Multiple whitish nodules in the thoracic cavity at the site of the thymus were observed in a 101-week-old male ICR mouse. In a histopathological examination, the neoplastic cells were predominantly fusiform in shape and proliferated in sarcomatoid growth patterns. Some neoplastic cells showed epithelial growth patterns, such as the ductal structures. Mitotic figures were frequently seen, and small necrotic foci and invasion to adjacent thoracic organs were noted. In Alcian blue staining, bluish materials were observed between fusiform-shaped cells and in some of the lumens of the ductal structures. In immunohistochemistry, both fusiform-shaped and ductal structure-forming cells were positive for vimentin and weakly positive to positive for cytokeratin. Based on the aforementioned findings, the thoracic nodules were diagnosed as a mixed type of malignant mesothelioma. Hatano high- and low-avoidance rats (HAA and LAA strains, respectively) were selected and bred according to the avoidance rate in a shuttle-box task. Although they have clear strain differences in ovarian function, their endocrine mechanisms still remain to be clarified. Differences in female reproductive endocrinology between the strains were investigated by means of measuring the plasma concentration of reproductive hormones during the estrous cycle. LAA rats showed approximately threefold lower basal and surge levels of LH, a more than fourfold lower level of FSH surges and higher levels of inhibin A and inhibin B during the estrous cycle compared with the levels seen in HAA rats. The concentration of estradiol-17β in the proestrous stage was significantly lower in LAA rats than in HAA rats. Additionally, LH and FSH secretions from primary cultured anterior pituitary cells with or without in vitro GnRH stimulation were lower in the cells derived from LAA rats and, in terms of FSH secretion, were unresponsive to GnRH in contrast to cells derived from HAA rats. Although an increased number of preantral follicles in diestrus were observed in LAA rats, number of hCG-induced ovulation was lower in LAA rats. LAA rats may have much more follicle growth during the early stage of folliculogenesis, but most follicles might not grow into mature follicles. These results strongly suggest that the strain difference in ovarian function of these two Hatano rats is due to the difference in the regulation of hypothalamo-hypophyseal system for gonadotropins secretion. 相似文献

The objective of this study was to investigate the cellular immunolocalization of inhibin a and inhibin/activin (β(A) and β(B)) subunits in the muskrat testes and scented glands during the breeding season. Inhibin α and inhibin/activin (β(A) and β(B)) subunits were expressed in Sertoli cells and Leydig cells of testes and glandular cells of scented glands, respectively. Also, positive signals of inhibin α and inhibin/activin (β(A) and β(B)) subunits by Western blotting were both observed in testicular and scented glandular tissues. These results suggested that the testes and scented glands of the muskrats had the ability to synthesize inhibins and activins and that activins and inhibins might play an important role in testicular and scented glandular function in muskrats. 相似文献

We have characterized the mitochondrial cox1 gene copies in two apple cultivars ‘Golden Delicious’ and ‘Delicious’. Both the cultivars contained an intact copy and a truncated copy of cox1. The intact ‘Golden Delicious’ and ‘Delicious’ cox1 genes, designated G-cox1 and D-cox1, respectively, were both found to be actually transcribed to give an RNA of approximately 1.7 kb. The two intact cox1 and two truncated copies (G-φcox1 and D-φcox1) shared a common 1115-bp segment flanked by four combinations of two different 5′- and 3′-sequences. PCR assay demonstrated that the configurations bearing G-cox1 and G-φcox1 existed in substoichiometric amounts within the mitochondrial genome of ‘Delicious’ whereas substoichiometric molecules carrying D-φcox1 were present in the ‘Golden Delicious’ mitochondrial genome. Although ancestor/descendant relationships cannot be inferred between the G-cox1 and D-cox1 arrangements, the results led us to hypothesize that (1) the 1115-bp segment containing part of the progenitor cox1 was duplicated, thereby generating a pseudo-cox1 copy, and (2) this was followed by homologous recombination across a portion of the 1115-bp repeats which gave rise to the descendant cox1 and pseudo-cox1 arrangements. 相似文献

T cells are the dominant lymphocytes in the endometrium and are considered to play a
crucial role in implantation and in the maintenance of gestation through cytokine
production and immune regulation. The mechanisms underlying immunoregulation at the
feto-maternal interface are still obscure for this complex system. Understanding the
role of T cells is a key factor in understanding the endometrial immune system. In
this study, the distribution of endometrial CD3+ T cells in bovines was
examined by immunohistochemical analysis. The estrous cycle and gestation was divided
into 4 stages, and the number of CD3+-positive T cells was counted in each
stage. CD3+ cells were found in the endometrium in significant numbers
throughout the estrous cycle and were mostly located in the subepithelial area. The
number of CD3+ cells significantly increased in the early and mid-luteal
phases but decreased after implantation with the progression of gestation. No T cells
were found in the placentome or specifically in the tissues near the fetus, including
the trophoblastic area. In addition, very few T cells were found in stromal regions
close to the myometrium of the endometrium. These findings suggest that
downregulation of bovine endometrial CD3+ T-cell functions is closely
related to the successful maintenance of gestation in a spatiotemporal manner. 相似文献

Although endoscopy is the definitive diagnostic method for the detection of colonic
ulcers, the equipment required for performing the test is costly and difficult to use.
Therefore, a simple cost-effective and reliable screening test for intestinal tract
bleeding is needed. To this end, we measured carbonic anhydrase isozymes (CA-I and CA-II)
originating from erythrocytes by ELISA in order to determine if they could be used as
markers of occult blood in feces. For fecal extract preparation, 2 g of feces were mixed
with 4 ml of 0.01 M Tris-HCl (pH 8.0) containing 0.01% thimerosal. The concentrations of
CA-I and CA-II in the fecal samples of 13 clinically normal racehorses were found to be
30.0 ± 10.0 and 34.0 ± 13.0 ng/ml, respectively. Increased concentrations of CA-I were
detected in the fecal samples of 5 horses after blood administration; however, no increase
was observed in CA-II. The concentrations of CA-I and CA-II in the fecal samples of 88
racehorses with clinical signs of equine gastric ulcer syndrome (EGUS) were 115.3 ± 79.0
and 41.0 ± 42.0 ng/ml, respectively. Thus, our results indicate that CA isozymes can be
useful as markers of occult blood in the fecal samples of horses with intestinal tract
