This study evaluated the Mycobacterium tuberculosis protein antigen MPT-51, the trimeric antigen 85 (Ag85) complex, and Bacillus Calmette-Guérin (BCG) in an indirect ELISA to diagnose bovine tuberculosis (TB) from serum samples. Serum was collected from 208 intra-dermal tuberculin test (ITT)-positive and 54 ITT-negative animals from a region where bovine TB is endemic. Using the Ag85 and BCG antigens, the indirect ELISA was able to discriminate ITT-positive from ITT-negative animals. This level of discrimination was not achieved when using the MPT-51 antigen. The highest sensitivity (Se) and specificity (Sp) of the test was found when BCG was used (Se, 82%; Sp, 91%). Further work in different epidemiological settings and with larger numbers of animals will be required to validate these findings.  相似文献

Background, Aims and Scope   Contaminated land is a high priority environmental problem in most of Europe and North-America. Sweden is no exception and generic guideline values have been developed for the initial assessment, but site-specific assessments are also needed. The generic guideline values are not applicable when the exposure conditions are different from the typical Swedish conditions or when the site contains a particularly sensitive ecosystem. The Swedish guideline values have, like in many other countries, been set by using deterministic point estimates for all variables and constants in the used multimedia model. The same approach is common also for site-specific assessments, and a limitation is that it fails to quantify variability and uncertainty. Probabilistic risk assessment provided a method to deal with this problem. Variability and uncertainty in the input parameters (variables or constants) are described by probability distributions, and likewise the output (risk or exposure) is presented as a probability distribution. A substantial number of probabilistic risk assessments for contaminated land at sites in North America, Europe and Asia have been published. However, an extensive review of the literature did not identify any study where probabilistic risk assessment was applied to a site contaminated by an iron or steel industry. Here we will describe such a case, where we have compared a deterministic point estimate with a probabilistic risk assessment for six elements and benzo[a]pyrene. Methods   The site had different metallurgical plants in operation for more than 100 years. Most parts of the steel mill were closed by the mid 1980s, and today the site is used by small-sized enterprises. The soil is contaminated with metals from the previous industrial operations. The present owner plans to develop the site and has therefore initiated extensive investigations of soil contamination. Sixty-two soil samples collected between 1997 and 2000 provided a good coverage of the whole site, and were analyzed for the content of different elements and polycyclic aromatic hydrocarbons (PAH). The exposure assessments were focused on six elements with high concentrations compared to the generic guideline values; arsenic (As), lead (Pb), cadmium (Cd), chromium (Cr), copper (Cu) and zinc (Zn). In addition, benzo[a]pyrene was included due to the high toxicity and comparatively high concentrations. Variability and uncertainty were characterized in a Monte Carlo simulation of exposures (10 000 iterations), and the exposures were evaluated with two land use scenarios; less sensitive use and sensitive use. Results and Discussion   The deterministic point estimates and the probabilistic estimates of the 95th percentile are in approximately the same ranges in the scenario of less sensitive land use. It is only the exposure for arsenic that is slightly above the toxicological reference value (TRV) in the deterministic assessment. In the probabilistic assessment, the exposure for all elements is below the TRV. The results for sensitive land use are applicable to a scenario where the site is developed for general housing. The deterministic point estimates and the probabilistic estimates of the 95th percentile are also here in approximately the same ranges, but the exposure exceeds the TRV for arsenic, cadmium and lead. Drinking water, vegetables grown on site and soil ingestion are the major exposure pathways for this scenario. In this assessment, the estimated intake distributions are applicable to a randomly selected individual. The probability distributions used here to characterize the different soil parameters are typically representing both variability and uncertainty, and the same is true the majority of the exposure variables. We therefore decided not to attempt to separate variability and uncertainty at this stage, but with additional data from a more in-depth site investigation it might be possible to achieve this. Conclusions and Outlook   To the best of our knowledge, this study is the first report on a probabilistic risk assessment on a former iron and steel works site. The materials handled by this industry were less toxic than for many other metallurgical operations, but contaminants may still severely limit the options for future land use. This case study shows that probabilistic exposure estimates for a set of soil contaminants can be quite similar to deterministic point estimates. The main difference is instead to be found in the additional information obtained with the probabilistic assessment. The sensitivity analyses show pathways and input variables that contribute most to variations in the total intake of each contaminant, e.g. dermal contact and ingestion of soil, vegetables and drinking water. This information can be used both in the planning of future land use and for active measures to reduce current exposure. The probabilistic assessment also provides information on the magnitude of exposure and the margin of safety. This information may facilitate risk communication between decision-makers and stakeholders. The presentation of results from probabilistic risk assessments is only briefly discussed in the literature and here we see a need for research and opportunities for enhancement. The choice of data analytical tools may then be of importance, since more complex multimedia models are rather difficult to decipher when implemented within traditional spreadsheet software. Semen collection has an essential role in the initial bacterial load in boar ejaculates and extended semen. The study aimed to explore the efficacy of an adjusted penis fixation in a semi-automatic collection system on reducing bacterial contamination of ejaculates in two-boar studs with different scenarios. Historically, stud A had low levels of bacterial load in raw semen, while stud B had a high level of contamination. A total of 56 mature boars had their semen collected using two methods of penis fixation: (a) Traditional: The penis was fixed directly with the artificial cervix and transferred to the adjustable clamp; (b) Adjusted: The fixation was performed with one gloved-hand, and after exteriorization, the penis was gripped using the artificial cervix with the other gloved-hand and transferred to the adjustable clamp. The bacterial load (p = .0045) and the occurrence of ejaculates >231 CFU/ml (p = .0101) were reduced in the Adjusted compared to the Traditional method. Bacterial load was reduced when using the Adjusted method in stud B (p = .0011), which showed a greater occurrence of critical factors for bacterial contamination (p ≤ .0034). The Adjusted method reduced the occurrence of ejaculates >231 CFU/ml when the preputial ostium was dirty (p = .016) and the duration of semen collection was >7 min (p = .022) compared to the Traditional method. In conclusion, the Adjusted penis fixation was efficient in reducing bacterial load of ejaculates, mainly in boar stud B, which had high contamination challenges.  相似文献

