Purification and characterization of hepatic triacylglycerol lipase isolated from rainbow trout,Oncorhynchus mykiss

Trialcylglycerol (TG) lipase was isolated and partially purified from rainbow trout liver. Triacylglycerol lipase activity was assayed by measuring¹⁴C-oleic acid release from¹⁴C-triolein.¹⁴C-oleic acid release was linear for up to two hours. Optimal activity occurred at pH 7.0 and 15°C. Most of the lipase activity was recovered in the cytosolic fraction. A 27,000-fold purification was achieved after Sepharose (Bio-gel A 0.5 M, 200–400 mesh) chromatography of a resuspended 20% ammonium sulfate fraction. The molecular weight of the trout hepatic lipase as determined by size-exclusion chromatography and by SDS-polyacrylamide gel electrophoresis was 40–43 kD. Lipase-mediated hydrolysis of TG resulted in the production of diacylglycerols, monoacylglycerols, and fatty acids. Kinetic analysis indicated that V_max=0.016 nmol/h/mg protein and that K_m=0.28 mM triolein. Lipolytic activity was enhanced in the presence of cAMP/ATP-Mg²⁺. These results suggest that the liver of trout possesses a neutral TG lipase that is responsible for mobilizing stored TG and is catalytically activated by phosphorylation.A part of this work was presented at the Annual Meeting of the American Society of Zoologists, December 26–30, 1990, San Antonio, TX.

猪血免疫球蛋白的热稳定性及动力学研究

为了使猪血免疫球蛋白在各类食品中得到较好的应用 ,对猪血免疫球蛋白 (PIgG)的热稳定性及热变性动力学进行了分析研究。结果表明 ,PIgG在 6 0℃以下很稳定 ,6 0～ 6 5℃比较稳定 ,大于 70℃变性明显加快 ,超高温杀菌 (12 0℃ ,4s)条件下 ,PIgG几乎失活 ;其热动力学反应为 1 2级 ;在 6 0 ,6 5 ,70 ,75 ,80 ,85和 90℃ ,PIgG热变性的D值分别为 33 333× 10 3 ,14 2 86× 10 3 ,3 333× 10 3 ,0 4 76× 10 3 ,0 2 2 2× 10 3 ,0 16 9× 10 3 和 0 0 76× 10 3 s ,在此温度范围内的Z值为 10 81℃ ,表观活化能为 2 2 7 31kJ·mol-1。     

三种环境因子对大白菜软腐欧氏杆菌根毛吸附的影响

 温度、pH和离子条件是影响病原细菌对寄主吸附的基本因子,文章测定了它们对大白菜软腐欧氏杆菌(Erwinia carotovora pv.carotoyora)在寄主根毛上吸附的影响。病菌在5个给定温度下对寄主根毛吸附量的变化,呈现为由吸附起始温度(18℃)、温度稳恒渐近范围(18-22℃)和温度饱和点(22℃)组成的吸附动力曲线。病菌在12个pH梯度下的运动状态、生活力及对寄主根毛吸附量的变化表明,病菌吸附以pH5.8-7.2为适宜范围,pH4.6和8.2为低限和高限;偏酸(pH5.0以下)和偏碱(pH7.2以上)可通过影响病菌的生活力和运动性而影响吸附。5×10^-6M的Ca++可使病菌吸附量提高2.5-4.2倍,同浓度的Mg++可使吸附量提高2.7倍。文章讨论了温度和pH条件影响吸附的动力学,以及吸附调节的可能机制。     

甲状腺球蛋白免疫组化对甲状腺肿瘤的诊断价值

用免疫组化A B C法对60例甲状腺病理标本进行了甲状腺球蛋白检查,结果表明源于甲状腺滤泡上皮细胞的肿瘤,均呈阳性或弱阳性反应。大部分甲状腺癌呈弱阳性反应。作者认为甲状腺球蛋白可作为鉴别甲状腺滤泡上皮细胞源性与非滤泡上皮细胞源性的良好标记。     

丰宁“中国沙棘”资源及种群表型结构的研究

本文用样地调查和统计分析的方法研究了丰宁“中国沙棘”主要经济性状的变异,探讨了性状相关和各主要性状的组配情况,分析了主要经济性状变异的频数分布及种群特征和优种分布范围。认为丰宁的“中国沙棘”是以主干型、桔黄扁圆果、褐色种子植株为主体的多态型种群系统,是品质较好的种质资源,是沙棘树种改良的珍贵材料。     

Foot-and-mouth disease eradication efforts in the Republic of Korea

On March 20, 2000, a suspected vesicular disease in cattle was reported to the National Veterinary Research and Quarantine Service (NVRQS) of the Republic of Korea. This represented the index case of a foot-and-mouth disease (FMD) outbreak, which spread through several provinces. The Republic of Korea had been free of FMD for 66 years prior to the reintroduction of the virus and had recently suspended imports of pork and pork products from neighboring Japan owing to a reported FMD outbreak in that country. The Korean outbreak was ultimately controlled through the combination of preemptive slaughter, animal movement restrictions, and a strategy of ring vaccination. The purpose of this paper is to review the current FMD situation in Korea in the aftermath of its 2000 epizootic and how it may affect future efforts to eradicate or reduce risk of reintroduction of the disease into Korea.     

通用开放式绘图仪接口软件设计与实现

秸秆饲料开发利用现状及前景

农作物秸秆在自然条件下是一种劣质饲料，主要有玉米秸、麦秸、稻草、豆秸、红薯藤、油菜秸、花生秧等。其特点是：粗蛋白含量低，而且品质不佳；粗纤维含量高，其中含有不易被家畜消化的木质素；无氮浸出物含量高，主要成分是半纤维素和聚糖醛酸；有些秸秆质地粗硬，适口性差，采食率极低。但是，对秸秆采用青贮、氨化、碱化、热压力加工、高强度冷压、微生物等方法进行处理后，即能分解木质素，降低粗纤维含量，提高营养价值，提高利用率、消化率、采食速度，增强适口性，增加采食量，达到减少资源浪费的目的。1 秸秆饲料开发利用现状目前…     

Immunohistochemical detection of scrapie prion proteins in clinically normal sheep in Pennsylvania.

Following diagnosis of scrapie in a clinically suspect Suffolk sheep, 7 clinically normal flockmates were purchased by the Pennsylvania Department of Agriculture to determine their scrapie status using an immunohistochemical procedure. Two of the 7 euthanized healthy sheep had positive immunohistochemical staining of the prion protein of scrapie (PrP-Sc) in their brains, nictitating membranes, and tonsils. The PrP-Sc was localized in the areas of the brain where, histopathologically, there was neurodegeneration and astrocytosis. The PrP-Sc occurred within germinal centers of the affected nictitating membranes and tonsils and was located in the cytoplasm of the dendrite-like cells, lymphoid cells, and macrophages. These results confirm that immunohistochemical examination of the nictitating membrane can be used as a screen for the presence of scrapie infection in clinically normal sheep at a capable veterinary diagnostic laboratory. In sheep with a PrP-Sc-positive nictitating membrane, the diagnosis of scrapie should be confirmed by histopathology and immunohistochemical examination of the brain following necropsy. Following full validation, immunohistochemistry assays for detection of PrP-Sc in nictitating membrane lymphoid tissues can improve the effectiveness of the scrapie control and eradication program by allowing diagnosis of the disease in sheep before the appearance of clinical signs.
