Intramammary infusion of Bifidobacterium breve (B. breve)-induced somatic cell (SC) counts, chemiluminescent response (CL), lactoferrin (LF) concentrations and mastitis-causing pathogens from quarters with subclinical mastitis were measured to evaluate innate immune response of mammary glands in dairy cows at 3 to 4 weeks before drying off. SC counts in 7 quarters of 7 control cows and 5 quarters of 6 cows with mastitis increased markedly on day 1 and SC values in control cows were significantly (P<0.05) increased and returned to pre-infusion levels on day 5 after B. breve-infusion. CL values in both groups increased markedly on day 1 and then decreased after B. breve-infusion; however, CL values in cows with mastitis did not return to normal levels on day 5 and at postpartum. The CL values were highly correlated with their SC counts in milk from both groups. LF concentrations increased toward day 3 after B. breve-infusion and were higher in cows with mastitis. B. breve-infusion eliminated 16.6% (1/6) of pathogens from 6 quarters with chronic subclinical mastitis. B. breve-induced SC responses in quarters from 3 cows with mastitis showed characteristic patterns of recovery, persistent and new infections. B. breve-induced SC counts in quarters from the cows in the pre-drying off were lower (25.7–70.6%) than those of the cows in mid-lactation. The intrinsic innate immune response in cows on pre-drying off may be decreased and appears to be insufficient to eliminate pathogens from mammary gland in the pre-drying off.  相似文献

This study investigates the occurrence of diarrhetic shellfish toxins (DSTs) and their producing phytoplankton species in southern Brazil, as well as the potential for toxin accumulation in co-occurring mussels (Perna perna) and octopuses (Octopus vulgaris). During the spring in 2012 and 2013, cells of Dinophysis acuminata complex were always present, sometimes at relatively high abundances (max. 1143 cells L−1), likely the main source of okadaic acid (OA) in the plankton (max. 34 ng L−1). Dinophysis caudata occurred at lower cell densities in 2013 when the lipophilic toxins pectenotoxin-2 (PTX-2) and PTX-2 seco acid were detected in plankton and mussel samples. Here, we report for the first time the accumulation of DSTs in octopuses, probably linked to the consumption of contaminated bivalves. Perna perna mussels were consistently contaminated with different DSTs (max. 42 µg kg−1), and all octopuses analyzed (n = 5) accumulated OA in different organs/tissues: digestive glands (DGs) > arms > gills > kidneys > stomach + intestine. Additionally, similar concentrations of 7-O-palmytoyl OA and 7-O-palmytoly dinophysistoxin-1 (DTX-1) were frequently detected in the hepatopancreas of P. perna and DGs of O. vulgaris. Therefore, octopuses can be considered a potential vector of DSTs to both humans and top predators such as marine mammals.  相似文献

The effects of temperature on growth and production of Lipophilic Toxins (LT) by a monoclonal culture of Dinophysis caudata was studied. The cell density of D. caudata increased significantly with increasing temperature, and was the highest under 27, 30, and 32.5 °C. Temperature affected the average specific growth rate (µ) during the exponential growth phase (EG), which increased from 15 °C to 30 °C, and then decreased at 32.5 °C. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) revealed that this strain of D. caudata produced only pectenotoxin-2 (PTX-2) whose concentration increased significantly with incubation period, except at 32.5 °C. It was significantly different between temperatures ≤18 °C, ≥21 °C, and 32.5 °C. The cellular toxin production (CTP, pg·cell−1·day−1) showed variation with growth phase and temperature, except at 32.5 °C. The average net toxin production (Rtox) was not affected by temperature. During EG, the average specific toxin production rate (µtox) increased significantly with increase in temperature, reaching a peak of 0.66 ± 0.01 day−1 at 30 °C, and then decreased. Over the entire growth span, µtox was significantly correlated to µ, and this correlation was most significant at 27 and 30 °C. During EG, µtox was affected by both temperature and growth. This study shows that temperature affects growth and toxin production of this strain of D. caudata during EG. In addition, a positive correlation was found between toxin production and growth.  相似文献

To identify the effects of whole crop maize silage (MS) as a substitute for rice straw (RS) on feed intake and milk production of mid-late lactating buffalo and cattle in Tarai, Nepal, eight Murrah and eight Jersey-Hariana were fed the basal diet, RS ( ad libitum ) with concentrate (0.68% of bodyweight [BW] on a dry matter [DM] basis). A 4 × 4 Latin square design experiment was conducted in each animal species with graded levels of MS substitution for RS (0%, T1; 33%, T2; 67%, T3 and 100%, T4). The MS had higher digestibility and total digestible nutrient (TDN) than RS. The DM intake per BW of the both species was highest in T3. The substitution of MS for RS increased the crude protein intake and the TDN intake in the both species. Although the buffalo showed the highest milking performance in T4, the cattle showed no significant differences in their milking performance among the treatments. The substitution of MS for RS improved the feed intake and milk production in the buffalo. On the other hand, the milk yield was not raised in the cattle, though the feed intake was increased by the substitution.  相似文献

