The shortfin silverside Chirostoma humboldtianum has been considered for culture in Mexico, but success has been limited by a poor knowledge of its early development. First synthesis of the early development of the shortfin silverside is presented to determine conditions suitable for rearing. Brooder maturation was induced through photothermal cycles. C. humboldtianum ova were fertilized in vitro. The eggs were incubated in reconstituted water (160-180 mg/L CaCO3) at 18 °C and 5 gm of NaCl per litre. During the hatching day, 300 shortfin silversides were stocked and followed up until metamorphosis in order to establish the timing of exogenous feeding, changes in food type, growth and development during critical periods for survival, according to the theory of saltatory ontogeny. Free embryos hatched 12 days after fertilization at 18 °C. First critical point for survival is the beginning of exogenous feeding. Free embryos started mixed feeding on day four of post-hatching (dph), point of no-return was presented towards the end of mixed feeding on 6 dph, larval period began at six (dph) when the anus is opened, and metamorphosis to juvenile was presented at 65 dph with a SL of 19.34 ± 2.28 mm, when scales and fins were well developed. Differences in growth between periods were detected: free embryos growth slower than larvae but mouth size depicted a larger growth rate in the former. Cephalic length and mouth size were negatively related to standard length in embryos and larvae. Mouth size was positively related to cephalic length in free embryos but negative in larvae. Results suggest that during the free embryo phase, growth priorities are directed to the development of apparatuses and systems; whereas, during the larval period, energy is directed to growth in length, mouth size and development of fins, which allows them to increase their swimming velocity, grants them a greater capacity to obtain exogenous food and, in consequence, increases fitness for survival.  相似文献

  1. Marine protected areas (MPAs) are becoming a widely used tool for the conservation of biodiversity and for fishery management; however, most of these areas are designed without prior knowledge of the basic ecological aspects of the species that they are trying to protect.
  2. This study investigated the movement of two top predators: the Galapagos shark, Carcharhinus galapagensis, and the yellowtail amberjack, Seriola lalandi, in and around the Motu Motiro Hiva Marine Park (MMHMP) using MiniPAT satellite tags to determine the effectiveness of this MPA for the protection of these species.
  3. The Galapagos sharks (n = 4) spent most of their tag deployment periods inside the MMHMP. However, high intraspecific variability was observed in their movement dynamics. Daily individual maximum movements ranged from 17 to 58 km and the maximum distance from Salas y Gómez Island, the only emergent island within the MMHMP, ranged from 31 to 139 km.
  4. The maximum linear distance travelled for a female juvenile Galapagos shark (152 cm total length) was 236 km, which is greater than the maximum distance previously documented for juveniles of this species (<50 km).
  5. For the yellowtail amberjack (n = 1), 91% of the satellite geolocations were within the MMHMP, with a maximum daily distance travelled of 6 km. The maximum distance travelled between points was 111 km and the maximum distance from Salas y Gómez Island was 62 km.
  6. All archival tagged fish spent most of their time at depths of <50 m and never left the epipelagic zone. Daytime versus night-time differences were pronounced in all individuals but showed high interindividual variability.
  7. This study provides a baseline on the movement of these two top predators in the MMHMP and provides valuable insights for the creation of MPAs in the region and elsewhere.

The feeding habits of soil mesofauna have been a mystery for decades, and depending on the methods used, different degrees of feeding specialisation have been observed. A new way to study the almost unknown feeding habits of soil mesofauna e.g. oribatid mites is to use PCR-based techniques. When applying PCR on these small organisms, the low amount of ingested DNA can cause problems. Even more important is to certify that the amplified DNA does not originate from body-surface contamination. The aim of this study was to analyse if washing of the body surface combined with PCR can be a successful approach when identifying the food ingested by fungivorous mites. The method was developed in a laboratory system where we used the oribatid mite Archegozetes longisetosus as a model organism due to its relatively short life cycle and ease of laboratory culturing. The results demonstrated that surface contamination is a serious problem. Both washing and dissection was needed to remove surface contamination on such small organisms. To get a reliable result the samples also had to contain at least five pooled guts, but preferably ten. This is the first step towards a successful use of PCR-based methods to study natural feeding habits of species most likely contaminated on the body surface. When modified for field conditions, the results obtained by this method have a high potential to answer many questions about the animals feeding habits, and their functional role in the soil.  相似文献

This study was conducted to evaluate the effect of citric acid (CA) and amino acid chelated trace elements (AA) on the growth, phosphorus (P) utilization efficiency and loading in rainbow trout fed a diet formulated with a low‐fish meal (FM) concentration. A low‐FM basal diet with or without monocalcium phosphate supplementation served as positive and negative control correspondingly. Dietary treatments consisted of the basal diet supplemented either with 1% CA, AA (equivalent to 40 Zn, 20 Mn, and 4 Cu mg/Kg of diet instead of inorganic trace element mix) or a combination of both. Duplicate groups of 35 fish (13.2 g) were fed until apparent satiation during 12 wk. Group of fish fed diets supplemented with 1% CA or AA had a final growth and feed utilization similar to that observed in fish fed the positive control diet (P > 0.05). P absorption was increased with the addition of CA or AA. P retention efficiency rates for these diets were higher in comparison with the rest of the treatments consequently showing the lowest P loading values (P < 0.05). The results indicate that addition of 1% CA or AA, to a low‐FM diet without inorganic P supplementation enhanced fish growth, P retention, and decreased P load to the environment.  相似文献

