This study aimed to determine whether pig responses to heat stress (HS) were directly due to heat exposure (regardless of feeding level and pattern) or were indirectly due to the reduction of feed intake (FI) and to determine if increasing feeding frequency (splitting heat increments) can improve pig response to HS. A total of 48 pigs (66.1 ± 1.7 kg) were allocated to four groups in three replicates. After 7 d in thermoneutral (TN) conditions (22 °C; period 1 [P1; day −7 to −1]), pigs were placed in either TN or HS (32 °C) conditions for 20 d (period 2 [P2; day 0 to 19]). The diet was provided either ad libitum (AL; 2 distributions/d) or pair-fed (PF8; 8 distributions/d) using HS–AL pigs as the reference group. Thus, the four experimental groups were TN–AL, HS–AL, TN–PF8, and HS–PF8. The daily ration of PF8 pigs was distributed at every 90-min intervals from 0900 to 1930 hours. Data were analyzed using the PROC MIXED procedure with replicate (n = 3), experimental group (n = 4), and their interactions as fixed effects, and the REPEATED statement was used for repeated measures data. Pigs had a similar average daily feed intake (ADFI) during P1 (P > 0.05). In P2, HS–AL and PF8 pigs had lower ADFI (−19%), average daily gain (−25%), and final body weight (−6.1 kg) than TN–AL pigs (P < 0.01). TN–AL pigs had thicker backfat than TN–PF8 pigs (P < 0.05), while the HS pigs had intermediate results. HS pigs had a higher perirenal fat percentage based on the contrast analysis between PF8 pigs (P < 0.05). Thermoregulatory responses of pigs increased with HS exposure but did not differ between HS or between TN groups (P > 0.05). For TN pigs, variation in muscle temperature (Tmuscle) depended on feeding and physical activity, while for HS pigs, Tmuscle gradually increased throughout the day. The Tmuscle of PF8 pigs increased with each additional meal but plateaued earlier for HS–PF8 than TN–PF8 pigs; an increase in Tmuscle per meal was also lower in HS–PF8 than TN–PF8 (P < 0.05). Exposure to HS decreased plasma T3 and T4 (P < 0.05) and increased plasma creatinine (P < 0.05). Between the PF8 groups, HS pigs also had a transient increase in plasma insulin on day 8 (P < 0.05). The effect of HS on FI decreased the growth rate of pigs but there are heat-induced effects, such as altered physiological responses, which might explain the direct HS effects seen in other literature especially in terms of increased adiposity. The increased feed provision frequency in the present study did not improve the HS response of pigs.  相似文献

The two sympatric sibling species Culicoides obsoletus (Meigen) and Culicoides scoticus Downes and Kettle (Diptera: Ceratopogonidae), are known to be competent vectors for bluetongue virus in the Palaearctic region. However, morphological identification of constituent species is only readily applicable to adult males and these two species distinguishing traits have overlapping character states. As their vector competence may differ in space and time, the correct identification and quantification of specimens of both species are essential for understanding bluetongue epidemiology. However, no molecular tools are available for high-throughput identification of the two species. We therefore developed a quantitative duplex real-time PCR assay to determine the relative abundance of each sibling species in a sample using TaqMan probes. For each species, standard curves were constructed from serial dilutions of purified plasmid DNA containing ITS1-5.8S-ITS2 (rDNA) in the range of 10(-1) to 10(-5)ng/μL. Standard curves were used to quantify samples of mixed C. obsoletus/C. scoticus specimens. Specificity was evaluated with 5156 specimens representing 62 species. Based on the DNA quantities detected according to the standard curves, a quadratic model developed on 1100 males and validated on 555 females was able to predict the relative abundance of each species simultaneously in a one-shot reaction (Pearson coefficient of 0.999). Our assay showed a requirement of two specimens or less for 95% of the predictions, making it highly applicable to field collections. Extensive use of this real-time PCR assay will provide a better understanding of geographical distribution, dynamics, and bionomics on a species level, which is essential for risk assessment. This approach is an important contribution to medical entomology for investigating the vector role of arthropod sibling species.  相似文献

In this study we examined the influence of several factors, like the surface of skin with lesions attributable to Sarcoptes scabiei, the time of year and host sex and age on variations of mite load in Iberian wild goats, Capra pyrenaica, from the Sierra Nevada Natural Space, southern Spain. As well, the surface area of skin with scabietic lesions estimated during field surveys were compared with measurements taken in the laboratory and the potential for using categories based on surface areas estimated during monitoring and management programs was analyzed. The surface area of the skin lesions measured in the laboratory and the time of year were the main factors affecting mite load. Results also revealed discrepancies between the surface area of lesions estimated in the field and those measured in the laboratory. The highest error percentages were associated with lesions ranging between 25 and 75% of host skin surface area. By reducing the number of categories of mange infestation (from 5 to 3) the proportion of cases correctly classified using data from field surveys increases. Our analyses suggest that the estimated surface areas of scabietic lesions is a useful relative index of mite abundance and/or intensity of parasitation in Iberian wild goat. Therefore, we conclude that revised classifications should be used in the monitoring of sarcoptic mange in wild populations, since data can be obtained in an inexpensive, fast and non-invasive fashion.  相似文献

