The avian coronavirus infectious bronchitis virus (IBV) strain Beaudette is an embryo-adapted virus that has extended species tropism in cell culture. In order to understand the acquired tropism of the Beaudette strain, we compared the S protein sequences of several IBV strains. The Beaudette strain was found to contain a putative heparan sulfate (HS)-binding site, indicating that the Beaudette virus may use HS as a selective receptor. To ascertain the requirements of cell-surface HS for Beaudette infectivity, we assayed for infectivity in the presence of soluble heparin as a competitor and determined infectivity in mutant cell lines with no HS or glycosaminoglycan expression. Our results indicate that HS plays a role as an attachment factor for IBV, working in concert with other factors like sialic acid to mediate virus binding to cells, and may explain in part the extended tropism of IBV Beaudette.  相似文献

Abstract

Extensive forms of shrimp aquaculture have become an important source of income for farm households in the brackish water region of the Mekong Delta, Vietnam. The economic and production characteristics of farms are discussed in this paper, based on a 1997 farm survey, with particular emphasis on shrimp farming practices. Two types of shrimp fanning practice were observed, one based on natural recruitment of seedstock with few supplementary inputs, and the other based on relatively high cash investments in Penaeus monodon seedstock and other inputs. Households practising monodon culture made significantly more income, but faced high risk associated with shrimp mortality. An investigation of the factors affecting monodon yields indicated that investment in higher quality inputs had a positive effect on yield and income. However, further research is needed on the extent to which management can reduce risk of shrimp mortality by investing in such premium quality inputs.  相似文献

The aim of this study was to compare the efficacy of 2 surgical procedures for providing an adequate laryngeal airway in dogs after surgically induced laryngeal paralysis. The laryngeal function of 10 healthy, adult experimental dogs was assessed by clinical examination, laryngoscopy, arterial blood gas measurement and analysis of tidal breathing flow-volume loops before, and after, bilateral recurrent laryngeal neurectomy. A castellated laryngofissure with vocal fold resection was done on 5 dogs and bilateral arytenoid cartilage lateralisation performed on the remaining 5 dogs. Six weeks later, the laryngeal function of the 10 dogs was reassessed, before the dogs were killed and a necropsy performed. The visible changes in laryngeal structure were recorded. Both surgical procedures alleviated some of the airway obstruction caused by the bilateral recurrent laryngeal neurectomy, but bilateral arytenoid cartilage lateralisation produced more consistent clinical improvement, a wider rima glottidis, increased inspiratory air flow and a significant increase in post-operative arterial oxygen tension.  相似文献

Eight crossbred barrows (71 kg initial BW) were allocated to two treatments involving daily i.m. injections of either excipient (control) or recombinantly derived porcine somatotropin (120 micrograms/kg of BW). On d 8 of treatment, beginning 15 h after injection, glycerol and nonesterified fatty acid (NEFA) kinetics were determined using a primed, continuous infusion of [2-3H]glycerol and [9,10(n)-3H]oleic acid. Kinetics were examined under both basal conditions and during a hyperinsulinemic/euglycemic clamp. Plasma concentrations of NEFA and glycerol and their respective entry rates were highly correlated. Insulin had no effect on plasma glycerol or glycerol entry rate, probably due to the very low rates that were observed in both the control and somatotropin-treated pigs. However, both plasma NEFA and oleic acid entry rate were reduced during hyperinsulinemia. Although indices of fat mobilization tended to be higher in pigs treated with somatotropin, the magnitude of the increases were small and would be sufficient to account for only a minor fraction of the decreased lipid accretion observed in somatotropin-treated pigs.  相似文献

Twelve barrows (61 kg) received daily injections of either pituitary-derived porcine somatotropin (pST; 120 micrograms/kg body weight) or an equivalent volume of excipient. Blood samples were taken 1 d before initiation and on d 9 of treatment to characterize the temporal patterns of blood hormones and metabolites. Nitrogen (N) and energy (E) balances were performed on d 14 through 19 of treatment. Pigs treated with pST retained more N than controls (38.4 vs 23.0 g/d, P less than .01) and had a slightly higher apparent digestibility of N and E (88 vs 84% and 88 vs 85%, respectively, P less than .05). However, this improvement in apparent digestibility was probably a consequence of the lower feed intake (23% reduction) rather than a direct effect of pST. Treatment with pST resulted in chronic elevations in circulating concentrations of insulin-like growth factor I (406 vs 171 ng/ml), glucose (114 vs 86 mg/dl), insulin (3.9 vs 1.6 ng/ml), and nonesterified fatty acids (NEFA; 135 vs 59 mu Eq/liter). Diurnal patterns of glucose, insulin, and NEFA were also altered in pST-treated pigs. Plasma urea N levels were decreased with pST treatment (9.6 vs 24.0 mg/dl), but the diurnal pattern was unaltered. Our results indicate that the effect of pST is primarily on postabsorptive use of nutrients. In growing pigs, pST exerts a profound effect on N metabolism and on circulating concentrations of hormones and metabolites.  相似文献

