This study investigated the effects of atmospheric moisture on conidia development in cucurbit powdery mildew fungus (Podosphaera xanthii) through host-plant responses. Cucumber (Cucumis sativus L.) seedlings were grown under a high vapour pressure deficit (VPD) (3.8 kPa) or a low VPD (0.4 kPa). When the cotyledons had expanded, spores of P. xanthii were inoculated onto the adaxial surface. Inoculated seedlings for both treatments were then placed in a growth chamber maintained at a VPD of 2.1 kPa. The density of visible P. xanthii colonies on the high-VPD-acclimated cotyledons was 0.46 to 0.85 times that of the low-VPD-acclimated cotyledons 7 days after inoculation. It is likely the post-germination behaviour of P. xanthii such as the infection and consequent hyphal development was affected because spore germination did not differ between the treatments. The percentage of adaxial epidermal leaf cells with haustoria was also lower in the seedlings which had acclimated to a higher VPD. The high-VPD-acclimated cotyledons were thicker and had greater dry mass per area. The water potential of cotyledons did not differ between the treatments, although the stomatal conductance of high-VPD-acclimated cotyledons was lower than that of low-VPD-acclimated cotyledons. From these results, we conclude that the inhibition of P. xanthii conidia development on high-VPD-acclimated cotyledons was mainly caused by changes in leaf morphological properties.  相似文献

归一化植被指数(NDVI)时间序列数据拟合目的是降低时序数据的噪声水平,重建高质量的NDVI时序数据,有利于多种参数反演和信息提取。针对国际上普遍应用的两种NDVI时间序列数据拟合方法,即Savitzky-Golay滤波法和非对称性高斯函数拟合法,该文在介绍两种方法基本概念的基础上,利用直接比较法和间接比较法在中国对两种拟合方法进行了比较分析。结果表明,Savitzky-Golay滤波法和非对称性高斯函数拟合法的拟合效果总体上一致,但二者之间还是存在区域差异性,这种区域差异与两种方法的自身特点和中国区域自然条件紧密相关。不同数据拟合方法的比较研究可以弄清每种方法的优缺点和区域适宜性,有助于研究人员针对不同研究目的和研究区域选择适宜的NDVI数据拟合方法,减少遥感数据处理中的误差,提高研究精度。  相似文献

Phospholipid fatty acid (PLFA) analysis and amino sugars analysis were compared with respect to their capacity to reliably indicate microbial biomass and gross community structure (e.g. fungi to bacteria ratio). We sampled three sets of soils beneath dominant canopy-tree species in an old growth forest and extracted them using both PLFA and amino sugars procedures. Amino sugars data had a lower coefficient of variation among sample replicates and therefore may be better for quantitative application compared to PLFA data. However, when considering microbial community structure, PLFA had an advantage over amino sugars due to its more straightforward interpretation. Finally, because each biomarker derives from different portions of the microbial biomass, a conversion factor between cell membrane PLFA and cell wall amino sugars may be valuable in understanding soil carbon cycling.  相似文献

Gizzard erosion in broiler chickens was investigated at 18 slaughterhouses in Japan. The condition was observed in 13 of them, and adenoviral gizzard erosion (AGE) was diagnosed histologically, immunohistochemically and virologically in the eroded gizzards from nine of these 13. The antigen-positive intranuclear inclusion body of group 1 fowl adenovirus was observed in the epithelial cells of the affected gizzards, and fowl adenoviruses were isolated from the lesions. In two of the slaughterhouses the total weights of the gizzards disposed of in three years were 3590 kg (0.40 per cent of the gizzards inspected) and 2880 kg (0.19 per cent). Sixteen of the 19 outbreaks of gizzard erosion on 15 farms that were confirmed in three of the slaughterhouses, including the previous two slaughterhouses, were diagnosed as AGE, and the condition was suspected in the other three outbreaks. Most of the adenoviruses isolated were identified as serotype-1 by PCR-restriction fragment length polymorphism. No apparent clinical signs were observed in any of the affected flocks.  相似文献

