Hypoxic conditions in various cancers are believed to relate with their
malignancy, and hypoxia inducible factor-1α (HIF-1α) has been shown to be a major
regulator of the response to low oxygen. In this study, we examined HIF-1α expression in
canine lymphoma using cell lines and clinical samples and found that these cells expressed
HIF-1α. Moreover, the HIF-1α inhibitors, echinomycin, YC-1 and 2-methoxyestradiol,
suppressed the proliferation of canine lymphoma cell lines. In a xenograft model using
NOD/scid mice, echinomycin treatment resulted in a dose-dependent regression of the tumor.
Our results suggest that HIF-1α contributes to the proliferation and/or survival of canine
lymphoma cells. Therefore, HIF-1α inhibitors may be potential agents to treat canine

The Spontaneously Diabetic Torii (SDT) fatty rat, a model for obese type 2 diabetes,
shows bone quantitative abnormalities, namely low bone mineral density (BMD). The
objective of this study was to evaluate bone morphological changes, in particular
identifying the bone qualitative abnormalities, in the SDT fatty rat. Male SDT fatty rats
showed increases in total trabecular area and trabecular number and decreases in
trabecular thickness in cancellous bones of the proximal tibia, indicating trabecular
miniaturization. The SDT fatty rat is useful for investigation of pathophysiological

We investigated changes in the predicted functions of the rumen bacterial community in Japanese Black beef cattle during fattening. Nine cattle were fed a high-concentrate diet during the early, middle, and late fattening stages consecutively (10–14, 15–22, and 23–30 months of age, respectively). The rumen fluid and solid samples collected at each stage were subjected to sequencing analyses. The sequencing results were clustered and classified into operational taxonomic units (OTUs). Representative sequences and a raw counting table for each OTU were submitted to the Piphillin website. The predicted functions were revealed by the Kyoto Encyclopedia of Genes and Genomes database as the ratio of the total sequence. In the early stage, “Biosynthesis of secondary metabolites” was significantly higher in the fluid fraction than in the solid fraction. “Two-component system” in the middle stage was significantly lower and “Purine metabolism” in the late stage was significantly higher in the fluid fraction than those in the solid fraction. The fluid fraction was significantly correlated with acetic acid, propionic acid, and bacterial metabolism, such as “Biosynthesis of secondary metabolites” and “Sugar metabolism.” Moreover, the solid fraction was correlated with “Purine metabolism” and “Biosynthesis of secondary metabolism”. These results suggest that the rumen bacterial community in Japanese Black beef cattle adapts to changes in rumen conditions by altering their functions in response to a long-term high-grain diet. 相似文献

Quantitative determination of cyproconazole by gas chromatography–mass spectrometry (GC/MS) analysis was investigated. The results suggest that cyproconazole determination is significantly affected by wood extracts. In the presence of methanol extracts of Japanese cedar (Cryptomeria japonica) heartwood, cyproconazole showed 50 % larger peak area than that without the extracts. This phenomenon whereby contaminants in the sample solution affect the intensity of the peak signal is known as the matrix effect. An investigation for eliminating the matrix effect revealed that adding sorbitol as an analyte protectant drastically increased the analyte peak intensity and successfully mitigated the matrix effect of the wood extracts. Adding hexaconazole as an internal standard was also useful in reducing the deviation of the data. The efficacy of a novel sample preparation method consisting of adding sorbitol and hexaconazole as analyte protectants and the internal standard, respectively, were applied to the GC/MS analysis of cyproconazole in the presence of wood extracts of the other nine species. Effect of temperature and cell density on ethanol fermentation by a thermotolerant
aquatic yeast strain isolated from a hot spring environment 总被引:2,自引:0,他引:2
ABSTRACT: A thermotolerant, fermentative yeast strain named RND13 from a hot spring drainage was evaluated for its ethanol-producing ability at elevated temperatures at a high substrate concentration [15% (w/v) glucose] close to the level reflecting industrial practice. The RND13 was capable of utilizing glucose almost completely at 40°C with increasing inoculum size, producing ethanol up to 6.6% (w/v), which is comparable to levels (7.0–7.2%) at 30°C. The maximum rate of ethanol production by the RND13 was found to be 9.0 g/L per h at 40°C in an inoculum sized 5% (w/v). At 43°C, however, the RND13 could not utilize glucose to completion and showed a slight drop in the extent of produced ethanol [6.0% (w/v)]. Thus, the culture at 40°C with a 5% cell inoculum was considered to be the optimal condition for ethanol production at higher temperatures in terms of batch fermentation. In the phylogenetic analysis based on the small-subunit rDNA sequence, the strain was grouped together with both Candida glabrata and Kluyveromyces delphensis , which are relatively close to Saccharomyces cerevisiae . 相似文献

Exotoxins prepared from streptococcal bacteria of Seriola spp. origin were grown in fluid culture and fractionates of the supernatant used in toxicity studies in Seriola by oral and percutaneous exposure. Toxic fractions were then applied simultaneously with oral challenge with Streptococcus spp. bacteria.
