The production of piscine viruses, in particular of koi herpesvirus (KHV, CyHV‐3) and infectious salmon anaemia virus (ISAV), is still challenging due to the limited susceptibility of available cell lines to these viruses. A number of cell lines from different fish species were compared to standard diagnostic cell lines for KHV and ISAV regarding their capability to exhibit a cytopathic effect (CPE) and to accumulate virus. Two cell lines, so far undescribed, appeared to be useful for diagnostic purposes. Fr994, a cell line derived from ovaries of rainbow trout (Oncorhynchus mykiss), produced constantly high ISA virus (ISAV) titres and developed a pronounced CPE even at high cell passage numbers, while standard cell lines are reported to gradually loose these properties upon propagation. Another cell line isolated from the head kidney of common carp (Cyprinus carpio), KoK, showed a KHV induced CPE earlier than the standard cell line used for diagnostics. A third cell line, named Fin‐4, established from the fin epithelium of rainbow trout did not promote efficient replication of tested viruses, but showed antigen sampling properties and might be useful as an in vitro model for virus uptake or phagocytosis.  相似文献

Restriction fragment length polymorphisms were generated for 52 species of lignicolous European polypores belonging to 31 different genera. An enzymatically amplified portion of the nuclear-encoded ribosomal DNA was found to be about 1800–1900 bp long for most taxa. After digestion with Hpa II, 44 distinct phenotypes were detected for the species under study; application of the additional restriction endonucleases, Hin 6 I and Hinf I, resulted in 48 species-specific and two genera-specific phenotypes. The procedure described should allow for a reliable identification of unknown mycelia of lignicolous fungi.  相似文献

Oxidative stress results from a disruption of the prooxidant/antioxidant cellular balance and monitoring free radical status becomes an interesting challenge in animal and human nutrition. In the present work, merits and limitations of different analytical techniques (HPLC, GC-MS, fluorometric and colourometric assays, ELISA, gel electrophoresis) for the measurement of radical mediated alterations in the cellular integrity of lipids (malondialdehyde, hydrocarbon gases, F2-isoprostanes) proteins (protein carbonyls, 3-nitrotyrosine) and DNA (8-hydroxy-2'-deoxyguanosine) are discussed. Besides these indirect methods, owing to the fact that free radicals are paramagnetic, electron paramagnetic resonance spectroscopy combined with spin trapping has become a valuable tool to directly assess and to better understand the mechanisms of free radical reactions. With this approach a radical that is too short-lived to be detected, adds to a spin-trapping agent to form a relatively long-lived radical adduct. Information obtained from the hyperfine splitting of the spin-trapped adduct can provide identification and quantification of the originally generated free radicals.  相似文献

Studies of the effects of above-ground herbivory on soil organisms and decomposer food webs, as well as the processes that they regulate, have largely concentrated on the effects of non-living inputs into the soil, such as dung, urine, body parts and litter. However, there is an increasing body of information which points to the importance of plant physiological responses to herbivory in regulating soil organisms and therefore, implicitly, key soil processes such as decomposition and nutrient mineralisation. In this review we identify the mechanisms by which foliar herbivory may indirectly affect the soil biota and associated below-ground processes through affecting plants, so as to better understand the nature of interactions which exist between above-ground and below-ground biota. We consider two broad pathways by which above-ground foliar herbivory may affect soil biotic communities. The first of these occurs through herbivore effects on patterns of root exudation and carbon allocation. These effects manifest themselves either as short-term changes in plant C allocation and root exudation or as long-term changes in root biomass and morphology. Evidence suggests that these mechanisms positively influence the size and activity of the soil biotic community and may alter the supply of nutrients in the rhizosphere for plant uptake and regrowth. The second of these involves herbivores influencing soil organisms through altering the quality of input of plant litter. Possible mechanisms by which this occurs are through herbivory enhancing nitrogen contents of root litter, through herbivory affecting production of secondary metabolites and concentrations of nutrients in foliage and thus in leaf litter and through selective foliar feeding causing shifts in plant community structure and thus the nature of litter input to the soil. While the effects of herbivory on soil organisms via plant responses may be extremely important, the directions of these effects are often unpredictable because several mechanisms are often involved and because of the inherently complex nature of soil food-web interactions; this creates obvious difficulties in developing general principles about how herbivory affects soil food-webs. Finally, it is apparent that very little is understood on how responses of soil organisms to herbivory affect those ecosystem-level processes regulated by the soil food-web (e.g. decomposition, nutrient mineralisation) and that such information is essential in developing a balanced understanding about how herbivory affects ecosystem function.  相似文献

Atmospheric carbon monoxide (CO) exhibits mass-independent fractionation in the oxygen isotopes. An 17O excess up to 7.5 per mil was observed in summer at high northern latitudes. The major source of this puzzling fractionation in this important trace gas is its dominant atmospheric removal reaction, CO + OH --> CO2 + H, in which the surviving CO gains excess 17O. The occurrence of mass-independent fractionation in the reaction of CO with OH raises fundamental questions about kinetic processes. At the same time the effect is a useful marker for the degree to which CO in the atmosphere has been reacting with OH.  相似文献

We used a farm-level modeling approach to estimate on-farm compliance costs and environmental effects of a grassland extensification scheme in the district of Ostprignitz-Ruppin, Germany. The behavior of the regional farm population (n = 585) consisting of different farm types with different production orientations and grassland types was modeled under the presence and absence of the grassland extensification scheme using the bio-economic model MODAM. Farms were based on available accountancy data and surveyed production data, while information on farm location within the district was derived from a spatial allocation procedure. The reduction in total gross margin per unit area was used to measure on-farm compliance costs. A dimensionless environmental index was used to assess the suitability of the scheme to reduce the risk of nitrate-leaching.Calculated on-farm compliance costs and environmental effects were heterogeneous in space and farm types as a result of different agricultural production and site characteristics. On-farm costs ranged from zero up to almost 1500 Euro/ha. Such high costs occurred only in a very small part of the regional area, whereas the majority of the grassland had low on-farm costs below 50 Euro/ha. Environmental effects were moderate and greater on high-yield than on low-yield grassland. The low effectiveness combined with low on-farm costs in large parts of the region indicates that the scheme is not well targeted. The soft scheme design results from an attempt to achieve environmental and rural development objectives with only one scheme. Improving the efficiency of the scheme would require designing separate instruments for the two distinct objectives. This is in line with the Tinbergen rule, which states that consistent economic policy requires that the number of instruments equals the number of targets.  相似文献

Fluorescein isothiocyanate dextran (FITC-dextran) exchange between the primary (PCS) and secondary (SCS) circulatory systems in the Atlantic cod, Gadus morhua (Linnaeus, 1752), were studied using 20-kDa (n = 4) and 500-kDa (n = 4) FITC-dextran. In order to give a qualitative perspective of the general connection between the PCS and SCS, distribution of plasma-borne tracers (FITC-dextran) in the PCS and SCS were examined. In this study, a total of eight cod were cannulated in the ventral aorta (PCS) and dorsal cutaneous vessel (SCS), for investigation of FITC-dextran disappearance in the PCS and its subsequent appearance in the SCS. FITC-dextran of both sizes was found to be in equilibrium between the PCS and SCS in less than 20 min. This indicates a profound connection between the PCS and SCS in the Atlantic cod, and rapid mixing of tracers between the PCS and SCS. The destination of the injected 500-kDa FITC-dextran was also examined, and it was observed that of the 500-kDa FITC-dextran lost from the primary and secondary vascular systems, 63.0 ± 9.2% could be recovered from the liver  相似文献
