In the present work, direct methods for the determination of chromium, copper, and nickel in honey by electrothermal atomic absorption spectroscopy were developed using experimental design as an optimization tool. Once the optimum conditions for the individual methods were established, a direct method for the combined determination of the three elements was optimized using the response surface tool. Palladium was used as chemical modifier in all cases. Honey was diluted in water, hydrogen peroxide, and nitric acid. Triton X-100 was added to minimize the matrix effect and the viscosity of the sample. The RSD (better than 10%) and the analytical recovery (98-103%) were acceptable for all of the developed methods. Calibration graphs were used in the four methods to determine the concentration of the analytes in the sample. The detection limits of the combined method (0.21, 0.35, and 0.37 microg L(-)(1) for Cr, Cu, and Ni, respectively) were similar to those obtained for the individual methods (LOD = 0.17, 0.21, 0.33 microg L(-)(1) for Cr, Cu, and Ni, respectively). The direct-combined proposed method has been applied to the determination of chromium, copper, and nickel content in representative honey samples from Galicia (northwestern Spain). The concentrations found in the analyzed samples were in the range of (5.75 +/- 0.64)-(26.4 +/- 0.38) ng g(-)(1) of Cr, (79 +/- 7.8)-(2049 +/- 80) ng g(-)(1) of Cu, and (12.6 +/- 1.36)-(172 +/- 6.88) ng g(-)(1) of Ni. 相似文献
A neutral peroxidase isozyme (pI 7.2) from turnip roots (TNP) was purified to homogeneity and partially characterized. TNP is a monomeric glycoprotein with 9.1% carbohydrate content and a molecular weight of 36 kDa. Optimum pH values for activity using 2,2'-azinobis(3-ethylbenzthiazoline-6-sulfonic acid (ABTS) and guaiacol as H donors were 4.5 and 5.5, whereas the K(m) values were 0.7 and 3.7 mM, respectively. The ABTS K(m) was approximately 7 times higher than that reported for basic commercial horseradish peroxidase (HRP-C). TNP retained approximately 70% activity after 11 min of heating at 65 degrees C, whereas the activation energy for inactivation (132 kJ/mol) was higher than or comparable to that of other peroxidases. The low ABTS K(m) and high specific activity (1930 units/mg) gave a high catalytic efficiency (500 M(-1) s(-1)). The formation of inclusion complexes with beta-cyclodextrin was studied for several popular fungicides of different types: prochloraz, 2-phenylphenol, thiophanate methyl, 8-hydroxyquinoline, and benalaxyl. Phase solubility diagrams showed that in all cases complexation takes place, leading to an important increase of water solubility in prochloraz and benalaxyl. Equilibrium association constants could be determined from the phase solubility data and from NMR titrations in the case of 2-phenylphenol. Because of the low solubility of the complex formed between 8-hydroxyquinoline and beta-cyclodextrin, the corresponding association constant could not be determined. The solid complexes of fungicide-cyclodextrin were prepared and isolated by different methods. The isolation of real complexes and not physical mixtures was confirmed in the cases of prochloraz, 2-phenylphenol, and benalaxyl by differential scanning calorimetry. 相似文献
OBJECTIVE: To evaluate dietary habits in Spanish children and adolescents based on a Mediterranean Diet Quality Index tool, which considers certain principles sustaining and challenging traditional healthy Mediterranean dietary patterns. DESIGN: Observational population-based cross-sectional study. A 16-item Mediterranean Diet Quality Index was included in data gathered for the EnKid study (in which two 24-hour recalls, a quantitative 169-item food-frequency questionnaire and a general questionnaire about socio-economic, demographic and lifestyle items were administered). SETTING: Spain. SUBJECTS: In total, 3850 children and youths aged 2-24 years residing in Spain. RESULTS: Of the sample, 4.2% showed very low KIDMED index results, 49.4% had intermediate values and 46.4% had high index results. Important geographical differences were seen, with subjects from the Northeast showing the most favourable outcomes (52% with elevated scores vs. 37.5% of those from the North). Lower percentages of high diet quality were observed in low socio-economic groups, compared with middle and upper income cohorts (42.8%, 47.6% and 54.9%, respectively). Large cities had more positive results and only slight variations were seen for gender and age. CONCLUSIONS: The KIDMED index, the first to evaluate the adequacy of Mediterranean dietary patterns in children and youth, confirms that this collective is undergoing important changes, which makes them a priority target for nutrition interventions. The application of sludge from wastewater in agriculture has increased in recent years, and it is therefore important to assess the effect that such treatment has on both the soil and the plant. The aim of the study described here was to ascertain whether there is a variation in the properties of the soil and to determine if this addition has an impact on the plant.
Materials and methods
The area of investigation was close to the municipality of Villarrubia de los Ojos (Ciudad Real). In this work, six samples were taken from the surface horizon in the studied plot at a depth of 35 cm. A further three samples were taken: (i) a surface horizon of a soil close to the area under investigation but without treatment (control sample), (ii) a sample of sludge from the wastewater treatment plant and (iii) a sample of the mixture used by farmers as fertilizer. Laboratory tests were conducted in accordance with the SCS-USDA (1972) guidelines. Trace element samples were analysed by X-ray fluorescence spectrophotometry (Philips PW 2404).