Background

Measurement of salivary cortisol has been used extensively as a non-invasive alternative to blood sampling to assess adrenal activity in ruminants. However, there is evidence suggesting a considerable delay in the transfer of cortisol from plasma into saliva. Previous studies in cattle have used long sampling intervals making it difficult to characterise the relationship between plasma and salivary cortisol (PLCort and SACort, respectively) concentrations at different time points and determine whether or not such a time lag exist in large ruminants. Therefore, the objective of this study was to characterise the relationship between plasma and salivary cortisol and determine if there is a significant time lag between reaching peak cortisol concentrations in plasma and saliva across a 4.25 h time-period, using short sampling intervals of 10–15 min, following social separation in dairy cattle.Five cows were separated from their calves at 4 days after calving, and six calves were separated from a group of four peers at 8 weeks of age. Following separation, the animals were moved to an unfamiliar surrounding where they could not see their calves or pen mates. The animals were catheterised with indwelling jugular catheters 1 day before sampling. Blood and saliva samples were obtained simultaneously before and after separation.

Results

In response to the stressors, PLCort and SACort increased reaching peak concentrations 10 and 20 min after separation, respectively. This suggested a 10 min time lag between peak cortisol concentrations in plasma and saliva, which was further confirmed with a time-series analysis. Considering the 10 min time lag, SACort was strongly correlated with PLCort (P < 0.0001).

Conclusions

Salivary cortisol correlates well with plasma cortisol and is a good indicator of the time-dependent variations in cortisol concentrations in plasma following acute stress. However, there is a time lag to reach peak cortisol concentrations in saliva compared to those in plasma, which should be considered when saliva samples are used as the only measure of hypothalamic-pituitary-adrenal axis response to stress in cattle.  相似文献

