Articular cartilage (AC) covers the surface of bones in joints and functions as a cushion against mechanical loading. The tissue contains abundant extracellular matrix (ECM), which mainly consists of proteoglycans (PG) and collagen (COL) fibres. The property of AC is gradually changing by ageing with gravity loading. To know the property change of AC by initial gravity loading during short period after birth, we performed histological assays and proteomics assay on the AC of the femoral condyle in knee joints of perinatal rats. The water content (%) was significantly decreased in neonate AC compared with fetal AC. During the perinatal stages (E19 and P0), the localizations of glycosaminoglycan (GAG) and type I and II COLs were homogeneous. The density of chondrocytes was significantly decreased in the deeper layers comparing with the surface layer in neonate AC. In addition, we found a drastic change in the protein expression pattern on proteomic analysis. The expressions of ECM components were relatively increased in neonate AC compared with fetal AC. 相似文献

Cyclooxygenase-2 (COX-2), P-glycoprotein (P-gp) and multi-drug resistance-associated protein (MRP) are considered important tumor-associated proteins in humans and dogs. In the present study, we immunohistochemically evaluated the expression of these proteins in canine patients with transitional cell carcinoma (TCC). Of 52 cases, 30 (57.7%) were positive for COX-2, 40 (76.9%) for P-gp, and only 10 (19.2%) for MRP. In addition, 27 samples (27/52, 51.9%) were positive for two markers, while 3 (5.7%) and 5 (9.6%) cases were positive and negative, respectively, for all three markers. No significant correlations were seen for COX-2 and P-gp on Fisher's exact test and Mann-Whitney's test, but a significance was seen on Spearman's rank correlation analysis using the IHC scoring system (P=0.043). These results suggest that P-gp expression is induced by overexpression of COX-2 in canine patients with TCC. 相似文献

Vibrio vulnificus is a Gram-negative pathogenic bacterium that causes serious infections in humans and requires iron for growth. A clinical isolate, V. vulnificus M2799, secretes a catecholate siderophore, vulnibactin, that captures ferric ions from the environment. In the ferric-utilization system in V. vulnificus M2799, an isochorismate synthase (ICS) and an outer membrane receptor, VuuA, are required under low-iron conditions, but alternative proteins FatB and VuuB can function as a periplasmic-binding protein and a ferric-chelate reductase, respectively. The vulnibactin-export system is assembled from TolCV1 and several RND proteins, including VV1_1681. In heme acquisition, HupA and HvtA serve as specific outer membrane receptors and HupB is a sole periplasmic-binding protein, unlike FatB in the ferric-vulnibactin utilization system. We propose that ferric-siderophore periplasmic-binding proteins and ferric-chelate reductases are potential targets for drug discovery in infectious diseases. 相似文献

The short wavelength sensitive (SWS) opsin gene is expected to contain informative sites for understanding the speciation
of the family Delphinidae, because it is not functional in cetaceans. We determined partial SWS gene sequences from 15 delphinid
species of 12 genera and from harbor porpoise for comparison. We found a 39-bp insertion that was shared by six species (the
insertion group: Delphinus delphis, Delphinus capensis, Stenella longirostris, Stenella coeruleoalba, Lagenodelphis hosei, and Sousa chinensis) and common base substitutions shared by eight species (Stenella frontalis, Tursiops truncatus, and six species of the insertion group). As these insertions and substitutions are not found in the other seven delphinids
or in the cloven-hoofed mammals (which are close to cetaceans), it is suggested that these eight species are more closely
related to each other than to the other species. This hypothesis is supported by phylogenetic analyses. The eight species
with the substitutions formed a clade containing two sister clades, one consisting of the insertion group and the other consisting
of the two other species, in both neighbor-joining and Bayes analyses. Phylogenetic analyses also showed that Lissodelphis borealis and Lagenorhynchus obliquidens are closely related and that their common ancestor diverged from the others at an early stage of delphinid evolution. 相似文献

Lysophosphatidic acid (LPA) acts as a simple phospholipid that interacts with G
protein-coupled transmembrane LPA receptors. Recently, it has been reported that each LPA
receptor plays different biological roles in acquisition of the malignant property of
tumor cells. In this study, to assess the involvement of LPA receptor-3 (LPA3)
in cell survival after treatment with anticancer drugs, we generated
Lpar3-expressing FM3A-a3A9 cells from mouse mammary tumor FM3A cells
and examined the cell survival rate after treatment with anticancer drugs compared with
Lpar3-unexpressing cells. Cells were treated with 0.005 to 10 μM of
cisplatin (CDDP) or doxorubicin (DOX) for 3 days. For the CDDP and DOX treatments, the
cell survival rate of FM3A-a3A9 cells was significantly higher than that of
Lpar3-unexpressing cells. The expression level of the
Mdr1a gene in FM3A-a3A9 cells was higher than that of
Lpar3-unexpressing cells, whereas no significant difference in
multidrug resistance 1b (Mdr1b) and glutathione S-transferase
mu1 (Gstm1) expressions was found. These results suggest that LPA3
may enhance the cell survival rate after treatment with anticancer drugs in mouse mammary

Escherichia coli(E. coli) isolated from the uterus of a Thoroughbred mare with bacterial endometritis was used to evaluate the effect of progesterone (P(4)) on the immune response of mares. Peripheral blood mononuclear cells (PBMCs) were collected from 10 nonpregnant clinically healthy adult mares (range, 4-12 years) during diestrus, four Thoroughbreds and six Hokkaido native horses. Cell proliferation and expression of cytokine mRNA, including interferon (IFN)-γ, tumor necrosis factor (TNF)-α and interleukin (IL)-10, of PBMCs stimulated with E. coli and P(4) were examined in vitro. P(4) was shown to have significantly inhibited E. coli induced proliferation and expression of IFN-γ in PBMCs. These results indicate that P(4) inhibits the immune response to E. coli in mares. 相似文献

Abstract A water culture experiment on taro was conducted with two cultivars and two kinds of culture solution (I and II) with gradient omission treatment of calcium using newly devised culture equipment, in order to investigate their influence on so-called “Metsubure” symptoms. Calcium omission treatment in both culture solutions resulted in a considerable increase of “Metsubure” corms. The results confirmed Miyaji's data and conclusions in respect to the main cause of “Metsubure” symptoms. Solution II proved more suitable for water culture of taro than solution I as regards the ratio of its calcium to potassium content, the moderate renewal intervals and the slower lowering of pH. However, fürther improvements may be necessary to maintain an adequate range of pH for at least 10 days. The possibility that there might be factor(s) responsible for “Metsubure” corm formation other than calcium deficiency is also discussed. 相似文献
