The persistence of two herbicides, simazine and terbuthylazine, and appearance of their principal dealkylated chloro-s-triazine metabolites have been studied in agricultural soil after the addition of urban sewage sludge as organic amendment. Both herbicides and metabolites were monitored during long-term laboratory incubation (140 days) and analyzed by gas chromatography with a nitrogen-phosphorus detector (GC-NPD). Residues were confirmed by gas chromatography with a mass selective detector (GC-MSD). A sonication microextraction method was used to extract the compounds. The organic amendments used were urban sewage sludge and the humic fraction of this sludge, to increase the organic matter content of the soil from 1% to 2%. For both compounds, simazine and terbuthylazine, the degradation began earlier in the amended soils. Simazine showed a higher dissipation rate than terbuthylazine, the percentage of the former at the end of the experiment being lower than 2% in all cases, while for terbuthylazine the corresponding percentage ranged from 5% to 46%. Organic amendment, mainly its humic fraction, caused a certain stabilization of terbuthylazine in the soil, but did not greatly influence the residual amount of simazine at the end of the experiment. The periodic aeration of the soil caused a greater degradation in the case of terbuthylazine. Only mono-deethylsimazine and deethylterbuthylazine were isolated from the soil during the time the experiment lasted, while the di-deethylated metabolite of simazine was not found.  相似文献

The relationship between the levels of cyanogenic compounds (amygdalin and prunasin) in kernels, leaves, and roots of 5 sweet-, 5 slightly bitter-, and 5 bitter-kernelled almond trees was determined. Variability was observed among the genotypes for these compounds. Prunasin was found only in the vegetative part (roots and leaves) for all genotypes tested. Amygdalin was detected only in the kernels, mainly in bitter genotypes. In general, bitter-kernelled genotypes had higher levels of prunasin in their roots than nonbitter ones, but the correlation between cyanogenic compounds in the different parts of plants was not high. While prunasin seems to be present in most almond roots (with a variable concentration) only bitter-kernelled genotypes are able to transform it into amygdalin in the kernel. Breeding for prunasin-based resistance to the buprestid beetle Capnodis tenebrionis L. is discussed.  相似文献

To investigate the presence of furosine in commercial samples of jams and fruit-based infant foods a simple method by ion-pair reversed-phase liquid chromatography is described. The yield of furosine during the hydrolysis with hydrochloric acid was optimized. The reproducibility in the repeatability and recovery of the method, expressed in relative standard deviation percentages, proved to be in the ranges of 4.1-8.3% and 1.0-4.4%, respectively. The recovery percentages of furosine varied between 86.7 and 95.3%. The obtained results support the suitability of the method. Furosine was detected in all studied samples. Although a high variability in the content of furosine was noticed, in general terms, the lowest levels of furosine were observed in samples of fruit-based infant foods and the highest were observed in jams of more than 60% sugar. These results could be due to different heat treatment, storage conditions, and/or differences in the values of water activity (a(w)) and amounts of sugar. The results obtained in the present paper point out the usefulness of furosine as an indicator of Maillard reaction for jams and fruit-based infant foods.  相似文献

In this work, four different fluorescent intercalating dyes are compared for the ultrasensitive CGE-LIF detection of DNA from transgenic maize in flours. The fluorescent intercalating dyes compared are YOPRO-1, SYBR-Green-I, Ethidium bromide (EthBr), and EnhanCE. For all the four dyes optimum concentrations are established, and efficient separations of DNA fragments ranging in size from 80 to 1000 bp are obtained. The comparative study demonstrates that SYBR-Green-I and YOPRO-1 provide better limits of detection (LODs) than EnhanCE or EthBr (i.e., LODs are, respectively, 700, 1000, 11300, and 97400 zmol, calculated for a 200-bp DNA fragment). Separations using YOPRO-1 are faster than those using SYBR-Green-I (30 min vs 47 min for the analysis of the 80-1000 bp DNA fragments). Also, separations using YOPRO-1 are more efficient than those using SYBR-Green-I (e.g., 2.4 x 10(6) plates/m vs 1.6 x 10(6) plates/m, respectively, calculated for the 200-bp fragment). Also, buffer depletion and cost per analysis are worse with SYBR-Green-I than with YOPRO-1. Therefore, YOPRO-1 was selected as the preferred intercalating dye. Using this fluorescent compound, analysis time reproducibility for the CGE-LIF separation of the DNA fragments is determined to be better than 1.7% (% RSD, n = 10) within the same day, and better than 1.9% (% RSD, n = 30) for three different days. Moreover, the fluorescence signal obtained using this dye is shown to vary linearly with the DNA concentration in the range studied, i.e., 1-500 ng/microL. It is demonstrated that by using this method 0.01% of transgenic maize can be detected in flour by direct injection of the PCR-amplified sample.  相似文献

