Melatonin affects diverse physiological functions through its receptor and plays an important role in the central nervous system. In the present study, we compared immunoreactivity patterns of arylalkylamine N-acetyltransferase (AANAT), an enzyme essential for melatonin synthesis, and melatonin receptor type 1B (MT2) in the spinal cord of young adult (2~3 years) and aged (10~12 years) beagle dogs using immunohistochemistry and Western blotting. AANAT-specific immunoreactivity was observed in the nuclei of spinal neurons, and was significantly increased in aged dog spinal neurons compared to young adult spinal neurons. MT2-specific immunoreactivity was found in the cytoplasm of spinal neurons, and was predominantly increased in the margin of the neuron cytoplasm in aged spinal cord compared to that in the young adult dogs. These increased levels of AANAT and MT2 immunoreactivity in aged spinal cord might be a feature of normal aging and associated with a feedback mechanism that compensates for decreased production of melatonin during aging.  相似文献

The present study demonstrates the effects of different muscle types and chiller ageing periods on the chemical composition, meat quality parameters, sensory characteristics and volatile compounds of Karean native cattle beed. Longissimus dorsi (LD) and Semitendinosus (ST) muscles aged for 7 days and 28 days were used. Moisture, cooking loss, total collagen and Warner‐Bratzler shear force (WBSF) values for the ST were higher than the LD muscle regardless of ageing period (P < 0.05). The LD muscle had higher intramuscular fat (IMF) (P < 0.05). Ageing for 28 days decreased WBSF values whereas it increased thiobarbituric acid of both muscles. Moreover, tenderness, juiciness and flavor scores were significantly higher for the LD muscle at both ageing periods. Increased ageing time improved tenderness of both muscles, and increased juiciness of the LD muscle, whereas there was decreased flavor score of ST muscle (P < 0.05). The majority of the volatile compounds formed from the oxidation of lipids showed differences between the two muscles. Ageing for 28 days increased in the amounts of many volatile compounds; however, the amounts of some important volatile compounds were decreased. These results clearly demonstrate that muscle type and ageing have a potential effect on meat quality, sensory characteristics and volatile profile.  相似文献

A multiplex PCR protocol was established to simultaneously detect major bacterial pathogens in olive flounder (Paralichthys olivaceus) including Edwardsiella (E.) tarda, Streptococcus (S.) parauberis, and S. iniae. The PCR assay was able to detect 0.01 ng of E. tarda, 0.1 ng of S. parauberis, and 1 ng of S. iniae genomic DNA. Furthermore, this technique was found to have high specificity when tested with related bacterial species. This method represents a cheaper, faster, and reliable alternative for identifying major bacterial pathogens in olive flounder, the most important farmed fish in Korea.  相似文献

Various somatic cell nuclear transfer (SCNT) techniques for mammalian species have been developed to adjust species-specific procedures to oocyte-associated differences among species. Species-specific SCNT protocols may result in different expression levels of developmentally important genes that may affect embryonic development and pregnancy. In the present study, porcine oocytes were treated with demecolcine that facilitated enucleation with protruding genetic material. Enucleation and donor cell injection were performed either simultaneously with a single pipette (simplified one-step SCNT; SONT) or separately with different pipettes (conventional two-step SCNT; CTNT) as the control procedure. After blastocysts from both groups were cultured in vitro, the expression levels of developmentally important genes (OCT4, NANOG, EOMES, CDX2, GLUT-1, PolyA, and HSP70) were analyzed by real-time quantitative polymerase chain reaction. Both the developmental rate according to blastocyst stage as well as the expression levels CDX2, EOMES, and HSP70 were elevated with SONT compared to CTNT. The genes with elevated expression are known to influence trophectoderm formation and heat stress-induced arrest. These results showed that our SONT technique improved the development of SCNT porcine embryos, and increased the expression of genes that are important for placental formation and stress-induced arrest.  相似文献

