African catfish, Clarias gariepinus were exposed to total darkness (group D) or continuous light (L) during endogenous feeding. During external feeding some of the fish continued to be reared in darkness (DD) and some in light (LL), whereas two groups were exposed to reversed light conditions (groups DL and LD). Survival to the end of yolk absorption was 22% greater in fish exposed to darkness; during subsequent rearing survival decreased in the sequence DD>LD>DL>LL. The onset of external feeding was delayed by a few hours in the L-group as compared with the D-group. Fish reared in dark were larger than those reared in light; the size difference increased with age. In dark, the ratio of total metabolism to body growth (the Rtot/P ratio, both in terms of energy) was depressed, hence in the dark, energy used for locomotor activity may have been low, leading to increased investment in growth. We hypothesised that in juveniles light exerts an indirect effect by increasing locomotor activity which in turn promotes multiple encounters between individuals and enhances cannibalistic behaviour. During the fifth and sixth weeks post-fertilization the biomass of fish reared in the dark was about 175% of that in fish reared in light. Light restriction may be recommended as a simple, low-cost technique for intensification of production of C. gariepinus stocking material.  相似文献

Potex constitutes a potato fiber preparation widely used as an ingredient to meat and bakery products which thermal treatment results in creation of new compounds. Melanoidins are high molecular weight brown end products of Maillard reaction, and few data presenting tumor cell growth inhibiting activity of melanoidins have been reported. Thus, in present study we utilized water extract of Potex roasted (180 °C for 2 h), whose chemical characterization revealed the presence of melanoidin complexes. Heated Potex extract inhibited C6 glioma cell proliferation in a dose-dependent manner measured by MTT method. High molecular weight components present in initial extract were responsible for stronger antiproliferative effect compared with low molecular weight fraction. Impaired MAPK (mitogen-activated protein kinase) and Akt signaling was found in cells treated with the extract. Moreover, flow cytometry analyses revealed the extract to induce G1/S arrest in glioma cells. Simultaneously, Western blot analysis showed elevated levels of p21 protein with concomitant decrease of cyclin D1. In conclusion, observed antiproliferative activity of melanoidins present in heated Potex was linked to disregulated MAPK and Akt signaling pathways, as well as to cell cycle cessation. These results suggest potential application of Potex preparation as a functional food ingredient and chemopreventive agent.  相似文献

Coal carbonization by-products contain up to 10,000 aliphatic and aromatic compounds. Many of them show toxic, mutagenic, and carcinogenic character. In this study, we examined 51 pure bacterial cultures of their ability of coal tar constituent biodegradation. Bacterial cultures were isolated from both explosives and coal tar-contaminated areas. Among all of the investigated strains, 19 showed biodegradative activity. One of the isolates degraded 40% of the substrate in 14 days at a temperature of 15°C. The most active strain was identified by both classic and 16S ribosomal DNA sequencing methods and designated Rhodococcus erythropolis B10. The biodegradation of coal tar constituents, performed by identified strain, was assessed by GC/MS technique. The comparison of samples analyzed by GC/MS before and after biodegradation indicated high degradative potential of the chosen strain. It was able to degrade n-paraffins, n-olefins, benzene, alkylbenzenes, cadalene, and other PAHs, as well as recalcitrant heterocyclic compounds dibenzofuran and its methyl-substituted derivative. The B10 strain isolated and tested in this research shows promising possibilities to be used in field conditions. The biodegradation experiments indicated that satisfactory results may be obtained even in pure bacterial cultures.  相似文献

Heavy metals in running water are analysed within the Swedish Surface Water Monitoring Programme at about 80 stations. Data for selected rivers and brooks has been used to assess the effect of soil acidification on the concentrations of metals in waters. In southern Sweden, acidification shows a significant effect on Zn and Cd. In the upper parts of the drainage areas, there is an marked increased leakage of these metals from soils to waters indicated by elevated concentrations of Zn and Cd in brooks and also by high maxima during periods of low pH values. The increased leaching from forest soils also influences the concentrations in rivers in southern Sweden. During periods of high water flow, the pH values of the rivers decreases and the concentrations of Zn and Cd increases. At pH levels of 6.2–6.5 the concentrations of these metals are mostly 2–5 times higher as compared to pH levels of about 7.0. Since high concentrations of Zn and Cd coincides with high water flow, the transport of these metals has most certainly increased several times compared to the pre-industrial period. Hence, the by far most important human impact on the loading of Zn and Cd on the marine areas, surrounding southern Sweden is the increased leakage of these metals due to acidification. In northern Sweden the effects of acidification on Zn and Cd are less evident. In general, the concentrations of these metals are lower and the connection between pH and these metals are much less significant in the rivers. The links between acidification and the concentrations of Pb and Cu in the watercourses are comparatively much weaker. These two metals are more related to the content of organic matter in the waters and a possible effect of acidification is overshadowed by natural transport processes in soils and waters.  相似文献

As a result of increasing frequency and intensity of tropical cyclones, an accurate forecasting of cyclone evolution and ocean response is becoming even more important to reduce threats to lives and property in coastal regions. To improve predictions, accurate evaluation of the air-sea momentum exchange is required. Using current observations recorded during a major tropical cyclone, we have estimated this momentum transfer from the ocean side of the air-sea interface, and we discuss it in terms of the drag coefficient. For winds between 20 and 48 meters per second, this coefficient initially increases and peaks at winds of about 32 meters per second before decreasing.  相似文献

Hurricane Ivan, a category 4 storm, passed directly over six wave-tide gauges deployed by the Naval Research Laboratory on the outer continental shelf in the northeastern Gulf of Mexico. Waves were observed with significant wave heights reaching 17.9 meters and maximum crest-to-trough individual wave heights of 27.7 meters (91 feet). Analysis suggests that significant wave heights likely surpassed 21 meters (69 feet) and that maximum crest-to-trough individual wave heights exceeded 40 meters (132 feet) near the eyewall.  相似文献

Background

A single prolonged milking interval (PMI) e.g. after a technical stop in an automated milking system is of concern for the producer since it is associated with a short-lasting increase in milk somatic cell count (SCC), which is a major quality criterion used at the dairy plants. The content of polymorphonuclear leukocytes (PMN) and how the milk quality is influenced has not been much investigated. The SCC peak occurs without any obvious antigen challenge, possibly indicating a different leukocyte attraction mechanism after a PMI than we see during mastitis.