Testes mass is a key factor in male reproductive success and is potentially exposed to so-called 'parasitic castration'. This is the result of the direct destruction or alteration of reproductive cell lineages (parasitic castration sensu stricto), or the indirect detrimental effects - for example, via body condition - on the ability of progenitors to produce or rear offspring (parasitic castration sensu lato). There are enormous gaps in our knowledge on the effects of parasites on the testes of wild mammals and in an attempt to rectify this dearth of data we examined the relationship between the skin parasite Sarcoptes scabiei and testes mass in Iberian ibex Capra pyrenaica. We considered data from 222 males that were culled in the population from the Sierra Nevada in Spain. Our results provide evidence that sarcoptic mange is associated with reduced size-corrected testes mass in Iberian ibex which supports the hypothesis that parasitism is a determining factor in gonad plasticity in male mammals. We discuss several hypothetical causes of this relationship and highlight the need to deepen the sub-lethal effects of pathogens if we are to accurately understand their modulator effects on host population dynamics.  相似文献

There is increasing use of networks in ecology and epidemiology, but still relatively little application in phytopathology. Networks are sets of elements (nodes) connected in various ways by links (edges). Network analysis aims to understand system dynamics and outcomes in relation to network characteristics. Many existing natural, social, and technological networks have been shown to have small-world (local connectivity with short-cuts) and scale-free (presence of super-connected nodes) properties. In this review, we discuss how network concepts can be applied in plant pathology from the molecular to the landscape and global level. Wherever disease spread occurs not just because of passive/natural dispersion but also due to artificial movements, it makes sense to superimpose realistic models of the trade in plants on spatially explicit models of epidemic development. We provide an example of an emerging pathosystem (Phytophthora ramorum) where a theoretical network approach has proven particularly fruitful in analyzing the spread of disease in the UK plant trade. These studies can help in assessing the future threat posed by similar emerging pathogens. Networks have much potential in plant epidemiology and should become part of the standard curriculum.  相似文献

In boreal forests, historical variations in the area disturbed by natural disturbances or harvesting have rarely been compared. We measured temporal and spatial variations in areas affected by severe fires and clearcutting throughout the 20th century in a 57, 332 km2 section of the eastern Canadian boreal forest. We examined the effects of these disturbances on spatio-temporal variations in the abundance of forests >60 years. Natural variability for the abundance of forests >60 years was estimated from simulations of natural disturbance regimes. We also measured compositional and structural differences between three categories of stands originating from relatively recent disturbances (∼50 years; clearcutting, fires, and clearcutting followed by fires), and one category of stands that were undisturbed for at least 200 years. At the regional level, we observed that forests >60 years gradually became scarcer throughout the 20th century due to a gradual expansion of harvested areas, an effect most pronounced in the southern part of the region, where mature and old forest abundance was clearly outside the range of natural variability at the end of the studied period. At the stand level, forest composition and structure differed between stand-origin categories: clearcutting-origin stands contained more balsam fir (Abies balsamea), fire-origin stands more black spruce (Picea mariana), and fire/clearcutting-origin stands more hardwoods (Betula papyrifera and Populus tremuloides). Overall, we estimate that strict forest management targets based on natural disturbance regimes will be difficult to achieve in eastern North-American boreal forests, most notably because contemporary disturbance rates, including both clearcutting and fire, have gradually become higher than the fire rates observed during the preindustrial period.  相似文献

We analyzed the effect of simplifying assumptions in canopy representation of radiation transfer models, comparing modeled diffuse non-interceptance and photosynthetic photon flux density with measurements at different layers of complex pine-broadleaved canopy with large seasonal variation of leaf area index. The most detailed model included clumping of trees (i.e.,?stand density) and a vertical specification of leaf angle distribution and shoot clumping. A less detailed model replaced the vertically specified variables with their means. The most parsimonious model accounted for neither shoot clumping nor stand density. The vertical specification of shoot clumping and leaf angle distribution only slightly improved vertical and seasonal openness and light estimates over using mean values. Further simplification had little effect on total absorbed light but was more risky for estimates of the vertical distributions of openness and light absorbed by the canopy, which will affect photosynthesis estimates due to the non-linearity of photosynthetic light response. Including woody surfaces in winter, when leaf area was low, was essential for reproducing the measurements correctly. A sensitivity analysis showed that ignoring (i)?shoot clumping could result in a substantial overestimation of total absorbed light with errors increasing with decreasing leaf area and (ii) stand density in sparse stands could lead to substantial overestimation of total absorbed light, and the effect is largely independent of leaf area. Also, (iii) the effect of changing leaf angle distribution increased with decreasing leaf area, and was larger and more persistent along the leaf area range with increasing shoot clumping. Overall, accounting for the effect of tree clumping on absorbed light is most important in stands composed of species where leaves are not very clumped (e.g., broadleaved). However, even in forests with highly clumped shoots (i.e., coniferous), an accurate estimation of absorbed light distribution in stands requires incorporation of stand density in the model.  相似文献

Most long-path remote spectroscopic studies of the atmosphere rely on ambient light or narrow-band lasers. High-power femtosecond laser pulses have been found to propagate in the atmosphere as dynamically self-guided filaments that emit in a continuum from the ultraviolet to the infrared. This white light exhibits a directional behavior with enhanced backward scattering and was detected from an altitude of more than 20 kilometers. This light source opens the way to white-light and nonlinear light detection and ranging applications for atmospheric trace-gas remote sensing or remote identification of aerosols. Air ionization inside the filaments also opens promising perspectives for laser-induced condensation and lightning control. The mobile femtosecond-terawatt laser system, Teramobile, has been constructed to study these applications.  相似文献
