Our objective was to describe cow and quarter-level factors associated with drying-off, and to evaluate their impacts on new intramammary infections (IMI) during the dry period. Data from 300 cows in five research herds were collected starting 2 weeks prior to scheduled drying-off. Variables of interest included daily milk production, teat-end integrity, formation of the teat-canal keratin plug, and quarter-milk bacteriological culture results. Overall, 11% of quarters developed new IMI in the dry period; this varied by herd, parity and time of the study. Most new IMI were caused by environmental streptococci and coliform organisms (34 and 30%, respectively). Quarters that had a cracked teat-end had higher odds of developing new infections than those without cracks (15 and 10%, respectively). Quarters that formed a keratin plug early in the dry period had a lower odds than those that did not close (10 and 14%, respectively). After 6 dry weeks, 23% of quarters were still open. The hazard of quarters closing if milk production on the day prior to drying-off was >21 kg 1.8-times less.  相似文献

OBJECTIVE: To assess the impacts of the introduction of foot-and-mouth disease (FMD) and various FMD control programs in southern Thailand. ANIMALS: A native population of 562,910 cattle and 33,088 buffalo as well as 89,294 animals legally transported into southern Thailand. PROCEDURES: A quantitative risk assessment was used to ascertain the probability of FMD introduction, and an intrinsic dynamic model was used to assess impacts. Value for the transmission rate (beta) was estimated. Five scenarios created to assess the impacts of nonstructural protein (NSP) testing, mass vaccination, and culling were examined. Impacts were assessed through an examination of the estimated annual cumulative incidence (ACI) of FMD. The ACIs of various scenarios were compared by use of the Tukey Studentized range technique. RESULTS: beta was estimated at 0.115. Approximately 35,000 cases of FMD would be expected from the baseline situation. A 30% reduction of ACI was detected with the introduction of NSP antibody testing. Prophylactic vaccination resulted in an 85% reduction of ACI. Concurrent use of NSP antibody testing and vaccination reduced the ACI by 96%, and the addition of an eradication policy resulted in a slightly greater decrease in the ACI (98%). CONCLUSIONS AND CLINICAL RELEVANCE: The study used epidemiologic models to investigate FMD control interventions. Results suggested that vaccination has more impact than the use of NSP testing. Use of the NSP test reduced ACI during peak seasons, whereas vaccination diminished the underlying incidence. The best mitigation plan was an integrated and strategic use of multiple control techniques.  相似文献

AIMS: To examine the association between the interval from internal teat sealant (ITS) administration to calving and the incidence of farmer-recorded clinical mastitis in the first 30 days of lactation in pasture-based dairy heifers.

METHODS: Heifers that were administered an ITS by a single veterinary business in the South Island of New Zealand over the winter of 2014 were enrolled in a cross-sectional observational study. ITS was administered to all heifers on each participating farm on a single calendar day. The dates of calving and farmer-diagnosed clinical mastitis were recorded by farm staff. The interval from ITS administration to calving was categorised into four approximately evenly sized groups: <35, 35–48, 49–69 and >69 days. The quartile of the farm’s calving period in which each heifer calved was also investigated as a potential confounding variable. A hierarchical logistic regression model was constructed to determine the association between the interval from ITS administration to calving with the odds of clinical mastitis in the first 30 days of lactation.

RESULTS: Analysis was performed on 7,126 eligible heifers from 31 farms, with ITS administered between 9 May and 11 July 2014. The mean interval from ITS administration to calving was 52.9 (SD 24.4, min 1, max 137) days. Clinical mastitis was diagnosed in 420/7,126 (5.9 (95% CI=5.4–6.5)%) heifers between calving and day 30 of lactation. In the final multivariable model, which included calving period quartile, interval from ITS administration to calving was not associated with the odds of clinical mastitis (p=0.516). Compared to an interval from ITS administration to calving of <35 days, the adjusted OR of clinical mastitis for intervals of 35–48, 49–69 and >69 days were 0.83 (95% CI=0.59–1.17), 0.71 (95% CI=0.45–1.11) and 0.68 (95% CI=0.36–1.29), respectively.

