Amplification of a gene related to mammalian mdr genes in drug-resistant Plasmodium falciparum

The malaria parasite Plasmodium falciparum contains at least two genes related to the mammalian multiple drug resistance genes, and at least one of the P. falciparum genes is expressed at a higher level and is present in higher copy number in a strain that is resistant to multiple drugs than in a strain that is sensitive to the drugs.     

Fluorometric detection of enzyme activity with synthetic supramolecular pores

The reversible blockage of synthetic pores formed by rigid-rod beta barrels, either by substrates or products, was used to sense a variety of enzymatic reactions in high-throughput format with "naked-eye" fluorescent detection. Improvement of sensor sensitivity beyond three orders of magnitude by straightforward internal mutations underscores the functional plasticity of rigid-rod beta barrels. Such detectors of enzyme activity with the aforementioned characteristics are needed in areas as diverse as proteomics and environmentally benign organic synthesis.     

Lhx2 selector activity specifies cortical identity and suppresses hippocampal organizer fate

The earliest step in creating the cerebral cortex is the specification of neuroepithelium to a cortical fate. Using mouse genetic mosaics and timed inactivations, we demonstrated that Lhx2 acts as a classic selector gene and essential intrinsic determinant of cortical identity. Lhx2 selector activity is restricted to an early critical period when stem cells comprise the cortical neuroepithelium, where it acts cell-autonomously to specify cortical identity and suppress alternative fates in a spatially dependent manner. Laterally, Lhx2 null cells adopt antihem identity, whereas medially they become cortical hem cells, which can induce and organize ectopic hippocampal fields. In addition to providing functional evidence for Lhx2 selector activity, these findings show that the cortical hem is a hippocampal organizer.     

Seasonal speedup along the western flank of the Greenland Ice Sheet

It has been widely hypothesized that a warmer climate in Greenland would increase the volume of lubricating surface meltwater reaching the ice-bedrock interface, accelerating ice flow and increasing mass loss. We have assembled a data set that provides a synoptic-scale view, spanning ice-sheet to outlet-glacier flow, with which to evaluate this hypothesis. On the ice sheet, these data reveal summer speedups (50 to 100%) consistent with, but somewhat larger than, earlier observations. The relative speedup of outlet glaciers, however, is far smaller (<15%). Furthermore, the dominant seasonal influence on Jakobshavn Isbrae's flow is the calving front's annual advance and retreat. With other effects producing outlet-glacier speedups an order of magnitude larger, seasonal melt's influence on ice flow is likely confined to those regions dominated by ice-sheet flow.     

Giant piezoelectricity on Si for hyperactive MEMS

Microelectromechanical systems (MEMS) incorporating active piezoelectric layers offer integrated actuation, sensing, and transduction. The broad implementation of such active MEMS has long been constrained by the inability to integrate materials with giant piezoelectric response, such as Pb(Mg(1/3)Nb(2/3))O(3)-PbTiO(3) (PMN-PT). We synthesized high-quality PMN-PT epitaxial thin films on vicinal (001) Si wafers with the use of an epitaxial (001) SrTiO(3) template layer with superior piezoelectric coefficients (e(31,f) = -27 ± 3 coulombs per square meter) and figures of merit for piezoelectric energy-harvesting systems. We have incorporated these heterostructures into microcantilevers that are actuated with extremely low drive voltage due to thin-film piezoelectric properties that rival bulk PMN-PT single crystals. These epitaxial heterostructures exhibit very large electromechanical coupling for ultrasound medical imaging, microfluidic control, mechanical sensing, and energy harvesting.     

Theaflavins depolymerize microtubule network through tubulin binding and cause apoptosis of cervical carcinoma HeLa cells

Here we studied the antiproliferative activity of theaflavins in cervical carcinoma HeLa cells by investigating their effects on cellular microtubules and purified goat brain tubulin. Theaflavins inhibited proliferation of HeLa cells with IC(50) value of 110 ± 2.1 μg/mL (p = < 0.01), caused cell cycle arrest at G(2)/M phase and induced apoptosis with alteration of expression of pro- and antiapoptotic proteins. Along with these antiproliferative activities, theaflavins act as microtubule depolymerizers. Theaflavins disrupted the microtubule network accompanied by alteration of cellular morphology and also decreased the polymeric tubulin mass of the cells. The polymerization of cold treated depolymerized microtubules in HeLa cells was prevented in the presence of theaflavins. In vitro polymerization of purified tubulin into microtubules was also inhibited by theaflavins with an IC(50) value of 78 ± 2.43 μg/mL (P < 0.01). Thus, disruption of cellular microtubule network of HeLa cells through microtubule depolymerization may be one of the possible mechanisms of antiproliferative activity of theaflavins.     