Methods

Composite cow milk samples were taken at the milkings twice daily during 7 days before and 5 days after a PMI of 24 h. Milk was analyzed for SCC, PMN, fat, protein and lactose, and at some occasions also casein and free fatty acids (FFA).

Results

During the PMI the proportion of milk PMN increased sharply in spite of marginally increased SCC. The peak SCC was not observed until the second milking after the PMI, in the afternoon day 1. However, the peak SCC value in morning milk did not occur until one day later, concomitantly with a decrease in the proportion of PMN. After declining, SCC still remained elevated while PMN proportion was decreased throughout the study as was also the milk yield, after the first accumulation of milk during the PMI. Milk composition was changed the day after the PMI, (increased fat and protein content; decreased lactose, whey protein and FFA content) but the changes in the following days were not consistent except for lactose that remained decreased the rest of the study.

Conclusion

The PMI resulted in increased SCC and proportion of PMN. Additionally, it gave rise to minor alterations in the milk composition in the following milkings but no adverse effect on milk quality was observed. The recruitment of PMN, which was further enhanced the first day after the PMI, appeared to be independent of milk volume or accumulation of milk per se. Hence, we suggest that there is a special immunophysiological/chemoattractant background to the increased migration of leukocytes into the milk compartment observed during and after the PMI.  相似文献

ABSTRACT

The aim of the study was to assess the effects of different forest stands (Silver fir (Abies alba) and sycamore maple (Acer pseudoplatanus) with common hornbeam (Carpinus betulus)) on the enzymes activities and microbial biomass. The objective was to explore how changes in the detritus inputs affect soil organic matter (SOM) composition. The content of SOM fraction has been compared with soil enzyme activities. The investigation was carried out in the ?wi?tokrzyskie Mountains of central Poland. Twenty investigation plots were selected, including fir stands (10 plots) and maple with hornbeam stands (10 plots). Contents of organic C, N and base cations, pH, hydrolytic acidity, and soil texture were investigated. The content of C and N in the physically separated SOM fractions was distinguished. The study indicated only small changes in soil properties and stabilization of organic matter as a result of different detritus inputs. The maple-hornbeam and fir stands have a similar influence on microbiological processes and the SOM. Acidity of soil is a major factor affecting microbial activity and therefore pH affects enzyme dynamics. Differences in soil pH confirmed the stronger acidifying effects of fir stands compared to maple-hornbeam stands. Additionally, fir stands stimulate β-glucosidase activity, probably through a simultaneous interaction of mycorrhizal fungi in the roots of fir stands.  相似文献

ABSTRACT

This article presents the effects of nitrate/ammonium (NO3 ?/NH4 +), applied at different proportions to the root media with or without 5 mmol bicarbonate (HCO3 ?), on the yield and chemical composition of tomato fruit. Tomato plants were grown hydroponically (pH 6.9) in glasshouse conditions. The yield of fruit fresh matter from four clusters obtained from plants grown on the medium with NH4 + was about 25% lower than from the plants grown on the medium containing NO3 ? as the nitrogen (N) source. Supplying NO3 ?/NH4 + at a ratio of 4:1 increased the fruit yield by about 20% in comparison with the value recorded for NO3 ??plants. The enrichment of the medium with HCO3 ? stimulated the bearing, while the result depended on the ratio of NO3 ?/NH4 +. A combined treatment of HCO3 ? with NO3 ? or NH4 + in the medium increased yields by about 28% and 11%, respectively, in comparison to plants cultivated without HCO3 ?. The application of NO3 ?/NH4 + at ratios of 4:1 and 1:1 with HCO3 ? increased the respective yields by about 16% and 10% in comparison with plants grown without HCO3 ?. Modifications in the composition of the media affected the accumulation of organic solutions in the fruit. The NH4 + nutrition effected a 20% decrease in the accumulation of reducing sugars in the fruit in comparison to the fruit of plants grown in media with NO3 ?. In the cultivation of plants in media with various NO3 ?/NH4 + proportions the intermediate values of the reduced sugar concentrations were recorded in comparison with the values obtained for NO3 ??plants and NH4 +?plants. The enrichment of media with HCO3 ? increased the concentration of sugars in fruit from about 28% (for NO3 ??plants) to about 10% (for NH4 +?plants).

Malate and citrate are the main constituents of carboxylates in tomato fruit. The form of nitrogen applied to the medium did not significantly affect the concentration of carboxylates in fruit. Significant differences in carboxylate concentrations appeared in fruit grown on media enriched with HCO3 ? ions. In comparison with the cultivation without HCO3 ?, increases in the accumulation of carboxylates varied from about 22% to 30% depending on the form of the applied nitrogen. The concentration of amino acids in the fruit of plants grown with NH4 + exceeded that in NO3 ??plants by about 55%. In the plants grown on media of modified NO3 ?/NH4 + proportions, the concentration of amino acids in fruits were positively correlated with the level of NH4 + in the medium. The enrichment of media with HCO3 ? stimulated a further increase in amino acid concentration in fruit by about 9% in NO3 ? plants and about 21% in NH4 + plants compared with the respective control (without HCO3 ?).  相似文献
