We investigated the effects of inoculation of Rhodobacter sphaeroides, Lactobacillus plantarum, and Saccharomyces cerevisiae on cucumber plant growth promotion and on the contents of plant hormones, amino acids, and mineral nutrients. We showed that treatment with all three bio-inoculants significantly increased the shoot length, root length, shoot fresh weight, shoot dry weight, and chlorophyll content, via secretion of indole acetic acid and/or organic acids. Inoculation with R. sphaeroides had more favorable effect on plant growth than did inoculation with L. plantarum or S. cerevisiae, by significantly enhancing the gibberellin and reducing the abscisic acid contents. The results of amino acid analysis revealed that inoculation with R. sphaeroides, L. plantarum, and S. cerevisiae generally increased the contents of 17 amino acids, namely, aspartic acid, threonine, serine, glutamic acid, glycine, alanine, cysteine, valine, methionine, isoleucine, leucine, tyrosine, phenylalanine, lysine, histidine, arginine, and proline. With the exception of cysteine, all these amino acids were present in higher concentrations in plants inoculated with R. sphaeroides than in control plants or in plants inoculated with L. plantarum and S. cerevisiae. Furthermore, inoculation with R. sphaeroides significantly increased the calcium, potassium, magnesium, and phosphate contents. Our results suggest that the use of R. sphaeroides, L. plantarum, and S. cerevisiae in agricultural fields can improve plant growth. Moreover, inoculation of cucumber plants with R. sphaeroides regulates plant functional metabolites, thereby promoting plant growth.  相似文献

Pot and field experiments were conducted on alkaline calcareous soils of Pakistan to determine the effect of premixing superphosphate (SUP) with farmyard manure (FYM) on the fraction of plant phosphorus derived from the 30P labelled SUP (y value). Premixing (SUP with FYM) before their addition to soil, markedly increased the uptake of SUP-P by plants. The premixed treatment also increased crop yield on phosphorus deficient soli. Optimum quantity of FYM required was twice that of SUP. Higher quantities of FYM had no additional effect on phosphorus availability from SUP.

The possible mechanism of this effect and the implications of these observations in phosphorus nutrition of plants have been discussed.  相似文献

The accumulation of total soluble and cell wall-bound phenolics and total soluble proteins in Zea mays plants exposed to drought stress and foliar spray of salicylic acid (SA) at 10?4?mol/L and 10?5?mol/L was investigated. Drought stress was imposed at the four-leaf stage for 10 days (30–35% field capacity). Dehydration of maize leaves was accompanied by the accumulation of both total soluble and cell wall-bound phenolics, reduction in leaf relative water content (LRWC), and shoot and root growth attributes. Foliar spraying of SA further augmented the content of total soluble and cell wall-bound phenolics and total soluble proteins content under drought stress. SA ameliorated the adverse effects of drought stress on LRWC, shoot fresh weight, shoot dry weight, root fresh weight, root dry weight, root length and root area. The accumulation of both soluble and cell wall-bound phenolics by foliar spray of SA may be a mechanism related to SA-induced drought stress tolerance in maize. It was concluded that foliar spraying of SA at 10?5?mol/L can be highly economical and effective for modifying the effects of drought stress on maize at the four-leaf stage.  相似文献

SUMMARY An Australian strain of Newcastle disease virus, was evaluated for use as a vaccine following its administration by drinking water, aerosol and spray to chickens at 1 and 21 days of age. Haemagglutination inhbition antibody was produced and persisted for 11 weeks. Aerosol vaccination induced higher levels of haemagglutination inhibition antibody than the other methods of vaccination. No respiratory disease was observed following vaccination. Chickens vaccinated by aerosol and spray were fully protected when challenged at 5, 7 and 11 weeks of age with virulent Newcastle disease virus. Mortality of 10 to 30 per cent was observed in chickens vaccinated by drinking water and intranasally following challenge.  相似文献

1. Seven 35-week-old Hubbard broiler breeder males were subjected to three semen collection frequencies either once every 2 d (48 h), daily (24 h) or twice daily (12 h). 2. Semen characteristics including motility, volume, concentration and sperm numbers per ejaculate were determined for each ejaculate. 3. Sperm motility was unaffected by collection interval, but semen volume was lower at 12 than 24h intervals. Sperm concentration was lower at 12 than 48h intervals. 4. At 24 and 48 h number of sperm per collection (1.7+/-0.2, 1.8+/-0.2 x 10(9)) were higher than at 12 h (1.2+/-0.1 x 10(9)). 5. The number of semen doses over a 6-d period increased linearly as the frequency of collection increased from once every 2 d to twice daily. 6. It is concluded that output was theoretically maximal at twice daily collection, but in practice not all cockerels may be able to maintain full performance with such a demanding regime.  相似文献

BACKGROUND: The ability to sensitively detect Vibrio cholera with PCR-ELISA method represents a considerable advancement over alternative more time-consuming methods for detection of this pathogen. The aim of this research is to evaluate the suitability of a PCR-enzyme-linked immunosorbent assay for sensitive and rapid detection of V. cholera O1. METHODS: The 398-bp sequence of a gene that codes for the cholera toxin B subunit was amplified by PCR. The digoxigenin-labeled amplified products were coated on microplates and detected by ELISA. The PCR product was also hybridized with biotin labelled probe and detected by ELISA using streptavidin. RESULTS AND CONCLUSION: The specificity of the PCR was determined using 10 bacterial strains and 50 samples from south Iran. The detection limit was 0.5 pg of the genomic DNA and five bacterial cells. Adaptation of PCR into PCR-ELISA assay format facilitates specific and sensitive detection and diagnosis of human cholera disease. We conclude that this PCR-ELISA is a diagnostic method that specifically detects toxin genes in V. cholera O1 strains. It is more rapid and less cumbersome than other diagnostic methods for detection of toxicity in these strains.  相似文献

