The efficacy of intranasal vaccination in preventing or limiting disease of the lower respiratory tract induced by parainfluenza 3 (PI3) virus was evaluated under experimental conditions, using a commercially available live vaccine containing a temperature-sensitive strain of PI3 virus. In a preliminary study four colostrum-deprived calves were vaccinated intranasally at one week and again at two months of age, and two similar calves were given an intranasal placebo. After the second vaccination serum antibodies to PI3 virus were detected in all four vaccinated calves, but not in the control animals. Seventeen days after the second vaccination all six calves were challenged with virulent PI3 virus, and they were killed six days later. The clinical scores and the extent of pulmonary consolidation were reduced in the vaccinated animals; PI3 virus was detected in the upper and lower respiratory tract of the control calves but in none of the vaccinated calves. In a larger scale study with 14 colostrum-fed calves, seven were vaccinated at one week and again at five weeks of age, and seven were given an intranasal placebo. Two weeks after the second vaccination all 14 calves were challenged with virulent PI3 virus. The clinical scores and lung consolidation were significantly reduced in the vaccinated calves in comparison with the controls. Six days after infection, 10 of the 14 calves were killed; PI3 virus was detectable in the nasal secretions of all seven control calves but in only one of the vaccinated animals, and PI3 viral antigen was detected in the lungs of the control calves but not in those of the vaccinated animals. One of the vaccinated calves had developed a severe clinical response after the challenge, but it had only minor lung consolidation when killed.  相似文献

A blocking enzyme-linked immunosorbent assay (ELISA) was developed to detect antibodies to porcine rubulavirus (La Piedad Michoacan Virus [LPMV]) in serum samples from pigs. The test, based on a monoclonal antibody against the LPMV hemagglutinin-neuraminidase glycoprotein, had a sensitivity of 99% and a specificity of 97%. The results of this test were in agreement with those obtained by an indirect ELISA and hemagglutination inhibition, indirect immunofluorescence, and virus neutralization tests. The blocking ELISA is considered the most suitable test for routine screening for antibodies against LPMV.  相似文献

Porcine circovirus 2 (PCV2) is a recently identified agent that has been associated with postweaning multisystemic wasting syndrome (PMWS) in swine populations. In this report, the potential spectrum of disease associated with PCV2 is expanded by evidence of vertical transmission and associated reproductive failure. PCV2 was isolated from a litter of aborted piglets from a farm experiencing late-term abortions and stillbirths. Severe, diffuse myocarditis was present in 1 piglet associated with extensive immunohistochemical staining for PCV2 antigen. Variable amounts of PCV2 antigen were also present in liver, lung, and kidney of multiple fetuses. The presence of other agents that have been associated with fetal lesions and abortion in swine, including porcine parvovirus, porcine reproductive respiratory syndrome virus, encephalomyocarditis virus, and enterovirus, could not be established.  相似文献

Two concepts for use in the fabrication of three-dimensional (3D) microstructures with complex topologies are described. Both routes begin with a two-dimensional (2D) pattern and transform it into a 3D microstructure. The concepts are illustrated by use of soft lithographic techniques to transfer 2D patterns to cylindrical (pseudo-3D) substrates. Subsequent steps-application of uniaxial strain, connection of patterns on intersecting surfaces-transform these patterns into free-standing, 3D, noncylindrically symmetrical microstructures. Microelectrodeposition provides an additive method that strengthens thin metal designs produced by patterning, welds nonconnected structures, and enables the high-strain deformations required in one method to be carried out successfully.  相似文献

