The erythrocyte-exchanged chimera mouse model has become to be a significant tool for studying animal and human (hu) protozoan haemoparasites, though the usefulness of this model varies depending primarily on the acceptability of xenogeneic red blood cells (RBC). To find a superior recipient in comparison with C.B-17/Jcl mouse with severe combined immuno-deficiency (scid) mutation, we examined in this report the non-obese diabetes (NOD)/shi-scid mouse, a recently available strain of SCID. When 2.5 x 10(8) of fluorescent dye-labeled hu-RBCs were transfused, C.B-17scid mouse eliminated them logarithmically by a simple linear regression, while NOD-scid mouse eradicated hu-RBCs by a unique two-step fashion, i.e., a potent but only briefly functioning RBC eradication followed by a weak steadily functioning step. The means of regression line constance +/- their standard deviations (SD) of 205 C.B-17scid and of 213 NOD-scid mice for their short- and long-lasting steps were -0.73 +/- 0.63, -0.53 +/- 0.25 and -0.16 +/- 0.10, respectively. Hu-RBC half-lives determined from these means of C.B-17scid mice and of NOD-scid mice for the short- and long-living steps were 3.6, 4.9 and 16.3 hr, respectively. Higher hu-RBC acceptability of NOD-scid mouse, especially at their long-lasting step, was also demonstrated under at an activated state of mouse innate immunity. Treatment with 1.0 mg heat-killed Candida cells caused an acceleration of hu-RBC elimination in both mouse strains but the magnitudes for the short- and long-living steps of NOD-scid mice evaluated by "stimulation index" were only 1/2.6 and 1/7.6 of C.B-17scid mice, respectively.  相似文献

As a part of the elucidation of the pathogenesis of anemia in Theileria sergenti infection, oxidized-erythrocyte membrane proteins (OEMPs) collected from T. sergenti-infected calves were examined. The amount of OEMPs were seen to increase with the progress of the anemia and showed a maximum value around the crisis period of the infection. The increase of OEMPs coincided with band Nos. 1, 2, 2.1, 3, 4.1, 5, 6, and 7. The majority of them was located at the Triton X-100 un-extractive phase, and was confirmed as cytoskeletal proteins. This evidence indicates the enhancement of erythrocytic oxidation, and suggests that it might be one of the aggravating factors of anemia in T. sergenti infection.  相似文献

Three different polymerase chain reaction (PCR) protocols were evaluated for their ability to detect bovine herpesvirus 2 (BoHV-2): single-step PCR with 3 reaction stages (denaturation, annealing and extension), 2 reaction stages (denaturation and annealing/extension; shuttle PCR), and semi-nested PCR with 3 reaction stages. All the PCR protocols showed the same sensitivity (detection limit of 0.4 TCID(50)). A non-specific band sometimes appeared in mock cell DNA at annealing temperatures below 64 degrees C. The shuttle PCR was found to be superior to the other protocols under consideration because of the speed of its application. Furthermore, no non-specific band was detected in DNAs of eight other DNA viruses. Thus, the shuttle PCR seems to be an excellent diagnostic tool for BoHV-2 infections.  相似文献

Leaf blast suppression in multilines was evaluated based on the number of susceptible lesions observed in a pure stand of susceptible rice cultivar Sasanishiki, and in 1 : 1 and 1 : 3 mixtures of Sasanishiki and a resistant near-isogenic line, Sasanishiki BL4 or BL7, from 1998 to 2001. The number of lesions first observed in fields in the 1 : 1 and 1 : 3 mixtures were close to theoretical numbers calculated using the number of lesions observed in the pure stands and the ratios of the susceptible Sasanishiki in the mixtures. The ratio of the number of lesions in the 1 : 1 and 1 : 3 mixtures to the number in the pure stand was 0.29 and 0.09, respectively. The relationship between these ratios and the ratios of susceptible Sasanishiki in mixtures was defined in an equation to estimate the degree of leaf blast suppression. Validation studies for the ratios of the number of lesions in the 1 : 1 and 1 : 3 mixtures to the number in the pure stand were conducted in two different locations and showed that the ratios are almost acceptable. The calculated autoinfection to alloinfection ratio was 1.3 and 1.4 in the 1 : 1 and 1 : 3 mixtures, respectively, suggesting that the calculated ratio will affect the degree of leaf blast suppression. Thus, predictors were obtained to estimate leaf blast suppression for effective blast control in multilines.  相似文献

