Artificial breeding of mithun poses several challenges including lack of standard protocol for cryopreservation of spermatozoa. This is further complicated by harmful effects of hen's egg yolk (EY) as additive in extender. Purified low‐density lipoproteins (LDL) extracted from EY have been shown as beneficial over EY extender for long‐term semen storage in several species. This investigation explored use of LDL versus EY on semen quality and oxidative stress following freezing–thawing of spermatozoa. A total of 25 of 50 ejaculates based on biophysical parameters were selected for the experiment. After diluting with the Tris‐citrate‐glycerol (TCG) extender, each sample was split into three equal aliquots: Group I, control, EY; Group II and Group III contained 8% and 10% purified LDL, respectively. Frozen–thawed samples were evaluated for motility parameters (progressive, and in the bovine cervical mucus penetration test [BCMPT]), viability, sperm and nuclear abnormality, acrosome integrity, and enzymatic (leakage of intracellular contents) and biochemical (oxidative stress) profiles and in vitro fertility (IVF) assay. Study revealed a significant (< .05) improvement in viability, sperm and nuclear abnormality, acrosome integrity, motility (progressive and in cervical mucus), cholesterol content, and reduction in the leakage of intracellular enzymes in Group II. Moreover, intactness of acrosome and biochemical membranes was protected significantly (p < .05) in addition to significant (p < .05) improvement in binding per cent and binding index in IVF assay in extender containing 8% LDL. These results demonstrate that although cryopreservation of mithun's spermatozoa in EY was comparable with other species, addition of 8% LDL holds a clear advantage over EY or 10% LDL.  相似文献

Coffee leaf rust (CLR) caused by the fungus Hemileia vastarix is a devastating disease in almost all coffee producing countries and remote sensing approaches have the potential to monitor the disease. This study evaluated the potential of Sentinel-2 band settings for discriminating CLR infection levels at leaf levels. Field spectra were resampled to the band settings of the Sentinel-2, and evaluated using the random forest (RF) and partial least squares discriminant analysis (PLS-DA) algorithms with and without variable optimization. Using all variables, Sentinel-2 Multispectral Imager (MSI)-derived vegetation indices achieved higher overall accuracy of 76.2% when compared to 69.8% obtained using raw spectral bands. Using the RF out-of-bag (OOB) scores, 4 spectral bands and 7 vegetation indices were identified as important variables in CLR discrimination. Using the PLS-DA Variable Importance in Projection (VIP) score, 3 Sentinel-2 spectral bands (B4, B6 and B5) and 5 vegetation indices were found to be important variables. Use of the identified variables improved the CLR discrimination accuracies to 79.4 and 82.5% for spectral bands and indices respectively when discriminated with the RF. Discrimination accuracy slightly increased through variable optimization for PLS-DA using spectral bands (63.5%) and vegetation indices (71.4%). Overall, this study showed the potential of the Sentinel 2 MSI band settings for CLR discrimination as part of crop condition assessment. Nevertheless further studies are required under field conditions.  相似文献

French bean (Phaseolus vulgaris L.) cv. ‘Anupama’ was grown in refined sand at variable levels of cobalt (Co), i.e., 0.0001 (control), 0.05, 0.10, 0.20, 0.40, and 0.50 mM, supplied as cobalt sulfate. The symptoms of excess Co (0.50 mM) appeared at day 35. In addition to growth depression, the young leaves developed chlorosis from the apex leading toward the base; the chlorosis intensified, changed to necrosis, and the infected leaves dried and withered. Under excess Co (> 0.0001 mM), the flowers produced were fewer in number, smaller, and many failed to mature, leading to lower seed yield. Excess Co (> 0.0001 mM) decreased the biomass, seed yield (number and weight), concentration of chlorophyll, Hill reaction activity, and activity of catalase, and deteriorated the quality of produce (sugars, starch, and protein nitrogen (N)), but it increased the concentration of phenols and activity of certain enzymes, i.e., peroxidase, ribonuclease, and acid phosphatase. The concentration of Co in various parts of French bean increased with an increase in Co supply and that of iron (Fe) decreased concomitantly. The values of threshold of toxicity and toxicity of Co were 26 and 72 μg g?1 in young leaves of French bean, respectively.  相似文献

