The objective of the present study was to evaluate the protective effect of humoral and cellular immunities on turkeys infected with turkey coronavirus (TCV). Two trials were conducted with two separate hatches of turkey poults. Turkey's were experimentally immunosuppressed with cyclosporin A (CsA) or cyclophosphamide (CY) and infected with TCV. Prior to infection, treatment with CsA selectively suppressed T cell activity as revealed by 2-3 fold decreased (p < 0.1) lymphocyte proliferation responses to a T cell mitogen, concanavalin A (Con A). Treatment with CY mainly induced B cell deficiency as indicated by significant reductions (p < 0.05) in antibody responses to sheep erythrocytes 7 days after injection. Body weight gain of turkeys treated with CY was significantly lower (p < 0.05) than that of untreated turkeys at 9 days post-infection (PI). Turkeys treated with CY had 1-2 fold higher immunofluorescent antibody assay (IFA) scores for TCV antigens (p < 0.05) in the intestine than untreated turkeys at 9 or 14 days PI. Fungal mats of Hysterangium crassum (Tul. and Tul.) Fischer occupied a mean of 9.6% of the upper 10 cm of soil developed under a 40–65 yr old stand of Douglas-fir (Pseudotsuga menziesii (Mirb.) Franco) in Oregon. This hypogeous basidiomycete exudes large amounts of oxalic acid, some of which precipitates with Ca in microscopic crystals of calcium oxalate, resulting in a mean CaC2O4 content of 82g m?2 for the entire soil. Soil oxalate concentration was significantly greater within fungal mats (P < 0.01) and soil pH was significantly lower (P < 0.01) than in soil adjacent to mats. The quantity of Ca present as CaC2O4 is 0.5 the amount of exchangeable Ca in the soil and exceeds the mass of Ca lost annually in runoff. Scanning electron micrographs show intense chemical weathering, attributable to oxalate attack, in the immediate vicinity of hyphae. X-ray diffraction patterns of clay indicate bulk weathering is more intense within the fungal mats than in adjacent uncolonized soil. 相似文献

The pH buffer capacity of a soil (pHBC) determines the amount of lime required to raise the pH of the soil layer from its initial acid condition to an optimal pH for plant growth and the time available under current net acid addition rate (NAAR) until the soil layer acidifies to a critical pH leading to likely production losses. Accurate values of pHBC can also be used to calculate NAAR from observed changes in soil pH. In spite of its importance, there is a critical shortage of pHBC data, likely due to the long period of time needed for its direct measurement. This work aimed to develop quick, simple and reliable methods of pHBC measurement and to test these methods against a slow (7‐day) titration used as benchmark. The method developed here calculates pHBC directly from the pH buffer capacity of the buffer solution and the increase in soil pH and corresponding decrease in pH of the buffer solution following mixing and equilibration. The pHBC values calculated using Adams and Evans or modified Woodruff buffers were in accord with those measured by slow titration. Buffer methods are easily deployed in commercial and research laboratories as well as in the field. The advantage of using buffer solutions to calculate pHBC instead of lime requirement is the broad application of this soil property. The pHBC of a soil is an intrinsic property that would not be expected to need remeasurement over periods of less than decades. Recurring lime requirement can be calculated from the soil's pHBC, initial and target pH values. A large proportion of the variability in pHBC was explained by the soil organic carbon content. This relationship between pHBC and soil organic carbon content allowed us to develop local pedotransfer functions to estimate pHBC for different regions of Australia. 相似文献

Conidia of Cochliobolus sativus and five other pigmented fungi lysed when incubated in natural soil. Lysis followed perforation of the spore wall by holes of varying dia. Three possible causes of perforation were investigated, namely autolysis, mechanical puncture by soil animals and enzymatic erosion by soil micro-organisms. Results indicated that soil micro-organisms were the likely causal agents although no micro-organism able to perforate conidia has yet been isolated. Colonization of conidia by the soil microflora was studied by electron microscopy. On the basis of these direct observations, possible perforation mechanisms are suggested. Reports of perforation of fungal, plant and bacterial cell walls are briefly summarized and the perforation phenomenon discussed in relation to the biodegradation of pigmented fungal propagules in soils. 相似文献

Age-related changes in mitogen-induced lymphocyte proliferation and polymorphonuclear neutrophil function in the piglet 总被引:5,自引:0,他引:5
Changes in concentration of plasma cortisol, mitogen-induced lymphocyte proliferation and the phagocytic and killing abilities of polymorphonuclear neutrophils (PMN) were assessed in 24 crossbred piglets at .5, 1, 3 and 6 wk of age. Concentrations of blood cortisol were high at birth but decreased (P less than .001) thereafter. Spontaneous lymphocyte proliferation was approximately 10-fold higher in the newborn than in pigs 6 wk old. The methods by which data on lymphocyte proliferation were expressed greatly influenced the results of the age-related changes in lymphocyte response. When lymphocyte proliferation was expressed as a stimulation index, there was an increase in mitogen-induced lymphocyte proliferation with advancing age. However, when lymphocyte proliferation was expressed as total counts per minute, mitogen-induced lymphocyte proliferation decreased with increasing age. In general, no significant age-related changes were observed in PMN function except for a transient decrease in PMN phagocytic ability at wk 3. Approximately 10 of 100 young heifers that had recently delivered their first calf--members of a large Colorado dairy herd--had a syndrome of swollen teats and distal portions of the hind limbs, prefemoral lymphadenopathy, transient fever, rough coat, decreased milk production, and subsequent weight loss and reproductive inefficiency. Acute clinical signs of disease were associated with large numbers of Eperythrozoon wenyonii seen on blood smears, and resolution of signs correlated with reduction or disappearance of the parasite. Other known causes of peripheral edema could not be documented. The parasite was transmitted to 4 of 7 nonlactating dairy cows destined to be culled and a splenectomized calf via IV inoculation of blood from parasitemic heifers, but clinical signs of infection were not induced. 相似文献

Ninety dogs naturally infected with Dirofilaria immitis and 103 noninfected dogs, as determined by necropsy, were used to compare the sensitivity and specificity of the cuticular and somatic reactions of the indirect fluorescent antibody test (IFA-C and IFA-S, respectively) and 2 enzyme-linked immunosorbent assays (ELISA). In microfilaremic and amicrofilaremic heartworm-infected dogs, negative results were common for all serotests. In dogs without adult heartworms at necropsy, 32% to 49% were positive, using 1 ELISA, 27% to 29% were positive with the other ELISA, 15% to 36% were positive with the IFA-S, and 0% to 1% were positive using the IFA-C, depending on the classification of borderline reactions. The prevalence of false positive serotests was probably not due to the detection of precardial stages of D immitis in dogs obtained from areas of low endemicity. Until the causes of the false-positive tests are resolved, the use of currently available serotests for routine diagnostic screening or as criteria for instituting treatment is not recommended. 相似文献