Effect of Temperature on Bio-Kinetic Coefficients in UASB Treatment of Municipal Wastewater

Upflow anaerobic sludge blanket (UASB)reactors were used to treat municipal wastewater attemperatures of 6, 11, 15, 20 and 32 °C and athydraulic retention times (HRTs) ranging from 48 to 3 h overan operational period of approximately 860 days. The Monodmodel was used to evaluate substrate utilization. TheArrhenius model was used to calculate the activationenergies from which temperature coefficients weredetermined. It was found that the maximum specific substrateutilization rate constant (k) decreased from 0.387 d^-1(at 32 °C) to 0.041 d^-1 (at 6 °C). Thedecay rate constant (k_d), yield coefficient (Y_g),half velocity constant (K_s) and maximum specific growthrate (μ_m ) were also impacted by temperature in theUASB treatment of municipal wastewater.     

Studies on bacteriological quality of nainital lake water in different seasons

The total coliform and E. coli load of Nainital lake water in different seasons has been studied. The variation of these organisms with season, rainfall and human activities in the catchment area were observed.     

Prioritisation of conservation areas in the Western Ghats, India

Areas of high conservation value were identified in the Western Ghats using a systematic conservation planning approach. Surrogates were chosen and assessed for effectiveness on the basis of spatial congruence using Pearson’s correlations and Mantel’s tests. The surrogates were, threatened and endemic plant and vertebrate species, unfragmented forest areas, dry forests, sub-regionally rare vegetation types, and a remotely sensed surrogate for unique evergreen ecosystems. At the scale of this analysis, amphibian richness was most highly correlated with overall threatened and endemic species richness, whereas mammals, especially wide-ranging species, were better at capturing overall animal and habitat diversity. There was a significant relationship between a remote sensing based habitat surrogate and endemic tree diversity and composition. None of the taxa or habitats served as a complete surrogate for the others. Sites were prioritised on the basis of their irreplaceability value using all five surrogates. Two alternative reserve networks are presented, one with minimal representation of surrogates, and the second with 3 occurrences of each species and 25% of each habitat type. These networks cover 8% and 29% of the region respectively. Seventy percent of the completely irreplaceable sites are outside the current protected area network. While the existing protected area network meets the minimal representation target for 88% of the species chosen in this study and all of the habitat surrogates, it is not representative with regard to amphibians, endemic tree species and small mammals. Much of the prioritised unprotected area is under reserve forests and can thus be incorporated into a wider network of conservation areas.     

Insecticidal activity of Arum maculatum tuber lectin and its binding to the glycosylated insect gut receptors

Mannose binding approximately 50 kDa homotetrameric lectin, purified from edible Arum maculatum tuber, was analyzed through SDS-PAGE and studied for its agglutination property using rabbit erythrocytes. Cross reactivity of the purified lectin was verified through western blot using Colocasia esculantum(Family, Araceae) tuber lectin antibody. The insecticidal activity of Arum maculatum tuber lectin (ATL) was tested against two economically important sucking pests, Lipaphis erysimi and Aphis craccivora, in an artificial diet. The LC(50) values for L. erysimi and A. craccivora were determined to be 21 microg/mL and 16 microg/mL, respectively. Addition of alpha-d-mannose in ATL-supplemented diet reduced the aphid mortality. Two major receptor proteins of ATL (approximately 40 kDa and approximately 35 kDa) were detected from the brush border membrane vesicle (BBMV) protein of L. erysimi and A. craccivora guts, respectively, using ligand-binding assay. Alpha-d-Mannose was found to be a deterrent to such binding of ATL to the BBMV receptors.
