The genetic diversities of 4 species of subg. Lithocerasus (Prunus, Rosaceae) revealed by RAPD analysis

RAPD analysis was applied to reveal the genetic diversities of 4 speciesof subg. Lithocerasus within the genus Prunus using 40accessions representing the subgenera Prunophora, Amygdalus,Lithocerasus and Cerasus. The accession of subg. Lithocerasus are phenotypically similar to members of subgenus Cerasus but different with them in interspecific crossing tests and isozymeprofiles. Two major clusters, `Prunophora and Amygdalus group' and `Cerasus group' were constructed in the phenogram. We revealthat the examined 4 species of subg. Lithocerasus species; P.tomentosa Thunb., P. japonica Thunb., P. glandulosa Thunb.and P. besseyi Bailey, were genetically closer to the members ofsubgenera Prunophora and Amygdalus than to subg. Cerasus.     

Nucleotide-dependent single- to double-headed binding of kinesin

The motility of kinesin motors is explained by a "hand-over-hand" model in which two heads of kinesin alternately repeat single-headed and double-headed binding with a microtubule. To investigate the binding mode of kinesin at the key nucleotide states during adenosine 5'-triphosphate (ATP) hydrolysis, we measured the mechanical properties of a single kinesin-microtubule complex by applying an external load with optical tweezers. Both the unbinding force and the elastic modulus in solutions containing AMP-PNP (an ATP analog) were twice the value of those in nucleotide-free solution or in the presence of both AMP-PNP and adenosine 5'-diphosphate. Thus, kinesin binds through two heads in the former and one head in the latter two states, which supports a major prediction of the hand-over-hand model.     

Oxygen isotopic compositions of asteroidal materials returned from Itokawa by the Hayabusa mission

Meteorite studies suggest that each solar system object has a unique oxygen isotopic composition. Chondrites, the most primitive of meteorites, have been believed to be derived from asteroids, but oxygen isotopic compositions of asteroids themselves have not been established. We measured, using secondary ion mass spectrometry, oxygen isotopic compositions of rock particles from asteroid 25143 Itokawa returned by the Hayabusa spacecraft. Compositions of the particles are depleted in (16)O relative to terrestrial materials and indicate that Itokawa, an S-type asteroid, is one of the sources of the LL or L group of equilibrated ordinary chondrites. This is a direct oxygen-isotope link between chondrites and their parent asteroid.     

Snowcover Components in Northern Japan

Snowcover surveys were carried out in Hokkaido, northern Japan. Snowpack samples were collected using a steel pipe at 80 sites in 1988, 69 sites in 1992, and 66 sites in 1996 and 2000 during the time when the water equivalents of the snowcover are the greatest. Spatial distribution maps of water equivalents and of the concentrations and the accumulated amounts of hydrogen, non-sea-salt sulfate and non-sea-salt calcium ion of the snowcover drawn by Kriging method are discussed. The distribution maps show that water equivalents and the accumulated amounts of hydrogen and non-sea-salt sulfate in all the sample years were largest in the Japan Sea region, as were the ion concentrations of hydrogen and non-sea-salt sulfate. It was estimated this was caused by air pollutants transported from a great distance. The ratio of areas below pH 5.0 increased temporally from 1988 (20%) to 1992, 1996 and 2000 (66–80%). This trend was estimated to be affected by a decline of non-sea-salt calcium concentrations derived from alkaline road dust. In addition, the effect of snowcover components was assessed by comparison between the acid loads of the snowcover and the acid neutralizing capacity of lake water. We found that lakes in the middle of the Japan Sea coast have the potential to be acidified by snowmelt.     

Subepithelial elastic fibers in the bovine gastric mucosa

The arrangement of subepithelial elastic fibers in the bovine gastric mucosa was studied by light and electron microscopy, with special reference to the elastic architecture of the cornified papillae. A common feature of their arrangement was that the fibers, derived from the elastic plexus in the deep layer of the lamina propria, extend perpendicularly toward the epithelium giving off the slender branches that end at the epithelial basement membrane. The connecting branches were observed electron microscopically in the form of oxytalan or slender elaunin fibers. The high frequency of fibers was found in the mucosa of the upper part of the ruminal atrium, the ruminal pillars, the reticular cristae and two kinds of cornified unguiculiform and omasal papillae. Each papilla consisted of the core of three-dimensional elastic network and numerous connecting-twigs arising from the superficial points of the network. The mucosae of the ruminal sacs, the floor of the reticular cells and the abomasum except the pyloric part were relatively poor in elastic fibers. These results suggest that the variation in fiber-arrangement may be relevant to the condition of mechanical stress inflicting on the gastric innersurface.     

Induction of superovulation by immunoneutralization of endogenous inhibin in immature rats

The effects of passive immunoneutralization of endogenous inhibin on ovulation rate in immature rats were investigated. Efficiency of superovulation on production of fertilized oocytes was compared between the inhibin antiserum (inhibin-AS) and equine chorionic gonadotropin (eCG) protocols. Immature female Wistar strain rats were superovulated with a single injection of 100-200 microl inhibin-AS, with and without an injection of human chorionic gonadotropin (hCG). A total of 77.8% of the 26-30-day-old rats treated with a single injection of 100-200 microl inhibin-AS ovulated 72 h after treatment, while rats given normal goat serum (NGS; 200 microl) did not ovulate. At 28 days of age, all of the inhibin-AS treated rats ovulated when additional hCG treatment was given, whereas the number of ovulated oocytes was not affected. The number of ovulated oocytes in the inhibin-AS-hCG treated groups was significantly higher than that of the NGS-hCG treated group. In addition, plasma concentrations of FSH in the inhibin-AS-hCG treated group significantly increased compared with the NGS treated group. While the percentage of mated rats in the 200 microl inhibin-AS-hCG treated group was significantly lower than that of the 15 IU eCG-hCG treated group, the fertilization rate was comparable between the two groups. The number of fertilized oocytes in the 200 microl inhibin-AS-hCG treated group was significantly higher in comparison with the 15 IU eCG-hCG treated group. These results suggest that immunoneutralization of endogenous inhibin could be a reliable method for induction of superovulation to collect a large number of normally fertilized oocytes in immature rats.     

Identification of bovine dendritic cell phenotype from bovine peripheral blood

Dendritic cells (DCs) are professional antigen presenting cells, which initiate primary immune responses and also play an important role in the generation of peripheral tolerance. There is no reliable method established for the isolation of bovine peripheral blood DCs, and furthermore, the phenotypes and the functions of bovine DCs are still not fully clear. In the present study, we have attempted to identify bovine peripheral blood DCs by negative-selection. In bovine peripheral blood mononuclear cells (PBMC), we have newly characterized the phenotype of DCs, which is CD11c+/CD172a+. These cells display features of myeloid type DCs. In the thymic medulla, CD11c+/CD172a+ cells were also present and CD1+/CD172a+ cells were additionally detected as a population of DCs. The data suggest that one of the bovine DCs phenotypes from PBMC is derived from myeloid lineages lacking a CD1 molecule, which then drift to several tissues, and that they then may express a CD1 molecule upon their functional differentiation.
