This study investigates the association of semen traits with boar fertility. The fertility outcome (farrowing rate – FR and total piglets born – TB) of 14 boars was obtained from a field trial conducted during 10 week of breeding period on a commercial farm using multiparous sows (n = 948) through single‐sire mating with 2 × 109 motile sperm cells per artificial insemination (AI) dose. Sperm motion parameters, evaluated with computer‐assisted semen analysis system in raw and stored semen at 17°C for 240 h, in addition to morphological sperm defects, measured on the collection day, were included in the analysis to determine which semen traits were important to discriminate the fertility potential of ejaculates from these boars. The data underwent multivariate cluster, canonical and discriminant analyses. Four clusters of boars were formed based on fertility outcome. One boar, with the lowest FR and TB values (89.7% and 11.98), and two boars, with the highest FR and TB values (97.8% and 14.16), were placed in different clusters. The other boars were separated in two distinct clusters (four and seven boars), including boars with intermediate TB (12.64 and 13.22) but divergent values for FR (95.9% vs 91.8%). Semen traits with higher discriminatory power included total motility, progressive motility, amplitude of lateral head displacement and cytoplasmatic droplets. Through multivariate discriminant analysis, more than 80% of the 140 ejaculates were correctly classified into their own group, showing that this analysis may be an efficient statistical tool to improve the discrimination of potential fertility of boars. Nevertheless, the validation of the relationship between fertility and semen traits using this statistical approach needs to be performed on a larger number of farms and with a greater number of boars.  相似文献

We developed and tested a new method to collect CO2 from the surface to deep layers of a peatland for radiocarbon analysis. The method comprises two components: i) a probe equipped with a hydrophobic filter that allows entry of peat gases by diffusion, whilst simultaneously excluding water, and, ii) a cartridge containing zeolite molecular sieve that traps CO2 passively. We field tested the method by sampling at depths of between 0.25 and 4 m at duplicate sites within a temperate raised peat bog. CO2 was trapped at a depth-dependent rate of between ∼0.2 and 0.8 ml d−1, enabling sufficient CO2 for routine 14C analysis to be collected when left in place for several weeks. The age of peatland CO2 increased with depth from modern to ∼170 BP for samples collected from 0.25 m, to ∼4000 BP at 4 m. The CO2 was younger, but followed a similar trend to the age profile of bulk peat previously reported for the site (Langdon and Barber, 2005). δ13C values of recovered CO2 increased with depth. CO2 collected from the deepest sampling probes was considerably 13C-enriched (up to ∼+9‰) and agreed well with results reported for other peatlands where this phenomenon has been attributed to fermentation processes. CO2 collected from plant-free static chambers at the surface of the mire was slightly 14C-enriched compared to the contemporary atmosphere, suggesting that surface CO2 emissions were predominantly derived from carbon fixed during the post-bomb era. However, consistent trends of enriched 13C and depleted 14C in chamber CO2 between autumn and winter samples were most likely explained by an increased contribution of deep peat CO2 to the surface efflux in winter. The passive sampling technique is readily portable, easy to install and operate, causes minimal site disturbance, and can be reliably used to collect peatland CO2 from a wide range of depths.  相似文献

In the horse, breeding induces a transient endometrial inflammation. A subset of mares are unable to resolve this inflammation, and they are considered susceptible to persistent mating‐induced endometritis PMIE Select seminal plasma proteins cysteine‐rich secretory protein‐3 (CRISP‐3) and lactoferrin have been shown to affect the innate immune response to sperm in vitro. The objective of this study was to determine whether the addition of CRISP‐3 and lactoferrin at the time of insemination had an effect on the mRNA expression of endometrial cytokines in susceptible mares after breeding. Six mares classified as susceptible to PMIE were inseminated during four consecutive oestrous cycles with treatments in randomized order of: 1 mg/ml CRISP‐3, 150 μg/ml lactoferrin, seminal plasma (positive control) or lactated Ringer's solution (LRS; negative control) to a total volume of 10 ml combined with 1 × 109 spermatozoa pooled from two stallions. Six hours after treatment, an endometrial biopsy was obtained for qPCR analysis of selected genes associated with inflammation (pro‐inflammatory cytokines interleukin (IL)‐1β, IL‐8, tumour necrosis factor (TNF)‐α, interferon (INF)‐γ, anti‐inflammatory cytokines IL‐1RN and IL‐10, and inflammatory‐modulating cytokine IL‐6). Seminal plasma treatment increased the mRNA expression of IL‐1β (p = .019) and IL‐8 (p = .0068), while suppressing the mRNA expression of TNF (p = .0013). Lactoferrin also suppressed the mRNA expression of TNF (p = .0013). In conclusion, exogenous lactoferrin may be considered as one modulator of the complex series of events resulting in the poorly regulated pro‐inflammatory response seen in susceptible mares.  相似文献

Species diversity has declined in ecosystems worldwide as a result of habitat fragmentation, eutrophication, and land-use change. If such decline is to be halted ecological mechanisms that restore or maintain biodiversity are needed. Two long-term field experiments were performed in native grassland to assess the effects of fire, nitrogen addition, and grazing or mowing on plant species diversity. In one experiment, richness declined on burned and fertilized treatments, whereas mowing maintained diversity under these conditions. In the second experiment, loss of species diversity due to frequent burning was reversed by bison, a keystone herbivore in North American grasslands. Thus, mowing or the reestablishment of grazing in anthropogenically stressed grasslands enhanced biodiversity.  相似文献

Canine high-risk surgical patients were identified, and the variables that distinguished between survivors and nonsurvivors were determined. Fifty-two nonsurvivors and 176 survivors were randomly drawn from a population of 615 dogs undergoing general surgical procedures and entering the intensive care unit. Forty-two variables were examined to determine which variables were significantly different between nonsurvivors and survivors. Properative variables that were significantly different were age, hospital days prior to surgery, the number of concurrent diseases, American Society of Anesthesiologists (ASA) status, PCV, TP, platelet count, albumin concentration, alkaline phosphatase activity, bilirubin concentration, and surgery site (laparotomy = highest risk). Age, PCV, TP, platelet count, albumin concentration, alkaline phosphatase activity, and bilirubin concentration were then examined, using stepwise logistic regression for their ability to predict mortality in a separate population of 169 canine patients undergoing laparotomy. Age, TP, and platelet count were identified as predictive variables.  相似文献

Endotoxic Shock     
Endotoxic shock is a complex phenomenon resulting from systemic release of inflammatory mediators. Endotoxin interacts with inflammatory cells, platelets, and vascular endothelium. Cytokines, such as tumor necrosis factor and interleukins, and lipid mediators (platelet activating factor, thromboxane, prostacyclin, leukotrienes) are released. These primary mediators act synergistically to cause many of the harmful effects associated with endotoxemia. Multiple secondary mediators are released in response to the primary mediators, compounding the damage. The end result is the species-specific clinical syndrome recognized as endotoxemia.  相似文献