Results and discussion
The parcel of land studied is dominated by a sandy texture (88.3 % sand), and a decrease in pH was observed in areas in which the mixture (manure + sludge) was added (pH?=?8.0) compared to areas in which fertilizer was not applied (pH?=?8.5). It was observed that the addition of the compound led to an increase in the electrical conductivity of the soil. The trace elements can be organized into two groups based on the results obtained in this study. One group contains the trace elements that were only present in the rows that were treated with the fertilizer. The other group of trace elements was mobilized throughout the whole plot.
Conclusions
The application of sewage sludge on agricultural soils can be very useful as an organic amendment because it produces an increase in soil organic matter. However, sewage sludge must be applied with caution due to the changes in soil chemical properties (for example, pH and E.C.). The use of this type of waste for prolonged periods of time can cause problems of contamination in the soil.
The El Granero reservoir is the last reservoir of the Rio Conchos before it joins the Rio Grande at the Mexico–USA border. This reservoir, together with the San Marcos reservoir, is located in the arid region of Chihuahua, Mexico. High, naturally occurring radioactivity levels, as well as high arsenic (As) concentrations, have been found in both reservoirs. The main goal of this research was to establish the spatial and temporal distribution of trace and radioactive elements in surface sediments and cores collected from these reservoirs.
Materials and methods
Sediment cores were dated using 210Pb and 137Cs measurements and applying the constant rate of supply (CRS) model. Major, trace, and radioactive elements were determined in surface samples and three sediment cores. Radioactive elements were determined by both alpha and gamma spectrometry. Major and trace elements were determined by inductively coupled plasma optical emission spectrometry (ICP-OES) using the EPA 3051a method. Enrichment factors (EF), contamination factors (CF), and pollution load indexes (PLI) were calculated in order to identify the human impact in both reservoirs, whereas the chemical index weathering (CIW) was used to assess differences in the degree of weathering.
Results and discussion
High uranium (U) enrichment (EFs?=?24.9–54.7) was observed in core layers at the San Marcos reservoir, while in surface sediments, this enrichment was lower. The high variability of lead (Pb) and As in sediment cores from the Granero reservoir was attributed to human influence. Arsenic and Pb enrichment differences between entry and exit sediment cores were explained by the filtering capabilities of the elongated shape, the topography, and the presence of plants on the reservoir’s bed. The highest PLI was found at the entrance core of the Granero reservoir.
Conclusions
The natural element concentration levels of As, Pb, and U were established at the Granero reservoir. High EFs for As and Pb suggest an anthropogenic origin of these pollutants at specific time intervals. High U concentrations in the San Marcos area are explained as naturally occurring. The concentrations of As in most of the studied sediments could pose a risk to human health by As ingestion, since they are above the probable effect level (PEL).
In 1998, the pond containing the ore wastes from a pyrite mine in Aznalcóllar (SW, Spain) broke open, spilling some 36×105 m3 of acidic waters and 9?×?105 m3 of tailings containing high concentrations of As and heavy metals. The affected area was around 55 km2 of predominantly agricultural soils. After the clean-up of the tailings, many remediation actions were undertaken and the use of blocking agents to immobilize the As was one of the most extended measure. The first experiment performed was to determine the most important soil components in As adsorption under acidic conditions. A second experiment was conducted to neutralize the acidity caused by the solution coming from the tailings undergoing oxidation; an adequate liming material (sugar-refinery scum) was selected and the application rates were established. After the remediation measures, the zone was monitored for three years. A detailed study in four experimental plots located in the most polluted sector was carried out to test the influence of iron oxides in the As immobilization. Must obtained from Airén grapes was fermented in the presence of wood chips (4 and 7 g/L) of either French oak (from Vosges, central France, and Allier zones) or American oak. Fermentation yields were higher than in the control fermentations carried out in the absence of wood, and production of volatile substances during fermentation (alcohols, esters, and acetates) was also higher. The volatile substances that leached out of the wood were analyzed by GC-MS-SIR. The results showed that their concentrations depended on the type and amount of the oak; some of these substances were consumed in part by the yeasts during fermentation. A taste panel favorably assessed the wines produced by fermentation in the presence of oak chips, which retained part of the must original fruity aroma. 相似文献
Salsolinol, a tethrahydroisoquinoline present in banana and biosynthesized from dopamine, was oxidized by banana pulp polyphenol oxidase to its corresponding salsolinol-o-quinone. This oxidation was pH-dependent and showed a maximum at acidic pH values. At physiological pH of 5.0, the values obtained for the kinetic parameter (V(m) and K(m)) were 62.5 microM/min and 1.7 mM, respectively. When dopamine was added to the reaction medium to imitate physiological conditions, salsolinol was co-oxidized by dopamine-quinone. When this phenomenon was studied oxygraphically, an unexpected activation of dopamine oxidation was found in the presence of salsolinol. This activation was related with the enzyme's kinetic mechanism and was named "kinetic synergism", because a bad substrate activated a good one. A possible physiological role is discussed. 相似文献