CONCLUSIONS AND CLINICAL RELEVANCE: Within the range of intervals from ITS administration to calving observed in this study, there was no association with the odds of clinical mastitis in the first 30 days of lactation in dairy heifers. This study suggests that veterinary clinics may be able to extend their ITS administration service and treat dairy heifers earlier than the current recommendation of approximately 4 weeks before the planned start of calving.  相似文献

HistoryA 4‐year old, 500 kg Thoroughbred female horse diagnosed with bilateral forelimb laminitis and cellulitis on the left forelimb became severely painful and refractory to non‐steroidal anti‐inflammatory therapy (flunixin meglumine on days 1, 2, 3 and 4; and phenylbutazone on days 5, 6 and 7) alone or in combination with gabapentin (days 6 and 7).Physical examinationPain scores assessed independently by three individuals with a visual analog scale (VAS; 0 = no pain and 10 = worst possible pain) were 8.5 on day 6, and it increased to 9.5 on day 7. Non‐invasive blood pressure monitoring revealed severe hypertension.ManagementAs euthanasia was being considered for humane reasons, a decision was made to add an experimental new drug, trans‐4‐{4‐[3‐(4‐Trifluoromethoxy‐phenyl)‐ureido]‐cyclohexyloxy}‐benzoic acid (t‐TUCB), which is a soluble epoxide hydrolase (sEH) inhibitor, to the treatment protocol. Dose and frequency of administration were selected based on the drug potency against equine sEH to produce plasma concentrations within the range of 30 nmol L?1 and 2.5 μmol L?1. Pain scores decreased sharply and remarkably following t‐TUCB administration and blood pressure progressively decreased to physiologic normal values. Plasma concentrations of t‐TUCB, measured daily, were within the expected range, whereas phenylbutazone and gabapentin plasma levels were below the suggested efficacious concentrations.Follow upNo adverse effects were detected on clinical and laboratory examinations during and after t‐TUCB administration. No new episodes of laminitis have been noted up to the time of writing (120 days following treatment).ConclusionsInhibition of sEH with t‐TUCB was associated with a significant improvement in pain scores in one horse with laminitis whose pain was refractory to the standard of care therapy. No adverse effects were noticed. Future studies evaluating the analgesic and protective effects of these compounds in painful inflammatory diseases in animals are warranted.  相似文献

ObjectiveTo investigate the CD38/cADPR signaling pathway as possible underlying mechanism of the effects of medetomidine on insulin and glucose homeostasis.AnimalsThirty–two C57BL/6 mice of both sexes.MethodsWild–type (WT) and CD38–knockout (CD38?/?) mice received medetomidine (50 μg kg?1) or a similar volume of 0.9% NaCl (control) by intraperitoneal (IP) injection (each group n = 8). The mice were euthanized 45 minutes later with sodium pentobarbital IP and blood was sampled via cardiac puncture. Insulin and glucose concentrations were measured by radioimmunoassay and by the oxygen rate method, respectively. Data were analyzed with anova and Bonferroni post hoc (5% significance) and are shown as mean ± SD.ResultsPlasma insulin and glucose concentrations were similar between WT and CD38?/? mice under control conditions. As compared to controls, medetomidine administration produced a statistically significant decrease in plasma insulin concentrations in the WT mice whereas the decrease in the CD38?/? mice was not statistically significant. Correspondingly, medetomidine caused a significantly greater increase in plasma glucose concentrations in the WT than in the CD38?/? mice.ConclusionThe CD38/cADPR signaling pathway may be one underlying mechanism of the glucose and insulin effects of the alpha–2 adrenergic receptor agonist medetomidine and likely other drugs of its’ class.  相似文献

In the aquaculture industry, the physiological systems of fish can be chronically stressed by various biological, chemical and physical factors. Chronic stress leads to a decrease in the overall health and growth of the fish, making them more prone to diseases. Dietary iodine has been shown to reduce this stress response in chickens and increase disease resistance in dairy cattle, but the mechanisms by which iodine affects stress resistance and immunocompetency is not completely understood and has not been extensively studied in fish. This study investigated the effects of iodized feed as a nutritional supplement in relation to stress modulation in steelhead trout (Oncorhynchus mykiss). We investigated effects on primary, secondary and tertiary stress responses by measuring plasma levels of cortisol, glucose and thyroid hormones as well as haematocrit percentage and average growth rate in steelhead trout. Iodine‐supplemented fish, on average, had lower levels of plasma cortisol and glucose and lower packed cell volumes than fish fed with regular commercial feed (P<0.05). Iodine‐supplemented fish also showed higher levels of thyroid hormones and exhibited better growth over the period of the experiment (P<0.05). It was concluded that, used with other husbandry practices, use of iodine‐supplemented feed could lead to better host defence, growth and survival in fish raised in the aquaculture industry.  相似文献