The Δ6 and Δ5 desaturases and elongases show only very limited activity in marine fish, and little is known of the possibility of enhancing Δ6 desaturase gene expression in these fish. The use of plant oils in marine fish diets is limited by their lack of n−3 highly unsaturated fatty acids (HUFA) despite an abundant content of the 18C fatty acid precursor linoleic and α-linolenic acids. The objective of the present study was to determine the ability of larval gilthead seabream to utilize vegetable oils and assess the nutritional regulation of Δ6 desaturase gene expression. Seventeen-day-old gilthead seabream larvae were fed during a 17-day period with one of four different microdiets formulated with either sardine fish oil (FO), soybean, rapeseed or linseed oils, respectively, or a fifth diet containing defatted squid meal and linseed oil. Good larval survival and growth, both in terms of total length and body weight, were obtained by feeding the larvae either rapeseed, soybean or linseed oils. The presence of vegetable oils in the diet increased the levels of 20:2n−9 and 20:2n−6, 18:2n−9, 18:3n−6, 20:3n−6 and 20:4n−6, in larvae fed rapeseed and soybean oils in comparison to those fed FO. In addition, a sixfold increase in the relative expression of Δ6 desaturase-like gene was found in larvae fed rapeseed and soybean oils, denoting the nutritional regulation of desaturase activity through its gene expression in this fish species. However, feeding linseed oil did not increase the expression of the Δ6 desaturase gene to such a high extent.  相似文献

Porcine rubulavirus (PoRV), also known as blue eye disease (BED) of swine, causes respiratory and reproductive problems in pigs at several developmental stages. To study the effect of PoRV infection on semen production, five boars were infected with 1 x 10(6) TCID(50)/ml of PoRV strain PAC-3 and evaluated for 59 days post inoculation (DPI). Infected boars developed reproductive tract pathology that included swelling of the testes and epididymides. Analysis of the semen showed that the infection had little effect on semen production in four animals, but semen from one boar showed severe alterations in sperm concentration, motility, and morphology. When motility was analyzed in BTS-diluted semen after 24, 48, or 72 h, alterations were detected in all boars. Furthermore, viral antigen was detected in semen, the seminal plasma fraction, or sperm fraction from all boars. These results showed that PoRV is excreted via semen and, therefore, artificial insemination is a potential route of dissemination.  相似文献

Three types of ceramic mini-boluses, B1 [13.8 g; 10.5 x 51.0 mm (o.d. x length)], B2 (16.2 g; 12.2 x 42.2 mm), and B3 (20.1 g; 11.2 x 56.4 mm), were used to electronically identify as soon as possible after birth a total of 545 lambs of 3 breeds: Ripollesa (meat breed, n = 274), Manchega (dairy breed, n = 129), and Lacaune (dairy breed, n = 142). Boluses were administered by a trained operator using a balling gun or directly by hand. Lambs were also identified with 2 types of plastic ear tags in the left (temporary, 1.5 g) and the right (permanent, 4.1 g) ears. Lamb and identification device performances were checked during suckling (to wk 5 or 7) and fattening. At 24 kg of BW, lambs were slaughtered (n = 385) or kept for breeding (n = 144). No differences in performance were observed between the 2 dairy breeds, and their data were pooled. Minimum BW for bolus administration was lower in dairy than in meat lambs (P < 0.001). Across breeds, B1 and B3 did not differ in lamb age (27 d) and weight (9.2 kg) at administration, but B2 required older and heavier lambs (33 d and 11.1 kg; P < 0.01). Boluses did not affect lamb performance, but final readability at slaughter differed between B1 and B2 (97.7 and 95.2%, respectively) and B3 (100%), and between ear tags (temporary, 98.1; permanent, 100%). Bolus recovery was 100% in all cases, but the proportion of boluses found in the reticulum varied among bolus type (83.3 to 93.8%; P < 0.05). Three B1 (2%) were recovered from the abomasum. In a second experiment, effects of the intermediate mini-bolus (B2) on diet digestibility were evaluated. Digestibility of control and bolus-administered, Manchega ram lambs (14.9 kg of BW; n = 8) fed ad libitum with 2 pelleted concentrates and barley straw was assessed in digestibility crates. Feed intake and nutrient digestibility were measured in four 21-d periods, during which lambs received the 2 diets consecutively. No differences in intake, growth performance, or nutrient digestibility were observed between control and bolused lambs. In conclusion, the B3 mini-bolus proved to be an efficient device for identification of lambs before weaning (recommended age, >4 wk; recommended BW, >10 kg), allowing a reliable traceability of dairy and meat lambs until slaughter.  相似文献

Plants collected in thirteen wild populations of Capsicum annuum from Northwest Mexico were tested for resistance to the pepper huasteco begomovirus (formerly subgroup III) (PHV) that is transmitted by the white fly Bemisia tabaci Genadius. Plants were inoculated using both grafting and biolistic methods. Presence of viral DNA was detected by dot blot hybridization and densitometry. Populations varied in their resistance to PHV. Plants of only two of the populations either did not develop disease symptoms or showed very light symptoms after inoculation. In some cases, symptoms appeared several days after inoculation. In plants of these populations viral DNA was detected by dot-blot hybridization assays but they appear to be a good source of resistance (symptomless) for use in breeding programmes. Genetic progress through selection is directly related to the amount of variability present in the population and the quality of genes contributed by the parents. Genetic relationships between lines were studied using DNA marker-based estimates of genetic similarity. A statistical methodology using the width of a confidence interval was developed to determine the number of probes to be surveyed and the precision in the estimation of genetic distance between pairs of cultivars. Precision was affected by type of genetic distance used, the number of cultivars, and amount of genetic diversity present in the studied group. The width of a (1-α)% confidence interval decreased as the number of RFLP fragments increased. Oat and wheat diversity studies were used to illustrate the methodology.
