ABSTRACT: The mycotoxin T-2 toxin and Salmonella Typhimurium infections pose a significant threat to human and animal health. Interactions between both agents may result in a different outcome of the infection. Therefore, the aim of the presented study was to investigate the effects of low and relevant concentrations of T-2 toxin on the course of a Salmonella Typhimurium infection in pigs. We showed that the presence of 15 and 83 μg T-2 toxin per kg feed significantly decreased the amount of Salmonella Typhimurium bacteria present in the cecum contents, and a tendency to a reduced colonization of the jejunum, ileum, cecum, colon and colon contents was noticed. In vitro, proteomic analysis of porcine enterocytes revealed that a very low concentration of T-2 toxin (5 ng/mL) affects the protein expression of mitochondrial, endoplasmatic reticulum and cytoskeleton associated proteins, proteins involved in protein synthesis and folding, RNA synthesis, mitogen-activated protein kinase signaling and regulatory processes. Similarly low concentrations (1-100 ng/mL) promoted the susceptibility of porcine macrophages and intestinal epithelial cells to Salmonella Typhimurium invasion, in a SPI-1 independent manner. Furthermore, T-2 toxin (1-5 ng/mL) promoted the translocation of Salmonella Typhimurium over an intestinal porcine epithelial cell monolayer. Although these findings may seem in favour of Salmonella Typhimurium, microarray analysis showed that T-2 toxin (5 ng/mL) causes an intoxication of Salmonella Typhimurium, represented by a reduced motility and a downregulation of metabolic and Salmonella Pathogenicity Island 1 genes. This study demonstrates marked interactions of T-2 toxin with Salmonella Typhimurium pathogenesis, resulting in bacterial intoxication.  相似文献

Combinations of anthelmintics with a similar spectrum of activity and different mechanisms of action and resistance are widely available in several regions of the world for the control of sheep nematodes. There are two main justifications for the use of such combinations: (1) to enable the effective control of nematodes in the presence of single or multiple drug resistance, and (2) to slow the development of resistance to the component anthelmintic classes. Computer model simulations of sheep nematode populations indicate that the ability of combinations to slow the development of resistance is maximised if certain prerequisite criteria are met, the most important of which appear to concern the opportunity for survival of susceptible nematodes in refugia and the pre-existing levels of resistance to each of the anthelmintics in the combination. Combinations slow the development of a resistant parasite population by reducing the number of resistant genotypes which survive treatment, because multiple alleles conferring resistance to all the component anthelmintic classes must be present in the same parasite for survival. Individuals carrying multiple resistance alleles are rarer than those carrying single resistance alleles. This enhanced efficacy leads to greater dilution of resistant genotypes by the unselected parasites in refugia, thus reducing the proportion of resistant parasites available to reproduce with other resistant adults that have survived treatment. Concerns over the use of anthelmintic combinations include the potential to select for resistance to multiple anthelmintic classes concurrently if there are insufficient parasites in refugia; the potential for shared mechanisms of resistance between chemical classes; and the pre-existing frequency of resistance alleles may be too high on some farms to warrant the introduction of certain combinations. In conclusion, anthelmintic combinations can play an important role in resistance management. However, they are not a panacea and should always be used in accordance with contemporary principles for sustainable anthelmintic use.  相似文献

Aetiology, clinicopathological findings and treatment outcomes were documented in 23 cats (1.5-13 years of age) with sinonasal (SNA, n=6) or sino-orbital (SOA, n=17) aspergillosis. Cases recruited retrospectively and prospectively were included if fungal hyphae were identified on cytological or histological examination and the fungal pathogen was identified by PCR and DNA sequencing (ITS1 or ITS1-5.8S-ITS2 regions, rDNA gene cluster). Fungal culture was positive in 22/23 cases. In cases of SNA, the fungal pathogen was Aspergillus fumigatus (n=4), Neosartorya fischeri or A. lentulus (n=1) or a non-speciated Neosartorya spp. (n=1). In all cases of SOA (n=17), the fungal pathogen was identified as Neosartorya spp. Nine cats had brachycephalic conformation. Cats with SNA were more likely to be infected with A. fumigatus and had a better prognosis than cats with SOA.  相似文献

Grain weight and grain length are the most stable components of rice yield and important indicators of consumer preference. Considering the potentials of wild rice and to enhance the rice yields to meet the increasing demands, 185 Backcross Inbred Lines (BILs) in the background of O. sativa ssp. indica cv. PR114, including 63 rufi-BILs derived from O. rufipogon IRGC104433 and 122 glumae-BILs from O. glumaepatula IRGC104387 were evaluated for mapping QTLs for yield and yield component traits using Genotyping by Sequencing (GBS). Phenotypic evaluation of BILs in three seasons spanning two locations revealed significant differences compared with recurrent parent. BILs which did not show significant differences for any trait under investigation, or similar based on pedigree, were excluded from GBS. Some glumae-BILs had to be excluded from mapping QTLs due to less sequence information. A custom designed approach for GBS data analysis identified 3322 informative SNPs in 55 rufi-BILs and 3437 informative SNPs in 79 glumae-BILs. QTL mapping identified one QTL for thousand grain weight (qtgw5.1), two for grain width (qgw5.1, qgw5.2) and one for grain length (qgl7.1) in rufi-BILs. In the glumae-BILs, three QTL for thousand grain weight (qtgw2.1, qtgw3.1, qtgw6.1) and two for grain length (qgl3.1, qgl7.1) were identified. Most of the grain weight and width QTL showed positive additive effect contributed by wild species allele, whereas the grain length QTL showed positive additive effect contributed by recurrent parent allele. Based on their physical position, none of the QTLs were found similar to previously cloned QTLs. QTLs for grain traits identified from low yielding wild relatives of rice reveals their significance in improving further the rice yields and widen the genetic base of cultivated rice.  相似文献

