In North America, the only endemic focus for Angiostrongylus vasorum (French heartworm) was historically thought to occur in the southeastern part of the island of Newfoundland. However, reports of A. vasorum infection in wild canids in West Virginia, USA, and Nova Scotia, Canada, suggest the introduction of the parasite to mainland North America. We screened for A. vasorum in coyotes from across southern Ontario. Additionally, we evaluated the performance of ELISAs for detection of circulating A. vasorum antigen (Ag-ELISA) and antibodies against A. vasorum (Ab-ELISA) designed for use in sera or blood of foxes for use with coyotes in this region. Autopsies were performed on 397 coyotes, and lung tissue extract prepared from each carcass was tested via both ELISAs. The sensitivity and specificity for both tests were estimated in the absence of a gold standard using a 2-test single population Bayesian model; sensitivity and specificity priors were based on the performance of the assays in foxes in Switzerland. Eight coyotes tested positive for A. vasorum antigen; no animal was antibody positive. The estimated sensitivity and specificity of the Ag-ELISA were 90.8% (95% credible interval [CrI]: 83.8–95.6%) and 95.5% (95% CrI: 93.4–97.2%), respectively. For the Ab-ELISA, the estimated sensitivity and specificity were 41.9% (95% CrI: 32.1–51.9%) and 98.0% (95% CrI: 96.3–99.0%), respectively. Based on these findings and negative postmortem data for the same animals, there is insufficient evidence to suggest the presence of A. vasorum in southern Ontario coyotes.  相似文献

The effects of two different vinification techniques, traditional fermentation and carbonic maceration, on the anthocyanin composition and color of young red wines, made with Syrah grapes grown in a warm climate, were compared. Tristimulus colorimetry was applied to study the color of wines during the vinification, and a high-performance liquid chromatography (HPLC) procedure was used for the analysis of anthocyanins. Carbonic maceration led to wines with lower anthocyanin content, mainly monoglucosides, and total phenols. This was related to lighter wines, less saturated, but more colorful (higher chroma C*ab values), and hues hab similar to those of the Syrah wines made by traditional vinification. Thus, the lightness L* had much more influence on the saturation s*uv of the wines obtained by carbonic maceration than the chroma (s*uv = C*uv/L*). From a study of the color-composition relationships using linear and multiple regression, better relationships were found for the wines from traditional vinification, where the chromatic parameters L*, hab, and s*uv could be predicted from the 3-monoglucosides of delphinidin, petunidin, peonidin, and malvidin concentrations (R > 0.9). However, a good prediction of the chroma C*ab from the anthocyanin composition was not possible. On the contrary, C*ab was the best predicted parameter from the anthocyanins monoglucosides (R > 0.9) in the carbonic maceration wines.  相似文献

Purpose

Polycyclic aromatic hydrocarbons (PAHs) are a class of organic compounds commonly found as soil contaminants. Fungal degradation is considered as an environmentally friendly and cost-effective approach to remove PAHs from soil. Acenaphthylene (Ace) and Benzo[a]anthracene (BaA) are two PAHs that can coexist in soils; however, the influence of the presence of each other on their biodegradation has not been studied. The biodegradation of Ace and BaA, alone and in mixtures, by the white rot fungus Pleurotus ostreatus was studied in a sandy soil.

Materials and methods

Experimental microcosms containing soil spiked with different concentrations of Ace and BaA were inoculated with P. ostreatus. Initial (t 0) and final (after 15 days of incubation) soil concentrations of Ace and BaA were determined after extraction of the PAHs.

Results and discussion

P. ostreatus was able to degrade 57.7% of the Ace in soil spiked at 30 mg kg?1 dry soil and 65.8% of Ace in soil spiked at 60 mg kg?1 dry soil. The degradation efficiency of BaA by P. ostreatus was 86.7 and 77.4% in soil spiked with Ace at 30 and 60 mg kg?1 dry soil, respectively. After 15 days of incubation, there were no significant differences in Ace concentration between soil spiked with Ace and soil spiked with Ace + BaA, irrespective of the initial soil concentration of both PAHs. There were also no differences in BaA concentration between soil spiked with BaA and soil spiked with BaA + Ace.

Conclusions

The results indicate that the fungal degradation of Ace and BaA was not influenced by the presence of each other’s PAH in sandy soil. Bioremediation of soils contaminated with Ace and BaA using P. ostreatus is a promising approach to eliminate these PAHs from the environment.  相似文献

A novel, simple, and cheap method to synthesize antioxidant methacrylic-ferulic acid copolymer (PMAA-FA) by free radical polymerization was developed by employing a hydrogen peroxide-ascorbic acid pair to produce hydroxyl radicals acting as radical initiators. FT-IR spectra were performed to verify the insertion of ferulic acid into the polymeric chain, and the antioxidant activity of PMAA-FA was compared to that of a control polymer synthesized in the absence of antioxidant molecule. Good antioxidant activity was demonstrated by obtained materials, showing the efficiency of the polymerization method. This material could be useful in the pharmaceutical field and in the food industry (food packaging).  相似文献

A molecularly imprinted polymer able to recognize melamine in partially aqueous medium was synthesized using methacrylic acid as functional monomer and ethylene glycol dimethacrylate as cross-linking agent. The bound specificity and selectivity of the obtained material were verified by performing binding experiments with melamine and its structural analogue, 2,4,6-trimethoxy-1,3,5-triazine, respectively, using different aqueous binding media. Finally, the ability of MIP to selectively extract melamine from two real samples, a food supplement and a freeze-dried meat sample, was demonstrated.  相似文献

Purpose

Salt marsh plants are colonising wastes from a steel plant deposited on the Coina River Banks posing a potential contamination risk to the Tagus estuary ecosystem. The objectives of this study were to assess the uptake, accumulation and translocation of hazardous elements/nutrients in three spontaneous halophytic species, to evaluate the capacity of Tamarix africana to stabilise a contaminated salt marsh soil, and to evaluate the ecotoxicity of the pore water and elutriates from phytostabilised soils.

