Objective   To determine the efficacy of triclabendazole (TCBZ) against 28-day-old, early immature liver fluke in cattle and its pharmacokinetics following administration by the oral or topical (pour-on) route.
Procedures   Cattle (n = 18) were infected with 500 TCBZ-susceptible liver fluke metacercariae and randomly allocated to three groups. At 28 days after infection, the groups were: (1) untreated controls; (2) treated with oral TCBZ at 12 mg/kg in combination with oxfendazole and selenium (TOS); (3) treated with pour-on TCBZ at 30 mg/kg in combination with abamectin (TA). Blood samples were taken immediately prior to treatment and serially after treatment to assess the plasma profile of TCBZ metabolites. Ten weeks after treatment all animals were slaughtered and total liver fluke counts, fluke egg counts and liver pathology were assessed.
Results   Both the TOS and TA treatments resulted in significant reductions of 28-day-old liver fluke, as assessed by fluke counts and fluke egg counts at slaughter, and the reductions following TOS treatment were significantly greater than those following TA treatment. The blood profile of TCBZ metabolites in TOS-treated animals showed a significantly greater area under the plasma concentration time curve and a higher maximum observed concentration than those treated with TA. There was significantly less liver pathology in TOS-treated animals than in the TA-treated animals.
Conclusion   TCBZ administered orally at 12 mg/kg resulted in greater efficacy against 28-day-old, early immature liver fluke than was achieved by topical administration at 30 mg/kg. Plasma metabolites of TCBZ were higher and liver pathology was less in TOS-treated animals than in TA-treated animals.  相似文献

There is a need to improve the lean tissue content of ruminant animals destined for meat production. Muscle fiber number is set during fetal development. The effect of undernutrition of pregnant ewes on subsequent muscle fiber characteristics of their offspring was investigated. The trial involved 32 pregnant ewes carrying twins. The ewes were allocated randomly to one of four groups: three different treatment groups (n = 8) and a control group (n = 8). The diet of the treatment groups was dropped to 50% of their daily requirement to support the ewe and allow for conceptus growth for varying periods before being returned to 100% of their daily requirement until term. Group d 30-70 ewes were fed 100% of their daily requirement until d 30, the diet was then decreased to 50% until d 70; it was then returned to 100% of their daily requirement until term. Group d 55-95 ewes were similarly restricted from d 55 through 95, and Group d 85-115 ewes were restricted from d 85 through 115. The control group was fed 100% of their daily requirement to support the ewe and allow for conceptus growth throughout gestation. After parturition, the lactating ewes were fed a normal commercial diet. On d 14 (after parturition), the lambs were slaughtered and the LM, semitendinosus (ST), and vastus lateralis (VL) were dissected and snap frozen. The immunochemical determination of myosin heavy-chain slow (MHC-slow) and myosin heavy-chain fast (MHC-fast) proteins was measured by immunoprobing of Western blots. The number of fast and slow fibers and the diameter of these fibers also were measured in each muscle sample by histochemical techniques. Decreased maternal nutrition before fiber formation (d 30 through 70) was observed to change the muscle characteristics of the newborn lambs. These lambs had significantly fewer fast fibers (P < 0.001) and significantly more slow fibers (P < 0.001) in both the LM and VL compared with the other groups. Maternal nutrient restriction at the other periods had no effect on the number of muscle fibers in the newborn lambs; however, a decrease (LM, P < 0.05; VL, P < 0.01; ST, P = 0.08) in muscle weight was observed in the lambs born to the ewes restricted between d 85 and 115 of gestation compared with the other groups. This study has shown that decreased maternal diet before muscle fiber formation will alter the muscle fiber development in the fetus.  相似文献

Feeding sheep concentrate-based diets increases the oleic acid content of their tissues, whereas the cis-9, trans-11 conjugated linoleic acid (CLA) content is increased by feeding forage diets. Both these metabolic transformations could be attributable to increased activity of stearoyl-CoA desaturase (SCD). Therefore, the effect of forage or concentrate feeding regimens on the fatty acid composition of sheep tissues were investigated to determine whether any changes are related to an alteration of SCD mRNA levels. Twenty-four ewe lambs were randomly allotted to one of three dietary treatment groups: 1) dehydrated grass pellets, 2) concentrate diet fed to achieve a growth rate similar to that of the dehydrated grass pellets, and 3) the same concentrate diet approaching ad libitum intake. As expected, animals fed ad libitum concentrates grew at a greater (P = 0.001) rate (280 g/d) than those fed either of the other two diets (180 g/d), which were similar. In samples of liver and the three adipose tissue depots studied, the concentration of oleic acid from sheep fed either level of the concentrate diet was greater (P < 0.001) than from animals fed forage. This was associated with an increase (P < 0.05) in the ratio of SCD to acetyl-CoA carboxylase mRNA in adipose tissue and liver. Compared with concentrate-fed, the forage-fed lambs had increased (P < 0.05) levels of the cis-9, trans-11 isomer of CLA and C18:1, trans-11 in all their tissues, although the levels of SCD mRNA were lower. It therefore seems that the increased oleic acid content of sheep tissues in response to concentrate-rich diets is associated with an increase in SCD gene expression. By contrast, the increased concentration of CLA in animals fed forage-based diets is associated with an increase in substrate (C18:1 trans-11) availability.  相似文献

