Nitric oxide (NO*) synthesis is induced within many tumors. The timecourse of NO* synthesis was evaluated during intraperitoneal Meth A fibrosarcoma progression. While increasing macrophage recruitment into ascites was noted, inducible nitric oxide synthase (iNOS) antigen and function peaked between days 3-6 after tumor implantation. The capacity of cells to respond to LPS and IFNgamma stimulation was markedly depressed on day 9 and 11. Cellular proliferation correlated in an inverse fashion with levels of NO* synthesis. Electron paramagnetic resonance spectroscopy and nitrotyrosine immunostaining failed to show accumulation of characteristic target cell lesions induced by NO*. These findings lead us to conclude that NO* production was increasingly suppressed during Meth A tumor progression. Depression of NO* production did not correlate with levels of the inhibitory cytokines TGFbeta and IL-10, but could be partially overcome by addition of sepiapterin (a tetrahydrobiopterin prodrug). Thus, depletion of essential co-factors necessary for iNOS function may contribute to depressed NO* responses during cancer progression. 相似文献

Morphological data recorded from field trials using Citrullus lanatus germplasm collected in Namibia were used to analyse and compare the various morphotypes of this species. The experiment comprised wild types and local landraces as well as commercial cultivars. Cluster analysis supported the indigenous classification system used in Namibia, in which Citrullus types are distinguished based on gross morphology, ecology and usage and grouped into seed, cooking and fresh-eating (watermelon) types. Commercial watermelon cultivars formed a distinct cluster. Wide variation was found within the local types whereas the genetic basis of the commercial type appears to be narrow. Changes in the activity of the cell wall hydrolases – polygalacturonase (EC 3.2.1.15), pectinesterase (EC 3.2.1.11) and β-galactosidase (EC 3.2.1.23) – have been investigated following wounding of tomato fruit pericarp tissue (Lycopersicon esculentum cv. Ailsa Craig). In ripening fruit wounding appears to arrest the further synthesis of polygalacturonase. β-Galactosidase synthesis may also have been arrested in ripening fruit. The level of pectinesterase declined over the first 24 h following harvest, and since this was apparent in both wounded and unwounded tissue may be related to a harvest, rather than a wounding effect. There was a recovery of activity in intact fruit by 48 h after harvest but this seems to be impaired in wounded tissue. In the case of pectinesterase, this observation was extended to examine the changes in isoform profile and it appeared that the decline of this enzyme may be associated with the reduction of one specific isoform — PE2. In contrast to ripening fruit, wounding of fruit at the fully ripe stage appears to have no significant effects on the activities of any of these three enzymes. 相似文献

Clubroot caused by Plasmodiophora brassicae is an emerging threat to canola (Brassica napus) production in western Canada, and a serious disease on crucifer vegetable crops in eastern Canada. In this study, seven biological control agents and two fungicides were evaluated as soil drenches or seed treatments for control of clubroot. Under growth cabinet conditions, a soil‐drench application of formulated biocontrol agents Bacillus subtilis and Gliocladium catenulatum reduced clubroot severity by more than 80% relative to pathogen‐inoculated controls on a highly susceptible canola cultivar. This efficacy was similar to that of the fungicides fluazinam and cyazofamid. Under high disease pressure in greenhouse conditions, the biocontrol agents were less effective than the fungicides. Additionally, all of the treatments delivered as a seed coating were less effective than the soil drench. In field trials conducted in 2009, different treatments consisting of a commercial formulation of B. subtilis, G. catenulatum, fluazinam or cyazofamid were applied as an in‐furrow drench at 500 L ha?1 water volume to one susceptible and one resistant cultivar at two sites seeded to canola in Alberta and one site of Chinese cabbage in Ontario. There was no substantial impact on the susceptible canola cultivar, but all of the treatments reduced clubroot on the susceptible cultivar of Chinese cabbage, lowering disease severity by 54–84%. There was a period of 4 weeks without rain after the canola was seeded, which likely contributed to the low treatment efficacy on canola. Under growth cabinet conditions, fluazinam and B. subtilis products became substantially less effective after 2 weeks in a dry soil, but cyazofamid retained its efficacy for at least 4 weeks. 相似文献

Historical data on the distribution and status of the purple gallinule (Porphyrio porphyrio) in Portugal were collected by bibliographic research, from museum catalogues and from questionnaires to wetland managers, ornithologists and birdwatchers. The population suffered severe decline and range contraction at the end of the 19th century and most of the 20th century, due mainly to habitat loss and degradation and excessive hunting. However, the population has begun to recover in the last decade mainly through direct protection of the species and of the remaining habitat, together with creation of artificial wetlands and a reintroduction programme, and probably also due to immigration from Spain. The population size has increased about 10-fold and the range has expanded to most of its former area. The estimated Portuguese breeding population in 2002 was 49-67 pairs. Densities varied between 0.05 and 7 individuals/ha in recently colonized areas and stable (saturated) areas, respectively. Although the population is recovering, its small size, high fragmentation and several natural and anthropogenic threats lead us to consider it still a species of major concern in Portugal. 相似文献
