Partially purified thymus products were used to evaluate the maturation of T lymphocytes in immunodeficient mice. Three different bovine thymic extracts, designated thymic extracts A, B, and C, were successful in increasing the longevity of conventionally raised nude mice. Daily injection of bovine thymic extracts A, B, and C and mouse thymus extracts failed to mature a population of T lymphocytes and restore the capacity to reject heart allografts. Preincubation of normal syngeneic bone marrow with thymic extract B in vitro before injection into nude mice also failed to reconstitute the host's ability to reject heart grafts. The number of antibody plaque-forming cells of sheep red blood cells in lethally irradiated, bone marrow-reconstituted mice could be increased by preincubating the bone marrow cells with bovine thymic extract fraction B before injection followed by daily injections. A similar but less marked increase in plaque-forming cells was obtained by the daily injection of bovine thymic extract fraction C. Complete functional maturation of T lymphocytes in immunodeficient animals appears to require more than thymic extract stimulation of bone marrow cells or pre-T lymphocytes. 相似文献
SUMMARY A severe cattle mortality in which 132 out of 340 animals died on a property in southern Queensland was investigated. Clinical signs shown by affected animals included fever, inappetance, depression, lethargy, salivation, diarrhoea, ataxia, and ulceration of the oral cavity. The most common lesions seen at autopsy of 6 affected animals were ulceration of the tongue, gums, dental pad and buccal mucosa, linear ulceration of the caudal third of the oesophagus, mild catarrhal enteritis and necrosis of lymph nodes draining areas of ulceration. Bovine herpesvirus type 1 (BHV 1) was isolated from 3 out of 5 animals from which virus isolation was attempted. BHV 1 was recovered from oesophageal ulcers, retropharyngeal lymph nodes, blood clot, and swabs from ulcers in the oral cavity but not from spleen, liver or mesenteric lymph node. Serum neutralising (SN) antibody to BHV 1 was detected in 4 out of 12 affected animals in the second of paired serum samples but not in the first. Mucosal Disease (MD) virus was not recovered from any of 17 animals from which isolation was attempted but moderate MD SN titres, without a rise on paired sera, were detected in affected animals. Fever, depression, inappetance, ulceration of the upper alimentary tract, and adrenal necrosis were produced in 2 susceptible animals following inoculation with third passage cell culture fluid containing BHV 1. The immune response to modified live-virus bovine viral diarrhea (BVD) vaccine and infectious bovine rhinotracheitis (IBR) vaccine was examined in calves that had received passive maternal antibodies to these viruses. Blood serum samples from vaccinated and control (nonvaccinated) calves were examined for more than 1 year to determine the rate of decline of passive anti-BVD and anti-IBR antibodies and the effect that vaccination had on these antibody titers. The control calves lost their antibodies to BVD and IBR viruses at the rate of one half their remaining antibody titer every 21 days. Calves serologically responded to BVD vaccine at a time when maternal antibody titers remained between 1:96 and 1:20. However, animals did not seroconvert to the IBR vaccine until maternal antibodies had decreased and become undetectable. Evidence is presented to show that although passive immunity will inhibit IBR vaccination, priming for a secondary response will occur so that on subsequent vaccination, at a time when maternal antibodies have disappeared, the animals will respond anamnestically to IBR vaccination. 相似文献
The development of bovine leukemia virus (BLV) infection was studied in 14 noninfected young adult cattle exposed to 25 to 30 BLV-infected cows in an area of approximately 0.5 ha. Of 7 cattle (group 1) exposed beginning in July and August (midsummer) of 1976, 4 were infected by October, and all 7 by November (4 months' exposure). Of 7 cattle (group 2) exposed from February 1977 (midwinter), all remained negative for 3 months, and only 1 was positive after 6 months. By October 1977, however, 4 cattle in this group were infected, indicating that contact transmission of BLV is prevalent during the summer months. This, and the fact that BLV-infected lymphocytes were recovered from tabanids allowed to feed on a BLV-positive cow, supports the idea that bloodsucking insects play a major role in the spread of BLV. 相似文献
An indirect solid-phase microradioimmunoassay (IRIA) was developed for detection and quantitation of antibodies to pseudorabies virus (PRV) in swine serum. Qualitative results of the IRIA compared closely with results of the serum neutralization test (NT) and the microimmunodiffusion test (MIDT). The IRIA was more sensitive than the NT for detection of antibodies to PRV in swine serum. The IRIA result is expressed numerically. With the IRIA and NT, antibody to PRV was first detectable in 3 experimentally infected pigs at 9 days after inoculation. With MIDT, antibody was detected in the 3 experimentally infected pigs at 9 days after inoculation. With the MIDT, antibody was detected in the 3 experimentally infected pigs at 7, 8, and 9 days after inoculation. The IRIA results are obtainable within a few hours; the NT and MIDT require 48 hours for completion. 相似文献
1. A study of genetic parameters of egg mass based on the weights of all eggs laid, individual food consumption, body weight and age at first egg was made. Data from three populations differing in average egg weight were available over two production periods, from 20 to 40 and from 40 to 60 weeks of age, respectively.
2. Selection indexes designed to maximise income over food costs (IOFC) were used to evaluate the usefulness of food consumption data for the genetic improvement of laying flocks.
3. Given reliable estimates correlating food consumption to egg mass, body weights and age at first egg it is possible to construct highly efficient selection indexes for genetic improvement of IOFC without the direct use of food consumption data.
4. Selection for genetic gains in egg mass appears to be the most important factor for genetic improvement of IOFC. Correlated increases in egg weight were observed for two of the populations.
5. Body weights showed very small and inconsistent genetic responses under index selection suggesting that a reduction in body weight need not be necessary for more efficient egg production.
6. Indexes which showed greatest genetic improvement also resulted in an increase of food consumption. Thus genetic efficiency in egg laying flocks would be achieved by increasing egg mass through earlier sexual maturity, larger eggs and higher production rates with increased food consumption and only slight, if any, reduction in body size.
7. Estimates of heritabilities and genetic correlations indicate that food consumption measurements can be based on partial records; high heritabilities were found for egg mass and food consumption measurements in both early (20 to 40 weeks) as well as late (40 to 60 weeks) production periods. 相似文献
