With the changing climatic scenario and increasing global mean temperature, heat stress became a major limiting factor for today's agriculture. To identify the underlying mechanism associated with heat tolerance in peanut, two experiments (field and growth chamber) were conducted with four genotypes (ICGS 44, GG 7, AK 159 and DRG 1) having differential high temperature stress sensitivity. Field grown plants under three different temperature (D1, D2 and D3) regimes simulated three temperature treatment effects with a variability of 3–4/4–5°C in mean day/night temperature, respectively. In growth chamber, imposition of heat shock (10°C above ambient inside growth chamber) revealed not only rapid induction (within 0.5 hr) of HSPs, especially small HSPs (HSP 17, HSP 40) in tolerant genotypes, but also its sustenance for longer duration (2 hr), which might help them to have better physiological adaptation strategies under high temperature stress. This was evident from significant advancement in phenophases observed with increase in temperature by 15–18 days at physiological maturity, while pollen viability and membrane stability reduced below 50% and 41%, respectively in DRG 1 with increase in mean day/night temperature. Maintenance of higher photosynthesis and transpiration rate and stomatal conductance helped the tolerant genotype ICGS 44 to keep relatively cooler canopy and higher photosynthates, ensuring better physiological condition in this genotype under heat stress. Significant increase (~2.5‐fold) in inositol and hexoses (glucose and fructose) content and reduction (>50%) in sucrose content in leaf tissues indicated degradation of storage carbohydrates for improved osmotic adjustment especially in tolerant genotypes under elevated temperature.  相似文献

The potential extra response that can be obtained from the optimal use of a known QTL in selection by optimizing weights in an index of breeding value for the QTL and polygenic EBV was investigated for a range of parameters. Optimal strategies were derived for a deterministic model of simultaneous selection on a QTL and polygenic effects using optimal control theory. Responses over 10 generations to the following selection strategies were compared: 1) standard QTL selection, with QTL weights equal to 1, 2) optimal QTL selection, 3) stepwise single-generation optimal QTL selection, and 4) non-QTL selection based on phenotype. Cumulative discounted response with discount rates of 10 or 30% per generation were evaluated and used as objective for optimal selection strategies. Optimal selection balanced the conflict between short- and long-term responses and gave greater cumulative discounted response than standard QTL selection of up to 20%, but less than 5% for most cases. Discount rate had limited impact. For a QTL with an additive effect of one polygenic standard deviation, cumulative discounted response from optimal QTL selection was less than 5% greater than response for non-QTL selection for most cases. Exceptions were traits with low heritability and recessive QTL at low frequency, for which extra response was up to 55% greater. Stepwise optimal selection resulted in less cumulative discounted response than standard QTL selection for QTL with negative dominance. The benefit of optimal over stepwise optimal selection was limited (less than 4%) for most cases, except for overdominant QTL. These results indicate that optimizing selection on an identified QTL can result in greater responses to selection but that extra responses tend to be limited for the situations studied here of single-stage purebred selection on a single QTL for a trait observed on both sexes.  相似文献

The present study was conducted to investigate the effects of different forms of soil acidities on microbial biomass C, ergosterol content, microbial metabolic quotient, microbial respiration quotient, and fluorescein diacetate-hydrolyzing activity of some tea-growing soils of India. Total potential and exchangeable acidity and extractable and exchangeable aluminum were higher in Tripura followed by Jalpaiguri and Kharagpur soil. Different forms of acidity were significantly and positively correlated with each other. All the microbiological properties investigated were significantly and positively correlated with soil organic C content. The ratio of organic C with microbial parameters was significantly and negatively correlated with different forms of acidity. Principal component analysis indicated that the microbial activities were not directly affected by the extractable aluminum and total potential acidity. Although the tea soils had higher microbial biomass and activities because of higher organic matter content than other soils, the ratios of microbial parameters/organic carbon indicated that inhibition of microbial growth and activities had occurred because of acidity stress.  相似文献