Sixteen crossbred sows (Yorkshire x Duroc) were used to determine the effect of recombinantly derived porcine somatotropin (pST) on lactational performance and the pattern of plasma metabolites and growth rate of nursing pigs. Daily s.c. injections of either pST (8.22 mg.sow-1.d-1) or excipient were administered at 1000 on d 12 through d 29 of lactation. Jugular cannulas were inserted in three sows/treatment and hourly blood samples were collected on d 11 to 13 and d 28 to 29 of lactation to determine the effect of treatment on plasma concentrations of somatotropin, glucose and nonesterified fatty acids in plasma. Milk production and weight of nursing pigs were determined pretreatment (d 9 and 10) and on d 16, 22 and 28. Milk production of sows receiving pST progressively increased above that of control sows and was 22% greater on d 28. Milk composition was not affected by treatment with pST (P greater than .10), so that the increase in yields of milk fat, lactose and solids paralleled the increases in milk yield. Total milk protein yield tended to be higher in sows receiving pST, but protein yield was greater (P less than .10) only on d 28. Pigs suckling sows treated with pST weighed .34 kg more at the end of the lactation period (P less than .05). Sows receiving pST consumed less feed (P less than .05) during the treatment period, and, as a result, lost more weight (P less than .10) and backfat (P less than .05) than control sows. Average concentrations of plasma somatotropin were elevated approximately 2.5-fold above baseline levels by exogenous pST. No acute alterations in plasma glucose or nonesterified fatty acids were observed in response to pST treatment, however, sows receiving pST had a chronic elevation of plasma glucose on d 29 of lactation.  相似文献

The objectives of this study were to determine the effects of daily injection of bovine somatotropin (bST) on the metabolism of N and 1-[14C]leucine and on hormone and metabolite concentrations in growing beef steers. Injection of bST increased N retention (P less than .05) primarily through decreased (P less than .10) urinary N excretion. Plasma concentration of somatotropin, insulin and glucose increased (P less than .01) and of urea-N (P less than .01) and alpha-amino-N (P less than .10) decreased with bST compared with excipient injection. Total leucine flux was not altered by treatment; however, the partition of flux was. Leucine oxidation decreased (P less than .05) and leucine used for protein synthesis (P less than .10) increased, with bST compared with excipient injection. During excipient injection, 10.3 g protein were synthesized for each gram crude protein deposited, whereas during bST injections only 6.4 g were required. The average maximum contribution of myofibrillar protein degradation to whole body protein degradation, calculated from excretion of 3-methylhistidine, was 16%. Although the ratio of protein deposition/protein synthesis was low for both excipient- and bST-injected steers, the incremental efficiency of protein deposition was 50%, reflecting a dilution of protein synthesis required for turnover and a proportionately greater increase in protein synthesis than protein degradation with bST injection. In growing beef steers, bST stimulated whole body protein synthesis and decreased leucine oxidation. The change in partition of leucine flux, but not of total flux (irreversible loss), demonstrates a chronic redirection in metabolism consistent with homeorhetic control. These data from steers injected with bST suggest mechanisms by which bST affects metabolism during normal growth.  相似文献

Fresh fecal samples were collected from seven adult female reticulated giraffe (Giraffa camelopardalis reticulata). Samples were collected for several weeks before, during, and for a few weeks after gestation. Fecal samples were analyzed for progesterone levels by radioimmunoassay. There were significant differences in progesterone levels between pregestational and gestational samples and between gestational and postgestational samples. These results demonstrate that fecal progesterone levels are useful in determining pregnant versus nonpregnant reticulated giraffe.  相似文献

The treatment of young guayule plants with 2-(3,4-dichlorophenoxy)-triethylamine stimulated the accumulation of polyisoprenoid rubber in the stem and root tissues. This finding suggests that rubber productivity can be improved by the use of chemical agents on guayule and other rubber-forming plants.  相似文献