A 4‐week feeding trial was conducted to test the use of marine yeasts purified from seawater and sediments as a dietary source in cultivating a cladoceran, Moina macrocopa, a potential alternative live food for fish larvae. Optimal growth conditions of two yeast strains were obtained for NaCl concentration, pH and temperature. Moina macrocopa was cultivated using different diets: marine yeasts (Debaryomyces hansenii Yeast‐14 and Candida austromarina Yeast‐16) and a commercial diet (Erythrobacter sp. Sπ‐I). The essential amino acids of both the yeast strains were nearly as great as those in M. microcopa. Further, the yeast‐fed M. macrocopa had essential amino acid profiles similar to the documented values for rotifers and Artemia enriched in microalgae and commercial diets. Erythrobacter sp. Sπ‐I lacked n‐3 polyunsaturated fatty acids, 20:5n‐3 and 22:6n‐3, which were also low but detected in both yeasts. An increase in the 20:5n‐3 [eicosapentaenoic acid (EPA)] and 22:6n‐3 [docosahexaenoic acid (DHA)] levels, compared with the levels in yeast strains themselves, was more pronounced in the 22:6n‐3 level of Moina fed the C. austromarina, resulting in a high DHA:EPA ratio. When the Moina diets were switched, their δ13C values shifted gradually towards the values of the switched diets. Diet switch from Erythrobacter sp. Sπ‐I to C. austromarina Yeast‐16 resulted in a more rapid turnover of Moina tissue carbon than that in the inverse case. When fed a mixed diet, the δ13C values of Moina tissue approached the value of marine yeasts immediately. These temporal changes in the δ13C values of Moina tissue indicate the preferential ingestion of marine yeasts and a selective assimilation of the carbon originated from marine yeasts. These findings suggest that marine yeasts, particularly C. austromarina Yeast‐16, are highly available to mass cultures of M. macrocopa, providing better nutritional and dietary values than the commercial diet (Erythrobacter sp. This study investigated the effects of different types of diet and sediment on the growth parameters of juvenile tiger crab Orithyia sinica under laboratory conditions, focusing on survival, growth and moulting. In the first set of experiments, juvenile tiger crabs were reared with five different types of sediment, i.e., soft sand, coarse sand, soft sand mixed with coarse sand, mud or bare bottom. Soft sand resulted in not only the highest survival rate of 78% but also the best rate of growth and feed intake. Crabs reared with a mud substrate showed the worst results in these parameters. The intermoult period, however, was not affected by sediment types. In the second set of experiments, crabs were fed different types of diet to determine their effects on the same growth parameters. Diets were composed of different combinations with Manila clam (Ruditapes philippinarum), brine shrimp (Artemia nauplii), microencapsulated feeds and marine diatom (Chaetoceros gracilis). Survival and growth in crabs fed Manila clam as a diet component were significantly higher than in those on other diets. The highest survival and growth were achieved with a diet that included all four components. These results provide valuable information on the appropriate bottom substrates and diet types necessary for artificial mass culture of the tiger crab.  相似文献

Highly aligned ultrafine fibers of Poly (amide-co-imide) PAI (torlon)/Poly (trimellitic anhydride chloride-co-4, 4′-methylene dianiline) (PTACM) blends have been prepared by using mechano-electrospinning. Employing a mixed solvent system of DMSO and THF alongwith water coagulation bath as a medium, continuous fibers with improved mechanical properties have been obtained. The continuity of the fibers is strongly dependent on the solvent mixing ratio of DMSO and THF (6:4). Continuous fibers with the most uniform diameter are obtained when a 30 % of PAI and PTACM (1:1) resins by weight is used in the blending solution. The aligned fibers are further coated with silver nanoparticles using a one-step process by passing the electrospunned fibers through PEG solution and a silver precursor solution followed by reduction in a continuous process. The advantage of our method lies in a uniform silver coating on a single fiber that can be coated continuously on a larger length scale (~Km). The mechanical properties of these fibers are dramatically improved after their alignment. Better electrical conductivity is obtained for these fibers after they are coated with silver nanoparticles. The highly aligned silver decorated fibers utilizes a novel electroless continuous process using Polyethylene glycol (PEG), which shows good binding properties and can be used for various bio and electronic applications.  相似文献