Haematological investigation is an important part of disease diagnosis. This is particularly so when investigating individual animal disease. It may also be important when investigating diseases in groups of animals, but the opportunity to perform necropsies to directly detect disease processes often diminishes its usefulness. Haematological investigation is essentially similar for all species. The presence of nucleated erythrocytes and thrombocytes in nonmammals requires alteration of haemoglobin measurement and cell counting. In addition, it may cause some confusion in identification of cells in peripheral blood films. Examination of blood films is an important component of haematological investigation and provides useful information on erythroid, leukocytic and platelet/thrombocytic alterations. Interpretation of alterations is essentially similar for all species. However, cell identification can at times be difficult. There are five basic leukocytes in all species: neutrophil (mammals) or heterophil (nonmammals), eosinophil, basophil, lymphocyte and monocyte. In nonmammals it may be difficult at times to distinguish between heterophils and eosinophils. In addition, lymphocytes may be confused with thrombocytes. However, a common-sense approach to the examination of the peripheral blood film will minimise this confusion.  相似文献

Two in vitro immunological assays were developed for detection of the epsilon (epsilon) antigen of Cl. perfringens type D. It was found that the reverse phase passive haemagglutination assay (RPHA) was able to detect concentrations of epsilon-antigen as low as 6 x 10-7 mg/ml whereas the single radial immunodiffusion techniques (SRID) was capable of detecting concentrations of epsilon-antigen above 0.01 mg/ml. When applied to gut contents from freshly dead infected sheep the RPHA test was found to be more sensitive than mouse toxicity assay in detecting the presence of epsilon-antigen. However, very low titres were detected in gut contents from normal sheep which meant that in a diagnostic situation interpretation of RPHA titres would be difficult. No epsilon-antigen was detected by SRID in gut contents from normal sheep or in gut contents from freshly dead infected sheep. The SRID assay could detect epsilon-antigen in gut contents from infected sheep allowed to decompose for 20 h post-mortem.  相似文献

Although de novo glucose synthesis was 47 per cent higher in wethers fed hay plus barley than in wethers fed hay alone, the plasma glucose concentrations were similar. When two chronic jugular vein catheters were implanted, plasma glucose concentrations rose by 17 per cent and 5 per cent after 4 h in those fed hay alone and hay plus barley respectively. The arthritic form of swine erysipelas was induced in pigs by multiple intravenous inoculation of 2 strains of Erysipelothrix rhusiopathiae. The strains differed significantly in their arthritogenicity but not in the number of cases of lameness induced. The use of 3 intravenous inoculations instead of 5 did not significantly affect the outcome. In a second trial, the more arthritogenic strain was injected in ten-fold dilutions from 5 x 10(9) to 5 x 10(4) organisms. Pigs receiving the lower doses showed high variability in their arthritic responses that precluded sensitive analysis of the dose effects on the number of arthritic and infected joints. However, slaughter weights showed a significant negative correlation with dose. Mean slaughter weights in treatment groups varied by 14.6 kg per pig, an average weight loss of 3 kg per pig for each ten-fold rise in dose of the highly virulent strain, and significantly correlated with the number of arthritic and infected joints. Culture of homogenised synovial membrane through selective horse meat-serum broth containing kanamycin, neomycin and vancomycin identified 66% and 59% more infected joints than primary blood agar culture of synovial fluid or synovial membrane homogenate, respectively.  相似文献

The clinical and pathological features of 19 neonatal Holstein-Friesian calves affected with moderate to severe neurological disease are presented. Most calves were recumbent from birth, and many developed variable neurological signs including hyperaesthesia or depression, limb extension, head tremor, nystagmus, apparent blindness, and opisthotonos when stimulated. Consistent lesions of moderate to severe, diffuse, axonal swelling and loss, with Wallerian-type degeneration and myelin depletion in the spinal cord and brainstem, and occasionally in the midbrain and peripheral nerve roots, were observed. The lesions indicated a pre-natal insult affecting mainly motor areas of the foetal neuraxis, however the aetiology of the disorder remains undetermined. It is suggested that the calves may have been affected by a hitherto unrecognised disease entity for which we propose the term, degenerative axonopathy.  相似文献

SUMMARY: Biochemical profiles, restriction endonuclease analysis (REA) and ribotyping were used to investigate Pasteurella multocida isolates from outbreaks of fowl cholera on 7 turkey farms in New South Wales. While only a single isolate was available from 5 of the farms, multiple isolates, 4 and 12 respectively, were available from the other 2 farms. The available field evidence suggested that 8 outbreaks had occurred with one farm suffering 2 outbreaks. The isolates obtained were all confirmed as Pasteurella multocida . Biochemical profiles allocated the isolates to 4 groups, 3 being variants of P multocida subsp multocida and the fourth being P multocida subsp septica . REA performed with Hpall established 7 groups. Ribotyping using the Hpall digests probed with the 16S rRNA operon of Haemophilus paragallinarum recognised the same 7 groups as REA. Unlike the biochemical profiles, both REA and ribotyping provided a fine subdivision that identified outbreaks as either related or unrelated. The REA and ribotyping patterns as well as biochemical profiles were stable for all isolates from the outbreaks in which multiple isolates were obtained from either the same bird or from different birds. REA and ribotyping were found to be superior to biotyping methods for the investigation of fowl cholera outbreaks.  相似文献
