significantly with mitotic rate. RNA‐seq was performed on a subset of CD4+ PTCL cases (n = 6) and compared with sorted control CD4+ T‐cells. The gene expression pattern of CD4+ PTCL was similar between all cases regardless of breed. PTCL was enriched in pathways representing G‐coupled protein receptor signalling, extracellular matrix remodelling and vascular development, immune signalling and mitotic activity. Furthermore, global gene expression changes were consistent with downregulation of PTEN signalling and upregulation of the MTOR‐PI3K‐ATK axis. In this study, we evaluated the correlations between flow cytometry, histopathology and gene expression within a large cohort of nodal TCLs. We further demonstrate the ability of flow cytometry to identify a subtype of T‐cell lymphoma, CD4+ PTCL, with a uniform histomorphology and gene expression profile.  相似文献