Materials and methods

The work comprises the following: fieldwork collection of soil samples from Coina River (an affluent of Tagus River) bank landfill, estuarine water and spontaneous plants (Aster tripolium, Halimione portulacoides and Sarcocornia sp.), and greenhouse studies (microcosm assay) with T. africana growing in one landfill salt marsh soil, for 97 days, and watered with estuarine water. Soils were analysed for pH, EC, Corganic, NPK, iron and manganese oxides. Soils total (acid digestion) elemental concentrations were determined by ICP/INAA. Estuarine waters, plants roots and shoots (acid digestion), soils available fraction (diluted organic acids extraction-RHIZO or pore water), and salts collected from the T. africana leaves surface were analysed for metals/metalloids (ICP-MS). Ecotoxicity assays were performed in T. africana soil elutriates and pore waters using Artemia franciscana and Brachionus plicatillis.

Results and discussion

Soils were contaminated, containing high total concentrations of arsenic, cadmium, chromium, copper, lead and zinc. However, their concentrations in the available fraction were <4 % of the total. The estuarine waters were contaminated with cadmium, but negligible ecotoxicological effect was observed. The spontaneous plants had significant uptake of the above elements, being mostly stored in the roots. Elemental concentrations in the shoots were within the normal range for plants. These species are not hazardous elements accumulators. Tamarix africana was well adapted to the contaminated saline soils, stored the contaminants in the roots, and had small concentrations of hazardous elements in the shoots. Excretion of hazardous elements by the salt glands was also observed. Elutriates from soils with and without plant did not show ecotoxicity.

Conclusions

The salt marsh species play an important role in the stabilisation of the soils in natural conditions. Tamarix africana showed potential for phytostabilisation of saline-contaminated soils. The low translocation of the elements from roots to shoots and/or active excretion of the elements by the salt glands was a tolerance mechanism in T. africana.

The natural regeneration of non-serotinous Spanish black pine (Pinus nigra Arn. ssp. salzmannii) is known to be null or limited after severe wildfires. However, it remains challenging to define efficient management strategies within the burnt area, which can help to increase the post-fire resilience of this species. We conducted a direct seeding experiment during 2011–2015 to assess the effect of different post-fire management treatments (control, soil ripping, woodchips, and soil ripping?+?woodchips) performed in opposite slope-aspects (north- and south-facing) on pine emergence, seed predation, survival, growth, and biomass allocation during the early stages of seedling development (1–5 years) after a severe wildfire. Our results showed that the slope-aspect did not have an important effect in both the seedling emergence and seed predation rates. In contrast, the influence of slope-aspect was significant as both survival and growth of pine seedlings were reduced at the south-facing slope-aspect compared to the north-facing slope-aspect due to harsher environmental conditions. A variable and reduced impact on seedling emergence and seed predation rates was induced by both soil ripping and woodchips treatments. Overall, post-fire management treatments helped to enhance survival rates during early growth stages, although this effect was short-lasting. This knowledge can be used to define post-fire seeding management strategies oriented to enhance the resilience of severely burnt pine forest stands; therefore, while both the soil ripping and woodchips treatments might not be practices leading to increase their post-fire regeneration, direct seeding at north-facing slope-aspects could be a more appropriate management strategy.  相似文献

Intact whole native AMF communities occurring across a 100-m-long field were used for the evaluation of plant performance, as determined by the actual fungal species colonizing host roots. The soil from distinct plots within a “hot spot” field was collected to set up 54 experimental units where three different plant species were grown, in order to test whether the whole native AMF communities were able to differentially affect plant growth, to assess the genetic identity of the AMF actually colonizing the tested plants and to analyse their community composition in the different hosts. Molecular analyses revealed that plant growth and nutrition of the crop plants were differentially affected by the diverse native arbuscular mycorrhizal communities colonizing the roots of the three plants, whose performance varied depending on the identity of plant hosts and fungal symbionts, more than on a rich and diversified AMF community. Such results, improving our understanding of AMF distribution at the local scale, represent a starting point allowing the selection, isolation and characterization of the most efficient AMF assemblages to be used as inoculants in sustainable food production systems.  相似文献

This paper assesses the implementation of four selected IWRM principles in four very different river basins in Europe and Asia. The four principles relate to all the different aspects of sustainable development—environmental, social, economic and institutional—as well as the factor that is particularly crucial in many countries of the South: implementation capacity. The paper is based on the work performed in the EC-funded STRIVER project, “Strategy and methodology for improved IWRM—An integrated interdisciplinary assessment in four twinning river basins”. The four basins—Tungabhadra and Sesan (in Asia), and Tagus and Glomma (in Europe) exemplify very different problems and challenges with regard to IWRM: different levels of socio-economic development and very varying problems with regard to water quality and availability. The paper shows that the implementation of IWRM is at a fairly early stage in all the four STRIVER basins; and that successful implementation of water resources is dependent not only on the existence of relevant policies, but also the degree to which laws and policies are in fact implemented.  相似文献