For the first time was characterized the semen of Pimelodus britskii, hormonally induced and non‐induced induced, during the reproductive period. The experiment 1 was conducted with 12 fish per month, divided into: (i) induced with Carp Pituitary Extract (CPE), and (ii) without hormonal induction (testes macerated). The experiment 2 was conducted, with 30 fish, divided into groups for comparison of different doses of CPE and human Chorionic Gonadotrophin (hCG; T1: control; T2: 3.0 mg kg?1 CPE; T3: 0.5 mg kg + 3.0 mg kg?1 CPE; T4: 0.5 mg kg?1 + 7.0 mg kg?1 CPE; T5: 200 UI kg?1 hCG), the same fish were used every month, and the semen was collected by abdominal massage. In both, experiments were assessed the sperm motility and velocity by means of the CASA software. In experiment 1, the concentration of spermatozoids was significantly increase by application of CPE compared untreated males. The volume and motility decreased gradually until the end of the experiment, the highest values were recorded for treatment induced (0.49 ± 0.25 mL and 62.18, respectively). The same occurred to Gonadosomatic Index, showing the smallest value at the end of the reproductive period. The fish from experiment 2 released reduced volume of watery semen (0.1 mL). The values of sperm concentration, motility velocity decreased gradually throughout the months. For P. britskii, the reproductive period influenced the production and sperm quality. Despite small seminal volume released the dose of 7.5 mg kg?1 of CPE proved effective.  相似文献

In each of two seasons, undisturbed lysimeters 0.8 m in diameter and 1.05 m in length taken from five soil types were cropped with winter wheat. They received autumn applications of the pesticides isoproturon and linuron as well as a bromide tracer and spring applications of dimethoate and MCPA. Leachate was collected at regular intervals and concentrations of the various solutes determined. Rainfall from December to March was 290 and 191 mm in the first and second seasons, respectively. Both springs were exceptionally dry with less than 50% of the mean April‐to‐June rainfall of 138 mm. Total flow from the lysimeters ranged from 335 to 477 mm (and from 0.78 to 3.95 pore volumes) over the two seasons. Leaching to drainage of bromide highlighted soils where preferential flow was influential with total losses ranging from 24% of applied for a strongly structured, alluvial clay loam to 79% for an unstructured sand. Leaching to drainage of isoproturon (Koc ≈ 100 ml g−1) was observed from all but a peat soil with losses greater (0.31–1.01% of applied) from the clay loam and a deep medium loam, where patterns of leaching clearly indicated preferential flow mechanisms, than from the sand and a light loam over gravel (0.04–0.18% of applied) where a broad breakthrough curve indicated that matrix flow was more important. Linuron (Koc ≈ 500 ml g−1) was detected in occasional samples of leachate from the clay loam, the light loam over gravel and the medium loam during the first season only (maximum loss 0.12% of applied). The sandy soil, often considered most vulnerable to leaching, gave the smallest total losses of pesticide of the four mineral soils, whilst significant preferential flow in the deep, medium loam was believed to result from a compacted topsoil. Residues of tri-allate in triticale (X Triticosecale Wittmack) were determined, after both preplant and pre-emergence soil-incorporated applications of tri-allate at 1.4 kg ha−1, at the following crop stages: 2–3-leaf, 5–6-leaf, fully headed and mature (straw and grain). Tri-allate residues were detected only in green tissue samples from the 2–3-leaf stage of the crop. Recoveries of tri-allate were in the order of 90 and 85 % from the green tissue and grain at the 100 and 50 μg kg−1fortification levels, respectively.  相似文献

Scenario-based investigations explore alternative future courses of action in a widening array of situations. Anticipating landscape patterns and the values behind them are recurring needs in such investigations. While it is accepted that how scenario assumptions are framed and who frames them matters, the sensitivity of resulting trajectories to contrasting scenario framing and modeling processes is rarely tested. Using comparable scenarios we contrast landscape change trajectories produced from two distinct approaches to modeling scenario assumptions: the first uses lay citizen groups and deterministic land allocation modeling, the second uses experts from biophysical and social sciences and agent-based modeling. Scenarios are defined and mapped for the year 2050 in western Oregon’s Willamette River Basin along a gradient of conservation oriented to development-oriented assumptions using first citizen-based and then expert-based approaches. The landscape variability and trajectories for the citizen-based Conservation 2050 and Development 2050 scenarios are then characterized and compared with those of the expert-based Conservation 2050 and Development 2050 scenarios. Results distinguish areas where trajectories always vary regardless of approach or scenario from those that never vary. Policy influence on trajectory is illustrated using agent-based model results where land conversion serves purposes of wealth production and ecosystem function. Results depict areas with strong coupling between policy and trajectory as those places experiencing the same pattern of change over time regardless of scenario. Results also indicate that the greater the variability of a given scenario’s trajectories, the more successful the scenario is at avoiding scarcity of wealth and ecosystem function.  相似文献