Oligodendrocytes and myelin in the corpus callosum of black tremor and normal hamsters aged over 1.5 years were ultrastructurally examined to determine the myelination index (ratio of myelin thickness/diameter of axon), percentage of naked axons, and proportions of oligodendroglial subtypes (light, medium and dark). The mutant hamsters were remarkably hypomyelinated, with a low myelination index and a high proportion of naked axons, and high proportions of the dark subtypes. Serum concentrations of thyroid hormones (T3 and T4) in 6-week-old mutant hamsters were 2-fold (T3) to 3-fold (T4) higher than those of age-matched normal animals. However, in the aged animals (over 1.5 years old) only T4 levels of the mutant hamsters were higher in the mutant than normal hamsters. The black tremor hamsters were hypomyelinated throughout their life and high serum level of thyroid hormones might have played a role in the hypomyelination.  相似文献

Introduction:  It has been reported that 40–50% of canine osteosarcoma cases have p53 mutations. The p53 tumor supressor gene plays a central role in cell cycle regulation and induction of apoptosis. We previously showed that adenoviral vector expressing canine P53 (AxCA‐cp53) inhibited growth of cultured canine osteosarcoma cell lines. Here, we evaluated anti‐tumor effect of adenovirus‐mediated p53 gene therapy on the growth of canine osteosarcomas transplanted into nude mice.
Methods:  Nine nude mice were subcutaneously injected with cells of a canine osteosarcoma cell line (POS) having p53 gene mutation. The transplanted tumors formed into nude mice were injected with AxCA‐cp53, AxCA‐LacZ (adenovirus vector expressing LacZ) or PBS (3 mice each) 7 times during 15 days. Tumor sizes were measured every 3 days for 27 days after injection with the adenovirus vectors. Expression efficiency of the adenovirus‐mediated gene transfer was examined by X‐gal staining and P53 immunostaining. Effects of the P53 expression on cell cycle control were examined by RT‐PCR for expression of p21 gene downstream of P53.
Results:  Significant differences in the tumor size was observed between the transplanted osteosarcoma tissues injected with AxCA‐cp53 and those injected with AxCA‐LacZ or PBS. Expressions of LacZ and P53 were confirmed at the injection sites of the tumors. Moreover, p21 mRNA expression was shown to be induced in the AxCA‐cp53‐injected tumors, indicating the funciton of P53 to induce cell cycle arrest.
Conclusions:  Adenoviral vector expressing canine P53 inhibited the growth of canine ostersarcoma transplanted into nude mice.  相似文献

Glucose delivery and uptake by the mammary gland is a rate‐limiting step in milk synthesis. Insulin resistance is believed to increase throughout the body following the onset of lactation. To study glucose metabolism in peak‐, late‐, and non‐lactating cows we analyzed the expression of an adipokine, namely, adiponectin, decreased insulin resistance, leptin, and a novel insulin‐responsive glucose transporter (GLUT12) in the adipose tissue and mammary gland by using real‐time polymerase chain reaction. Our results demonstrated that the mRNA level of adiponectin in the adipose tissue was greater in non‐lactating cows than in peak‐lactating cows. In the adipose tissue, there were no significant differences in the abundance of GLUT12 mRNA between the peak‐, late‐, and non‐lactating cows. In contrast, in the mammary gland, the mRNA level of GLUT12 was greater in non‐lactating cows than in peak‐ and late‐lactating cows. In the adipose tissue, the mRNA level of leptin and peroxisome proliferator‐activated receptor gamma 2 (PPARγ2) was greater in non‐lactating cows than in peak‐lactating cows. The results of the present study suggest that in lactating cows adiponectin plays an important role in insulin resistance in the adipose tissue; in the mammary gland, GLUT12 expression is believed to be an important factor for insulin‐dependent glucose metabolism.  相似文献

This report describes the pathological characterizations of a rare case of necrosis of the femoral head that was spontaneous, bilateral, avascular and nontraumatic. A 14-month-old beagle dog was presented with pain in the hind limbs. At necropsy, the articular surface in the bilateral femoral head was markedly irregular. There were no gross abnormalities other than in the hip joints. Microscopically, a wide range of trabecular bone necrosis localized in the subchondral area was observed in both femoral heads. In the right femoral head, fibrosis and proliferative vessels were noted in the subchondral area. The articular cartilage was thickened irregularly, but there was no evidence of cartilage necrosis. The bone marrow adjacent to the affected area showed severe depression. In the metaphysis, atrophic bone marrow, but not bone necrosis, was observed. This was a rare case of spontaneous necrosis of the femoral head in an experimental beagle dog.  相似文献