The degree of hydrolysis (DH) of the ingredients was evaluated for Centropomus undecimalis juveniles. The in vitro experiment included wheat gluten (WG), corn gluten (CG), soybean meal (SBM) and pretreated phytase (SBM + phytase), soy protein concentrate (SPC), canola meal (CAN) and pretreated phytase (CAN + phytase), poultry byproduct meal (PBPM), chicken meal (CHKM), dried whey (DW), Pota meal (PM: mix of giant squid, fish soluble protein concentrate, crustacean meal, and fish oil), and Protiblend (PTB: aquatic and render mix). The highest acidic DH occurred with PTB (0.38 ± 0.06), CHKM (0.33 ± 0.3), and PBPM (0.25 ± 0.03). In the alkaline condition, PTB (1.6 ± 0.17 and 0.98 ± 0.05 for pyloric caeca and intestine, respectively) and CG (1.04 ± 0.4 and 0.75 ± 0.2 for caeca and intestine, respectively) provided the highest DH values. In vitro digestibility demonstrated that PTB was the highest (85.3%) followed by PBPM (51.4%), CAN + phytase (47.6%), CG (45.1%), and CHKM (46.5%). The in vivo experiment concerned the WG, CHKM, PBPM, PM, and PTB diets plus a reference diet with fishmeal (Ref diet). The total %DH was different (P < 0.05) with the lowest values for the WG diet (0.34 ± 0.09%) and Ref diet (0.34 ± 0.15). Free amino acid released during digestion was displayed for these diets and a bifactorial analysis produced no difference (P > 0.05). The apparent digestive coefficients ranged from 89.8 to 92.9% for protein and from 68 to 71.4% for energy.  相似文献

Our work with both sheep and mouse models has revealed many of the neural substrates and signalling pathways involved in olfactory recognition memory in the main olfactory system. A distributed neural system is required for initial memory formation and its short-term retention-the olfactory bulb, piriform and entorhinal cortices and hippocampus. Following memory consolidation, after 8 h or so, only the olfactory bulb and piriform cortex appear to be important for effective recall. Similarly, whereas the glutamate-NMDA/AMPA receptor-nitric oxide (NO)-cyclic GMP signalling pathway is important for memory formation it is not involved in recall post-consolidation. Here, within the olfactory bulb, up-regulation of class 1 metabotropic glutamate receptors appears to maintain the enhanced sensitivity at the mitral to granule cell synapses required for effective memory recall. Recently we have investigated whether fluctuating sex hormone levels during the oestrous cycle modulate olfactory recognition memory and the different neural substrates and signalling pathways involved. These studies have used two robust models of social olfactory memory in the mouse which either involve social or non social odours (habituation-dishabituation and social transmission of food preference tasks). In both cases significant improvement of learning retention occurs when original learning takes place during the proestrus phase of the ovarian cycle. This is probably the result of oestrogen changes at this time since transgenic mice lacking functional expression of oestrogen receptors (ERalpha and ERbeta, the two main oestrogen receptor sub-types) have shown problems in social recognition. Therefore, oestrogen appears to act at the level of the olfactory bulb by modulating both noradrenaline and the glutamate/NO signalling pathway.  相似文献

The prevalence of anti-Toxoplasma gondii antibodies was evaluated by the indirect immunofluorescent-antibody test in serum of 57 wild canids from three different species: Lycalopex gymnocercus, Cerdocyon thous and Dusicyon vetulus from the northeast, southeast and southern regions of Brazil. The prevalence was 35.1%, with 20 of the 57 canids demonstrating antibodies anti-T. gondii at dilutions of 1:16 in 2, 1:32 in 4, 1:64 in 2, 1:128 in 2, 1:256 in 6, 1:512 in 2 and 1:2048 in 2 animals. None of the D. vetulus were positive. Among the L. gymnocercus 11 (91.7%) of the 12 samples were positive and among C. thous 9 (60%) of the 15 had antibodies anti-T. gondii.  相似文献

Babesia microti-like piroplasms are a recently recognized cause of illness in dogs in northwest Spain. Our objective was to describe the clinical characteristics and investigate the risk factors for azotemia and death among 58 B microti-like infected dogs. Twenty-one of the 58 (36%) dogs were azotemic at the time that the infection was diagnosed. The case fatality rate during the following week was 22%. Dogs with azotemia at the time of diagnosis were 10 times (95% CI, 3.26-28.8) more likely to die during the following week. Azotemia was the main cause of death for B microti-like infected dogs (attributable fraction = 90%). Severe anemia was present in 45 of the 58 (78%) dogs. Azotemic dogs also presented with hyperphosphatemia, hypoalbuminemia, hypercholesterolemia, proteinuria, and high urine protein: creatinine ratios, suggesting a glomerular component to the disease. Age was the only factor significantly associated with the risk of azotemia (P = .042): on average, a 4-year age increase doubled the risk of an infected dog being azotemic. The only factor significantly associated with mortality was azotemia (P = .001). We concluded that B microti-like infection is associated with a high risk of azotemia and mortality.  相似文献