The aim of this study was to investigate the molecular mechanism behind androgen-induced sex reversal and growth increase in Nile tilapia. Three-day-old mixed-sex tilapia juveniles were treated with differential dose (0, 5, 10, 20, 30 mg/kg diet) and duration (0, 15, 30, 45, 60 days) regimes of 17α-methyltestosterone. Treatment regimes 10, 20, 30 mg/kg-30, 45, 60 days showed ~95% males, while 0 mg/kg-0 days (control) showed ~50% males (P < 0.01). Next, tilapia was cultured for 6 months in cistern and pond culture systems with 17α-methyltestosterone-treated diet (10 mg/kg) for three different durations: 0 days (control), 30 days (30-day-treated), and 180 days (throughout-treated). Fish in pond culture showed significantly higher (P < 0.05) final weight compared with tilapia in cistern culture (235.76, 156.82 g, respectively). Both 30-day-treated and throughout-treated fish showed significantly higher (P < 0.05) final weight, length, daily weight gain, specific growth rate, and muscle protein content compared with control fish. The androgen receptor β mRNA and protein expression levels in both 30-day-treated and throughout-treated fish were significantly higher (~twofolds, P < 0.05) compared with hormone-untreated control males. However, there was not much difference for all these parameters among the two hormone-treated groups. This higher growth probably results from an androgen-induced increased ‘androgen-androgen receptor’ mediated anabolic pathway. Serum testosterone concentration of 30-day-treated fish after 6 months was similar to that of control males (0.9 ng/ml). Moreover, culture of 17α-methyltestosterone-treated monosex tilapia was found beneficial than hormone-untreated mixed-sex tilapia culture for maintenance of natural fish diversity in indigenous water bodies.  相似文献

Field experiments were conducted in 2002-2003 and 2003-2004 to evaluate the relative performance of synthetic (black polyethylene) and organic (paddy husk and straw) mulches on soil and plant water status vis-a-vis N uptake in wheat in a semi-arid environment of India. Scope of better utilization of soil moisture was documented through all the mulches, especially during initial crop growth stages, when the moisture content was 1-3% higher in mulches. Soil temperature was more moderate under organic mulches. Paddy husk recorded significantly higher plant biomass, while the effect of mulching in enhancing root growth was clearly documented. Organic mulches produced more roots (25 and 40% higher root weight and root length densities compared to no-mulch) in sub-surface (>0.15 m) layers, probably due to greater retention of soil moisture in deeper layers and relatively narrow range of soil temperature changes under these systems. Incremental N dose significantly improved all the plant parameters in both mulch and no-mulch treatments. Grain yield was 13-21% higher under mulch and so with increasing N levels. Nitrogen uptake was higher in organic mulches and also with higher N doses, while polyethylene mulch showed mixed trend. Mulches were effective in reducing 3-11% crop water use and improved its efficiency by 25%. Grain yield and biomass were well-correlated with leaf area index (r = 0.87 and 0.91, respectively) and water use was better correlated with root length than its weight. Results indicated substantial improvement in water and N use efficiency and crop growth in wheat under surface mulching, and the organic mulches, especially rice husk performed better than synthetic mulches.  相似文献

Hilsa (Tenualosa ilisha) is an important commercial fish in India. This study investigates relationships among Hilsa catch per unit effort (CPUE) and the corresponding lunar phase, rainfall variability, wind vector, and month in the northern Bay of Bengal. Hilsa catch during the monsoon season of three consecutive years (2013–2015) was analyzed by using a Generalized Least Square model with lunar phase and monsoon months (June–September) as categorical variables and wind direction as circular variable. Significantly higher Hilsa catch was observed during the waning crescent and waxing gibbous lunar phases and during easterly winds. There was no significant effect of wind velocity. Daily rainfall was significantly correlated with Hilsa migration toward the estuary. Among the four monsoon months, September exhibited the most positive significant correlation with the Hilsa CPUE. Model predicted CPUE underestimated the actual CPUE in 2016 by 20%.  相似文献