Using a preference test and operant conditioning in a Y-maze, this experiment examined the ability of heifers to discriminate between their own familiar herdmates and member(s) of an unfamiliar group. Sixteen Danish Friesian heifers, eight older animals (360.6 ± 24.2 days of age) and eight younger ones (190.1 ± 14.1 days of age) were used. Each age group was further divided into two experimental groups. Members of each of these groups were housed together in small pens before the experiments began. In experiment 1, each of the 16 animals was allowed to approach either a familiar or an unfamiliar individual in the Y-maze. The test was repeated 12 times, with a different unfamiliar subject for each test. In experiment 2, eight heifers were individually tested in a conditioning experiment to examine whether they could learn to discriminate between a group of their three herdmates and a group of three unfamiliar heifers. Test animals were rewarded when they chose their own group. In experiment 1, heifers did not show a preference between familiar and unfamiliar individuals. Interestingly the younger stimulus heifers but not the test animals showed an ability to discriminate between unfamiliar animals by vocalizing. In experiment 2, four of the eight test animals achieved the criterion for successful discrimination between the familiar and unfamiliar group ( P  < 0.003: binomial law). There was no age group difference in the ability to discriminate between familiar and unfamiliar animals. In conclusion, heifers did not show a preference toward familiar or unfamiliar individuals; but after conditioning, some heifers could learn to discriminate between familiar and unfamiliar groups.  相似文献

A necrotrophic pathogen, the tomato pathotype of Alternaria alternata (Aa) causes Alternaria stem canker on tomato. Its pathogenicity depends on the production of host-specific AAL-toxin. Pre-inoculation with nonpathogenic Aa or pretreatment an elicitor prepared from Aa reduced disease symptoms by the pathogen. Salicylic acid (SA)- and jasmonic acid (JA)-dependent defense responses in tomato are not involved in the resistance to the pathogen induced by nonpathogenic Aa. The results suggest that an alternative and unknown signaling pathway independent of SA- and JA-signaling might modulate the induced resistance by activating the expression of the multiple defense genes.  相似文献

Four Thai native steers were used to determine the adequate levels of supplementary desalted mother liquor (DML) for energy and nitrogen balances and ruminal fermentation. The crude protein and sodium chloride contents of DML were 25.5% and 60.3% on a dry matter (DM) basis, respectively. A 4 × 4 Latin square design experiment was conducted by adding different amounts of DML to three experimental diets (T1: 1.1%, T2: 2.2%, T3: 3.4% sodium chloride concentration with supplementary DML on a DM basis) and comparing their effects with those of a control diet (C) containing 1.0% commercial salt on a DM basis. The animals were given the experimental diets and rice straw daily at 1.2% and 0.8% of body weight, respectively, on a DM basis. No significant differences in the apparent digestibility of nutrients were observed among treatments. T3 achieved the lowest nitrogen retention (< 0.05), followed by C, T2 and T1. The ratios of energy retention to gross energy were higher in T1 and T3 than T2, and that in C was lowest (< 0.05). Supplementary NaCl concentration at 1% and 2% can be replaced with DML without an adverse effect on the digestibility of nutrients or on the nitrogen and energy retention.  相似文献

Hydrophobins are small secreted proteins unique to filamentous fungi. In this study, we cloned and characterized the class I hydrophobin gene BcHpb1 in the necrotrophic pathogen Botrytis cinerea. The BcHpb1 protein consisted of 117 amino acids. Similar to class I hydrophobins from other fungi, BcHpb1 contains eight conserved cysteine residues. The hydropathy plot of the BcHpb1 amino acid sequence was characteristic of a class I hydrophobin. These results indicated that the BcHpb1 gene encodes a class I hydrophobin. Vegetative growth of ΔBcHpb1 strains, null mutants of BcHpb1, was similar to that of the wild-type strain as were the conidiophores, conidia, appressoria and virulence on host plants. However, adherence of ΔBcHpb1 strains to hydrophobic surfaces was greatly reduced, implying that BcHpb1 is important for the hydrophobicity of conidia and that BcHpb1 may be required to adhere to plant surfaces under certain environmental conditions.  相似文献

To demonstrate the effect of excess soil moisture on the decline of a coastal Pinus thunbergii stand in Oshamanbe, southwestern Hokkaido, Japan, soil moisture content was monitored for 4 years. The saturated hydraulic conductivities (K S) of different soil types (coastal sand, supplied topsoil, and buried concreted andosol) and the distribution of the buried concreted andosol layer were investigated. We also examined needle length to verify the real-time response of P. thunbergii to excess soil moisture. Soil moisture content at the heavily damaged site was more heterogeneous than that at the slightly damaged site, and a sensor near the ground always reported a higher soil moisture content at the heavily damaged site than at the slightly damaged site. The buried concreted andosol layer was always found at the heavily damaged site. The K S of the andosol layer was 10−5, suggesting that this layer is less permeable to water, leading to excess soil moisture at this site. P. thunbergii needles from the heavily damaged site were shorter than those from the slightly damaged site, possibly because of water stress. Together with other symptoms observed at the study sites, i.e., crown dieback and intense lateral growth, this information leads us to conclude that the decline of P. thunbergii stands at the heavily damaged site in Oshamanbe was caused by excess soil moisture due to the less permeable buried concreted andosol layer.  相似文献