This work has evaluated the effects of thermally dried (TDS) or composted (CDS) dewatered sewage sludge on β-glucosidase activity, total (TCH) and extractable (ECH) carbohydrate content, microbial biomass carbon and basal respiration of soils from limestone quarries under laboratory conditions. Two doses (low and high) of the dewatered sludge (DS) or of the respective TDS or CDS were applied to a clayey and a sandy soil, both coming from working quarries. The soil mixtures and the controls (soils with no added sludge) were incubated for 9 months at 25°C and 30% of field capacity. The addition of sludge increased all the studied soil parameters, and the increase depended on the amount of sludge. Except in the case of TCH and ECH, the enhancing effect decreased with time, but at the end of incubation, parameters of the treated soils were higher than those of the control. The rank order of the initial stimulating effect was soil–TDS ≥ soil–DS ≥ soil–CDS, and probably, this order depended on the proportion of stable organic matter, which was the lowest in the TDS. Values of metabolic quotient (qCO2) were higher at the lower dose, and they did not change during incubation in the CDS-treated soils. Both TCH and ECH were the parameters with the greatest significant sludge and dose effects. Basal respiration, microbial biomass carbon and β-glucosidase activity were the best measured parameters in distinguishing the long-term effects of the three sludge types over the soils.  相似文献

The purpose of the present study is to simulate concentrations of ozone and its precursors (NO and NO2) in an area under the influence of the Candiota coal-fired power station by applying numerical models. The photochemical simulations were conducted during two distinct periods: in summer (from 24 to 26 January 2003) and in winter (from 10 to 12 August, 2004). With the RAMS atmospheric model (Regional Atmospheric Modelling System) we obtained meteorological fields to initialize the CIT photochemical model. The emission data were based on the AP-42 and only compounds emitted during coal burning were considered. The simulated results revealed high ozone levels in warmer periods, being related to high temperatures and water vapor concentrations. The simulated concentrations are located far from the emitting source, favoring reaction and mixing time between precursors to form ozone. Simulated winds of small intensity favored ozone transport, mixing and accumulation in areas far from the emitting source.  相似文献

The aim of the present article is to show the possibilities of chemometric tools and the parallel factor analysis (PARAFAC) model, as well as to understand the complexities of the fluorescence emission-excitation matrix (EEM) of water-soluble organic matter (WSOM) extracted from sewage sludge samples obtained with different origins and stabilization procedures. The variation in the composition of WSOM in the different sewage sludge samples could be correlated with the conditions of stabilization; therefore, the use of fluorescence spectroscopy to monitor these changes can be a useful tool to optimize the management of this residual flux, avoiding environmental pollution. However, the “contour density” EEM maps obtained with fluorescence present continuous spectra with different peaks, valleys, and shoulders where manual peak picking can be complicated and subjective, especially in complex matrixes such as sewage sludge. With the PARAFAC algorithm, it is possible to resolve the emission and excitation spectra of the different fluorescence components present in the samples and their contribution to the total fluorescence. Also, the contribution (scores) of the different fluorophors can be coupled with another chemometric tool to provide an effective classification method using the stabilization conditions of sewage sludge as main criterion. In this study, a wide survey of sewage sludge samples (287) was characterized by fluorescence emission-excitation matrix. The final molecular composition of the sewage sludge samples was not related to the stabilization (aerobic vs anaerobic) treatments. However, a sewage sludge classification based on a combined spectroscopic-chemometric approach was obtained, which could be used for the optimization of sewage sludge use in agriculture, minimizing the environmental risks.  相似文献

Phosphorus (P) and zinc (Zn) are important determinants of plant productivity, particularly in the tropical grasslands of Brazil. Nutrient deficiency is one of the most important factors limiting plant productivity, decreasing photosynthesis efficiency and plant development. The present study investigates in Brachiaria brizantha (Hochst. ex A. Rich.) Stapf. cv. ‘Marandu’: 1) the gas exchange measurements; 2) the total leaf area development; and 3) the dry matter production due to P and Zn nutrition. Plants of B. brizantha cv. ‘Marandu’ were grown in nutrient solution under five rates of P (0.1, 0.6, 1.1, 1.6, and 2.1 mmol L?1) and five rates of Zn (0.00, 0.75, 1.5, 2.25, and 3.00 μmol L?1), in a fractioned factorial. Plants were harvested two times. Phosphorus supply increased carbon dioxide (CO2) assimilation and stomatal conductance, and decreased intercellular CO2. The interaction P rates x Zn rates were significant for the total leaf area variables and shoot dry matter in the second growth period. The nutrition of P and Zn interfered in the B. brizantha productivity by changing the grass photosynthesis and leaf area.  相似文献

Nitrogen (N) by form of nutrition, ammonium (NH4+) or nitrate (NO3?), affects metabolic and physiological processes of plants. In general, a high proportion of N in NH4+ form results in poor growth. Nonetheless, a number of species exhibit optimum growth when high levels of NH4+ are provided. In the present study, lisianthus [Eustoma grandiflorum (Raf.) Shinn] was grown in rockwool cultures and irrigated with nutrient solutions containing 15 mM N with varying proportions of NH4+ and NO3?. The results showed that an increase in NH4+-N form increased plant height, number of flowers and leaves, leaf area, and shoot, stem, and leaf dry weight. The proportion of NH4+ also affected leaf concentration of phosphorus, potassium (K), calcium (Ca), and magnesium (Mg), although leaf N concentration was unaffected. Potassium leaf concentration was higher when a low proportion of NH4+ was supplemented in the nutrient solution; however, plants exhibited a decrease in leaf K concentration and a decrease in leaf Ca as the proportion of NH4+-N increased. Shoot dry weight was higher with low leaf K whereas high leaf Ca was associated with high shoot dry weight. Net photosynthesis rate was higher in plants irrigated with solutions containing 75% of total N in NH4+ form than in those irrigated with solutions of 0 or 25%. The results suggest that lisianthus can tolerate high levels of NH4+, probably associated with a higher assimilation of Ca.  相似文献