Pituitary-ovarian function was analyzed in a strain of miniature swine previously shown to produce a low ovulation rate resulting in the formation of only 8.6 corpora lutea (CL)/animal. Five multiparous (M) and four nulliparous (N) miniature pigs with a mean inbreeding coefficient of .39 were monitored for estrous behavior through four consecutive estrous cycles. Daily blood samples were collected from 5 d before to 5 d after the onset of the second, third and fourth estrus and at 48-h intervals during the remainder of the second and third estrous cycle. Laparoscopy was used to examine the ovaries 1 and 5 d after onset of the third estrus and 2 d after the beginning of the fourth estrus. For the entire group, temporal fluctuations among serum estradiol-17 beta, luteinizing hormone (LH) and progesterone concentrations and sexual behavior were similar to previously published data in standard swine breeds. Although the mean lengths of the estrous cycle were not different (P greater than .05) between parity subgroups (M, 23 +/- 1.3 vs N, 22 +/- .7 d), multiparous pigs were in estrus longer (P less than .05) than nulliparous females (M, 3.7 +/- .2 vs N, 2.2 +/- .4 d). Parity subgroups were similar with respect to the mean number of follicles forming CL (M, 8.8 +/- .7 vs N, 9.2 +/- .2). Although an average of 6.2 +/- 2.1 CL had formed by 24-h after onset of estrus in the nulliparous subgroup, no CL were detected in the multiparous subgroup at this time.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Blister blight (causal agent, Exobasidium vexans) is an economically devastating disease of tea (Camellia sinensis). To determine what metabolite changes occur with tea blister that could be linked to disease progression, metabolomic approaches were used on E. vexans infected tea from a Darjeeling (India) plantation. Samples were classified according to disease phenotypes, i.e. either healthy or at one of three stages of disease progression. Initial metabolite fingerprinting using Fourier transform infrared (FTIR) spectroscopy indicated that metabolite changes could be related to disease stage. Electrospray ionization mass spectrometry (ESI‐MS) highlighted caffeine and flavonoid metabolism changes as disease progressed. High‐performance liquid chromatography (HPLC) with online photodiode array detection and electrospray ionization‐tandem mass spectrometry (HPLC‐PDA‐ESI/MSn) was used to characterize the caffeine, flavan‐3‐ol, flavone and flavonol profiles. There were increases in quercetin and kaempferol glucosides, kaempferol triglycosides and some catechin‐class antioxidants, but also substantial reductions in apigenin and myricetin glycosides and, particularly, caffeine as disease progressed. The content of important defence hormones, salicylic acid and jasmonic acid, was also reduced in blister blight diseased samples. Thus, E. vexans infections perturb defence signalling and reduce many potentially antimicrobial compounds, such as caffeine, to aid disease progression.  相似文献

High consumer preference together with its polyculture potential has undoubtedly driven Rohu (Labeo rohita) and Catla (Catla catla) to top the list of the most preferred fishes among the Indian major carps. Commonly found in these fishes are hybrids that can be natural or man-made. Increasing emphasis on biodiversity issues has necessitated proper stock management of these through molecular genetics techniques. Also with few morphological differences that can be used to differentiate wild types and hybrids properly, the problem demands a straightforward molecular approach. Here, we report a simple PCR-based technique that can differentiate the hybrid variety from wild types easily using three different microsatellite markers. Three sets of primers were used to amplify three different microsatellite markers from the genomic DNA isolated from pectoral fins. When the PCR products using all three primer sets were analyzed, ‘hybrid–Rohu’ could be distinguished from wild types. Whereas the hybrid–Rohu DNA yielded specific PCR products with all three primer pairs, only two PCR products were obtained either from wild-type Catla DNA (by primer sets 1 and 2) or from wild-type Rohu DNA (by primer sets 1 and 3). This study clearly demonstrates that a simple PCR-based technique will help the fish breeders and hatcheries to identify and differentiate suspected hybrid–Rohu carp from the wild types within a few hours.  相似文献
