The present study describes the genotypic distribution of rotaviruses (RVs) in an Indian bovine population with unexpectedly higher proportions of G3 alone or in combination of G8/G10. PCR-genotyping confirmed that 39.4% (13/33) of the prevalent RVs were the G3 type while 60.6% (20/33) were dual G3G10 or G3G8 types. P typing revealed that 93.9% (31/33) of the samples were P[11] while 6.1% (2/33) possessed a dual P[1]P[11] type. Sequence analysis of the VP7 gene from G3 strains viz. B-46, 0970, and BR-133 showed that these strains had sequence identities of 90.5% to 100% with other bovine G3 strains. The highest identity (98.9% to 100%) was observed with RUBV3 bovine G3 strains from eastern India. The G3 strains (B-46, 0970, and BR-133) showed 97.5% to 98.8% sequence homologies with the Indian equine RV strain Erv-80. Phylogenetic analysis demonstrated that G3 strains clustered with bovine RUBV3 and J-63, and equine Erv-80 G3. Overall, these results confirmed that the incidence of infection by RVs with the G3 genotype and mixed genotypes in the bovine population was higher than previously predicted. This finding reinforces the importance of constantly monitoring circulating viral strains with the G3 genotype in future surveillance studies.  相似文献

Summary Mungbean is an important source of vegetable protein for the growing population in many developing countries of South East Asia. Its production is limited due to its susceptibility to diseases and insect pests besides many other undesirable agronomic traits. Strategies for increasing and stabilising its production have been to develop varieties resistant to diseases, pests and with other desirable agronomic traits. Genetic improvement of this crop by classical breeding has met with limited success due to the lack of sufficient and satisfactory level of genetic variability within germplasm. Recent advances in biotechnology have offered the opportunity to develop new germplasms. The development of such technologies largely depends on efficient regeneration of sexually mature plants from organs, tissues and protoplasts. An overview of plant regeneration by direct or indirect organogenesis and embryogenesis is presented. The use of in vitro and molecular techniques such as somaclonal variation, screening for various desirable traits, interspecific crossing and genetic transformation to supplement conventional breeding, for genetic improvement of this crop is described. The advantages and limitations of these techniques along with directions for future research are discussed.  相似文献

During 1998, hydropericardium syndrome was observed among 3-to-6-wk-old broilers in 45 different flocks of Haryana, India, with mortality ranging between 10% and 30%. Fowl adenovirus (FAV) was isolated from one of the affected flocks by chicken embryo liver cell culture. Serum neutralization test and polymerase chain reaction assay coupled with restriction enzyme analysis confirmed that the isolated virus belonged to FAV serotype 4. The disease was reproduced in 28-day-old broilers by subcutaneous and oral inoculation of isolated FAV4 alone. Typical hydropericardium and basophilic intranuclear inclusions in hepatocytes were observed in experimental birds by day 4 postinoculation.  相似文献

Green tea manufacture was standardized with respect to the inactivation of polyphenol oxidase (PPO), rolling, and drying for quality manufacture. Inactivation of PPO by parching, steaming, microwave heating, and oven heating was monitored in tea shoots. The inactivated shoots were rolled under regimens of high and low pressures and dried by microwave heating, oven heating, or sun-drying; total phenols and catechins were estimated. Parched and sun-dried teas contained the lowest levels of total phenols and catechins, and their infusions were dull in color with a slightly burnt odor. Microwave-inactivated and-dried teas showed the highest levels of total phenols and catechins, and their infusions were bright in color and sweet in taste with a subtle pleasant odor. In steam-inactivated and oven/microwave-dried teas, total phenol and catechin contents were intermediate between parched and sun-dried teas and microwave-inactivated and microwave-dried teas, and their infusions were bright with a umami taste.  相似文献

According to Starling's law of the heart, the force of contraction during the ejection of blood is a function of the end-diastolic volume. To seek the molecular explanation of this effect, a study was made of the effects of length on Ca2+ sensitivity during tension development by isolated demembranated cardiac muscle in which the cardiac form of troponin C was substituted with skeletal troponin C. The results of troponin C exchange were compared at sarcomere lengths of 1.9 and 2.4 micrometers. Enhancement of the myocardial performance at the stretched length was greatly suppressed with the skeletal troponin C compared with the cardiac troponin C. Thus the troponin C subunit of the troponin complex that regulates the activation of actin filaments has intrinsic molecular properties that influence the length-induced autoregulation of myocardial performance and may be a basis for Starling's law of the heart.  相似文献

Acquaintance of potassium (K) transformation in soil is crucial for K management in management system of arid India. Present study revealed that the extractable K like ammonium acetate–potassium (NH4OAc–K), nitric acid (HNO3)–K, and hydrochloric acid (HCl)–K under different systems were followed the order: irrigated low input > rainfed low input > irrigated high input > no input. The pearl millet systems in irrigated low input practice maintained a higher NH4OAc–K than irrigated high input practice. The exchangeable-K (Exch-K), non-exchangeable-K (Non-exch K) and lattice-K were observed highest in irrigated low input system and lowest in no input system. It is noticed that Exch-K contributed 2.3–2.6% over total-K, which readily available to plants. The Exch-K was highly significant correlated with K pools and soil properties, indicating that rapid establishment of equilibrium between the pools. Thus, knowing K reserve and distribution of K forms can help in understanding K replenishment capacity of arid soil under long-term intensive cultivation.  相似文献