Aeromonas hydrophila and Gyrodactylus cichlidarum are common pathogens that induce significant economic losses in farm‐reared Nile tilapia. Nowadays, the sudden appearance of fish mortalities was exaggerated due to mixed and multiple infections. During summer 2016, mass mortality among earthen pond‐farmed Nile tilapia was reported. Clinico‐pathological, bacteriological and parasitological examinations have been demonstrated. As well, the water quality parameters were assessed. The clinical and histopathological findings of the moribund and recently dead fish were characterized by generalized septicaemic signs. The water quality parameters were significantly elevated over the permissible levels, whereas there was an elevation in nitrite (0.04 mg/L), un‐ionized ammonia (0.8 mg/L), hydrogen sulphide levels (153.1 mg/L) and organic matter content (3.79 mg/L). A. hydrophila was identified based on phenotypic characterization, API 20E features and the homology of 16S rRNA gene sequence analysis. In addition, PCR data confirmed the presence of aerolysin (aerA) and haemolysin (hly) genes in the identified A. hydrophila isolates. Gene sequencing and phylogenetic analysis based on 16S rRNA sequence confirmed that A. hydrophila H/A (accession No. MN726928) of the present study displayed 98%–99% identity with the 16S rRNA gene of A. hydrophila. Furthermore, the monogenetic trematode, G. cichlidarum was identified in the wet mounts from the skin and gills of the examined fish with a high infestation rate. In this context, it was reported that the synergistic co‐infection of A. hydrophila and G. cichlidarum with deteriorated water quality parameters could induce exaggerated fish mortalities during hot weather.  相似文献

Background:

Enterohemorrhagic Escherichia coli (EHEC) O157:H7 is an infectious zoonotic pathogen causing human infections. These infections, in some cases, can lead to hemolytic uremic syndrome and its life-threatening complications and even death worldwide. The first intimate bacterial adhesion, intimin (I), with its own receptor translocated intimin receptor (Tir) and E. coli secreted protein A, acting as Tir conduit, are highly immunogenic proteins for vaccine development against E. coli O157:H7.

Methods:

A chimeric trivalent recombinant protein was previously found to be a suitable strategy for developing vaccines against E. coli O157:H7. In this study, the recombinant EIT (rEIT) was used to design a protective EHEC nasal nanovaccine. Chitosan and its water-soluble derivative, trimethylated chitosan (TMC), as muco-adhesive biopolymers, are good candidates for preparation of nanovaccines.  Using the electrospraying technique, as a novel method, we could obtain particles of rEIT loaded with chitosan and TMC on a nanometer scale. Mice were immunized with intranasal administration or intrapretoneal injection of rEIT.

Results:

The rEIT-specific immune responses (IgG and IgA) were measured by indirect ELISA. Only nasal administration of chitosan electrospray and TMC formulation produced significant secretion IgA. Intranasal administration of nanovaccine reduced the duration of bacterial fecal shedding on mice challenged with E. coli O157:H7.

Conclusion:

Since development of mucosal vaccines for the prevention of infectious diseases requires efficient antigen delivery; therefore, this research could be a new strategy for developing vaccine against E. coli O157:H7.Key Words: EnterohemorrhagicEscherichia coli, Nanoparticles, Intranasal vaccination  相似文献

This study identifies the potential use of the combined application of hardwood-derived biochar (BC) and phytohormone-producing endophytes for enhancing crop production under heavy metal stress. For this purpose, the endophyte Galactomyces geotrichum WLL1 was isolated from Trapa japonica inhabiting a Korean river whose rainfall catchment area included an abandoned zinc mine. Pyrolyzed BC derived from pine hardwoods was obtained commercially. Under growth chamber conditions, the combined or individual application of G. geotrichum WLL1 and BC (15 % w/w) significantly improved soybean growth with or without excess Zn (5,253.6 mg kg?1). The beneficial effect of treatments was observed in the low uptake of Zn by different plant tissues and its immobilization in the soil. Biochar and G. geotrichum WLL1 shared their beneficial properties synergistically and improved plant growth characteristics with or without Zn heavy metal stress (HMS). Further, intensive root colonization of G. geotrichum WLL1 in the presence of BC was observed irrespective of HMS. Most interestingly, BC and G. geotrichum WLL1 had a priming effect by inducing systemic resistance in soybeans as revealed by significantly large amounts of jasmonic acid. These properties of BC could be exploited under abiotic and biotic stress. Here, for the first time, the combined application of hardwood-derived BC and phytohormone-producing endophytes is recommended for environmentally friendly and cost-effective crop production under HMS.  相似文献

Cumulative infiltration into thesoil profile during constant on-off cycletimes of surge irrigation was estimatedusing surge-ring infiltrometer data. TheKostiakov equation was employed to modelthe cumulative infiltration behaviour andits reduction into the soil duringdifferent surges. The two-point techniquewas applied to determine the unknownparameters of the Kostiakov equation. Itwas concluded that the surging phenomenasubstantially reduced water movement andthe reduction in cumulative infiltrationvaries from 60 to 83% during first surgeand 60 to 27% for the subsequent surges. Asurge infiltration model was also developedby introducing a `surge factor' into theKostiakov equation to generate infiltrationdata for subsequent surges, using the firstsurge / continuous infiltration data.Results of the study revealed that theinfiltration data of the surge-ringinfiltrometer and that generated by surgeinfiltration model matched closely.  相似文献