The affirmation of acrylamide as a probable carcinogen by the European Food Safety Authority has reinforced the need to lower acrylamide content in fried potato products. Selected for low reducing sugars and acrylamide-forming potential, recently released cultivars ‘Alpine Russet’, ‘Dakota Trailblazer’, and ‘Ivory Crisp’ were evaluated for their processing quality when grown with varying nitrogen (N) fertilizer regimes. The objective of this study was to determine the effects of N fertilizer rate (34, 135, 202, 269 and 336 kg ha?1) on tuber glucose and acrylamide concentration following processing of new cultivars relative to standard cultivars ‘Russet Burbank’ and ‘Snowden’ at harvest, and after 3, 6 and 9 months of storage at 7.2 °C over 2 years. Glucose and acrylamide responses to N rate were similar for chip cultivars, which linearly decreased in 2011, and quadratically increased then decreased in 2012 with increasing N rate. The N rate effect on French fry glucose concentration varied by cultivar and either decreased or did not respond to elevated N rate. Glucose and acrylamide concentrations of chip cultivars generally increased during storage, with a dramatic increase in ‘Snowden’ resulting from senescence sweetening after 9 months of storage. Environmental conditions significantly affected glucose and acrylamide responses of French fry cultivars to storage time. Glucose and acrylamide concentrations of all French fry cultivars generally increased during storage in 2011. In contrast, glucose concentrations of French fry cultivars were stable or increased, while acrylamide concentrations generally decreased during storage in 2012. The relationship between chip color and glucose concentration was significant, but differed by year. Glucose and acrylamide concentrations of French fry and chip cultivars were significantly correlated (R2?=?0.52 and 0.66, in 2011 and 2012, respectively). Generally, acrylamide in fried potato products can be minimized by using low reducing sugar cultivars supplied with a N fertilizer rate that optimizes yield and quality during growing seasons with minimal environmental stress.  相似文献

About 7 million beef cattle are raised annually in the Texas High Plains, producing 16 million Mg of manure, which is land-applied as raw manure (RM) to crops. An interest in aerobic composting prompted a 2-year field study with the primary objective of evaluating corn (Zea mays L.) yield and soil fertility for RM as compared to composted manure (CM). The four treatments (TRT) consisted of low-rate composted manure (CM-L), high-rate composted manure (CM-H), RM, and inorganic fertilizer (IN). All four TRT received equal plant—available N by supplementing the RM and CM with IN. Yield was measured by both machine-harvesting and hand-harvesting methods. Machine-harvested yield was similar among TRT for both years. Hand-harvested yield was lower for IN than RM in the first year, with similar yields among TRT in the second year. Due to drought, yields were considerably lower in the second year for all TRT. After 2 years, CM-H had higher soil P, K, Zn, and organic carbon concentrations than IN, with no differences among TRT for soil NO3-N, pH, or electrical conductivity. Both CM and RM provided beneficial nutrients for corn production and improving soil fertility. Land application of CM and RM provides long-term benefits to soil health and sustainability in the Texas High Plains region.  相似文献

Sediments from 18 different road runoff detention systems, located on the Swedish West Coast, were assessed for their ecological hazard potential. Thirteen of the sites were detention ponds, three were manholes within the same sedimentation construction, and two were detention basins handling wash water from road tunnels. Sediments from all sites were analysed for a range of physico-chemical parameters and contaminants, and screened for acute toxicity using Hyalella azteca (sediment), Daphnia magna (elutriate), and Ceriodaphnia dubia (elutriate) as the test organisms, and for chronic toxicity using C. dubia as the test organism. The benthic fauna of the thirteen detention ponds was also studied. Sediment quality guidelines probable effect levels were exceeded for one or several contaminants at half of the sites, and one third revealed toxicity in some of the bioassays. Most of the detention ponds were dominated by tolerant taxa indicating low biological quality. Relationships between contaminant concentrations, toxicity in bioassays, and benthic fauna were, however, found to be weak. Extractable organic Zn, which was used as a tire wear marker, correlated with Zn, Cu, presumably from brake linings, and W, a common component of tire studs. The highest concentration, which was found in the manholes (14 mg kg?1 ds), corresponds to a tire wear concentration of 11 g kg?1 ds. The results of the present study have shown that traffic related contaminants accumulate in the studied runoff treatment systems, and, therefore, the maintenance of them is crucial in order to prevent contamination of surrounding waters.  相似文献