A spacing experiment for Japanese oak (Quercus mongolica var.grosseserrata) with Nelder's design have been studied at the Kyushu University Forest in Hokkaido, since 1975. The oak seedlings were planted according to systematic spacings which were 20 levels varying gradually from 40,816 to 1,086 seedlings per hectare with concentric circular arcs (22 in number) and spokes (41 in number). In 1992 when the planted oak seedlings were 18-year-old, DBH, crown diameter, total height and clear length of the survival trees were measured. Based on the results obtained from analyzing the relationships between planting density and the mean values of each block, the optimum planting density should be 7,241 seedlings per hectare. The title is tentative translation from the original Japanese title by the authors of this paper.  相似文献

We studied the effect of genotypes of planting stocks regarding the variation of the modulus of elasticity of tree trunks on standing trees (trunk-MOE), tree height (TH), and diameter at breast height (DBH) in a 19-year-old Japanese cedar plantation made with root cuttings. Trunk-MOE was assessed nondestructively using a tree-bending method. Genotypes of individual trees were detected using the random amplified polymorphic DNA (RAPD) technique. RAPD analysis revealed that the sampled plantation consisted of 14 genotypes. Genotypic effects on DBH and TH were unclear, and there was no significant difference among genotypes. This result indicated that an acquired variation should have more influence than an inherited variation on DBH and TH. For trunk-MOE, there were significant differences among the four largest genotypes at the 5% level. However the coefficient of variation in trunk-MOE of each genotype ranged from 7.5% to 26.8%. It seems reasonable to assume that the wide variation in trunk-MOE in a sampled plantation may depend on the environmental effect within a clone as well as on the genetic origin of clones. We therefore conclude that the use of multiple planting stocks from different cuttings for which the wood quality is unknown contributed strongly to the wide variation in trunk-MOE in the plantation of Japanese cedar.  相似文献

Japanese larch wood loaded with nickel (1%–4%) alone or with nickel and calcium (0.25%–1.5%) was carbonized at 800°–900°C for 0–120min with a heating rate of 5°–20°C min−1 in a helium flow of 5.8−46.4 ml STP cm−2 min−1 to examine the influence of these variables on the crystallization of carbon (the formation of T component) and the development of mesoporosity. From the obtained results, reaction conditions suitable for effective production of carbon with the dual functions of adequate electroconductivity and adsorption capacity in liquid phase were established, thereby explaining the factors that govern the process. It was also confirmed that mesopore having a diameter of about 4 nm was selectively produced at the cost of specific (BET) surface area in parallel with the formation of T component. This result provided good insight into how the simultaneous dual function could be realized.  相似文献

This report presents a new case of mucormycosis encountered in penguin characterized by morphological variation of hyphae and presence of sporangia with numerous sporangiospores. A 4.5-year-old Magellanic penguin (Spheniscus magellanicus) died after exhibiting anorexia, poor nutritional condition and dyspnea. Multiple nodular lesions were observed in the thoracic and abdominal regions. Histopathologically, hyphae of various sizes were seen in the lungs, air sac and nodular lesions. Myriad sporangiospores and several sporangia were observed in/around the bronchi or parabronchi. The very narrow and short hyphae in the nodules were not consistent with the characteristics of Mucorales. However, for most hyphae, including those in the nodules, sporangiospores and sporangia, immunohistochemistry revealed Mucorales-positive reactions. In addition, these fungi were identified as Rhizomucor pusillus by gene analysis.  相似文献