Cattle control body temperature in a narrow range over varying climatic conditions. Endogenous body heat is generated by metabolism, digestion and activity. Radiation is the primary external source of heat transfer into the body of cattle. Cattle homeothermy uses behavioural and physiological controls to manage radiation, convection, conduction, and evaporative exchange of heat between the body and the environment, noting that evaporative mechanisms almost exclusively transfer body heat to the environment. Cattle control radiation by shade seeking (hot) and shelter (cold) and by huddling or standing further apart, noting there are intrinsic breed and age differences in radiative transfer potential. The temperature gradient between the skin and the external environment and wind speed (convection) determines heat transfer by these means. Cattle control these mechanisms by managing blood flow to the periphery (physiology), by shelter-seeking and standing/lying activity in the short term (behaviourally) and by modifying their coats and adjusting their metabolic rates in the longer term (acclimatisation). Evaporative heat loss in cattle is primarily from sweating, with some respiratory contribution, and is the primary mechanism for dissipating excess heat when environmental temperatures exceed skin temperature (~36°C). Cattle tend to be better adapted to cooler rather than hotter external conditions, with Bos indicus breeds more adapted to hotter conditions than Bos taurus. Management can minimise the risk of thermal stress by ensuring appropriate breeds of suitably acclimatised cattle, at appropriate stocking densities, fed appropriate diets (and water), and with access to suitable shelter and ventilation are better suited to their expected farm environment.  相似文献

There are few reports of coagulation times in marsupial species. Blood samples collected from 14 Bennett's wallabies (Macropus rufogriseus) under anaesthesia during routine health assessments were analysed for prothrombin time (PT) and activated partial thromboplastin time (aPTT) using a point‐of‐care analyser (POC) (Abaxis VSPro®). The wallabies had an aPTT mean of 78.09 s and median of 78.1 s. The PT for all wallabies was greater than 35 s, exceeding the longest time measured on the POC. Although PT was significantly longer, aPTT was similar to the manufacturer's domestic canine reference range.  相似文献

An 8-year-old terrier cross and a 10-year-old German Shorthaired Pointer presented to the University Veterinary Centre, Sydney, for investigation of long-standing tenesmus and dyschezia. Both patients had an annular adenocarcinoma at the colorectal junction. Exploratory laparotomy was performed and the affected large intestinal segment was removed by resection and anastomosis. In both dogs, the caudal mesenteric artery was intimately associated with the mass, necessitating its ligation and transection. Postoperatively, there was no evidence of anastomosis breakdown in either case and both animals recovered well from surgery. The dogs were euthanased 8 and 10 months, respectively, after surgery because of clinical signs relating to metastatic disease.  相似文献

Connective tissue growth factor (CTGF) has been shown to mediate many actions of transforming growth factor-beta (TGF-beta) in the fibrotic response in several diseases. We compared expression of CTGF, TGF-beta, platelet-derived growth factor (PDGF), TNF-alpha, and interleukin-1 (IL-1) by in situ hybridization in Sprague-Dawley rats euthanized at 0, 2, 4, and 8 weeks after 5/6 nephrectomy using the rat remnant kidney model of renal failure. Collagen was evaluated by trichrome stains, immunohistochemistry, and electron microscopy. We compared expression patterns to cells undergoing metaplasia. Tubular epithelial regeneration and transdifferentiation to myofibroblasts were assessed morphologically and by proliferating cell nuclear antigen, smooth muscle actin, desmin, and vimentin immunohistochemistry. CTGF expression was minimal in controls, mild at 2 weeks and marked by 4 to 8 weeks in interstitial fibroblasts, coinciding with damage, regeneration, and fibrosis. TGF-beta expression was increased in many cell types at 2 weeks, increased further by 4 weeks, then remained constant. PDGF-B messenger RNA was found in many stromal cells at 2-4 weeks, but expression decreased at 8 weeks. No significant IL-1 or TNF-alpha staining was detected. We conclude that CTGF and interacting factors are associated with development or progression of chronic interstitial fibrosis. Proximity of CTGF, TGF-beta, and PDGF mRNA expression to regenerative epithelial cells and those transdifferentiating to myofibroblasts suggests that growth factors may modulate renal tubular epithelial differentiation.  相似文献
