Coculture is a productive technique to trigger microbes’ biosynthetic capacity by mimicking the natural habitats’ features principally by competition for food and space and interspecies cross-talks. Mixed cultivation of two Red Sea-derived actinobacteria, Actinokineospora spheciospongiae strain EG49 and Rhodococcus sp. UR59, resulted in the induction of several non-traced metabolites in their axenic cultures, which were detected using LC–HRMS metabolomics analysis. Antimalarial guided isolation of the cocultured fermentation led to the isolation of the angucyclines actinosporins E (1), H (2), G (3), tetragulol (5) and the anthraquinone capillasterquinone B (6), which were not reported under axenic conditions. Interestingly, actinosporins were previously induced when the axenic culture of the Actinokineospora spheciospongiae strain EG49 was treated with signalling molecule N-acetyl-d-glucosamine (GluNAc); this finding confirmed the effectiveness of coculture in the discovery of microbial metabolites yet to be discovered in the axenic fermentation with the potential that could be comparable to adding chemical signalling molecules in the fermentation flask. The isolated angucycline and anthraquinone compounds exhibited in vitro antimalarial activity and good biding affinity against lysyl-tRNA synthetase (PfKRS1), highlighting their potential developability as new antimalarial structural motif.  相似文献

This study used a field experiment and the means-end chain approach to examine the effect of providing information on the biofortification process on consumers' acceptance of orange-fleshed sweetpotato (OFSP). Negative, positive, and neutral information about the biofortification were verbally provided. Images were used to reinforce the verbal information. Meaning representation (mental models) of various constructs relating to OFSP was assessed. We found that the structure of mental constructs differed depending on the type of information provided and concluded that the type of information consumers receive about the biofortification process affects OFSP acceptance. Implications of the findings for policy and development practice are discussed.  相似文献

CASE DESCRIPTION: A 2-year-old Siberian Husky was evaluated because of a 2-week history of coughing and gagging and decreased appetite and activity level. CLINICAL FINDINGS: Radiography, surgery, and immunohistochemical examination revealed a solitary sclerosing mesothelioma extending from the left thoracic diaphragmatic surface that was adherent to the pericardium and the caudal mediastinum. TREATMENT AND OUTCOME: The tumor was resected along with most of the left hemidiaphragm, and the left transversus abdominis muscle was used to reconstruct the diaphragm. The 13th rib formed the base of the muscle flap. The muscle flap was transposed into the defect so that the mesothelium-lined surface faced the thoracic cavity and the deep aspect of the muscle formed the abdominal surface of the diaphragm. To minimize risk of adhesions, the exposed raw aspect of the abdominal surface was covered with porcine small intestinal submucosa. Recovery was uncomplicated, and the dog's appetite and activity level soon returned to normal. Evaluation 54 days after surgery revealed 2 subcutaneous masses on the thorax and masses in the liver and both kidneys; histologic and immunohistochemical analyses revealed metastasis of the original tumor. The dog was euthanatized. CLINICAL RELEVANCE: Hemidiaphragmatic reconstruction with a transversus abdominis muscle flap after resection of a diaphragmatic tumor was successful. The muscle flap was easily harvested and transposed into the diaphragmatic defect.  相似文献

A study was undertaken to investigate and mitigate the risk from zoonotic Cryptosporidium associated with dairy farming in Dagoretti division, Nairobi, Kenya. Outcome mapping (OM), a relatively new tool for planning and evaluation, was used to foster and then monitor changes in farmer management of health risks. Elements of the OM framework, including the vision, mission and expected progress markers, were developed in participatory sessions and a set of progress markers was used for monitoring behaviour change in farmers participating in the project (the boundary partners). Behaviour change (the outcome challenge) was supported by a range of awareness and educational campaigns, working with strategic partners (extension agents and administrative leaders). The farmers the project worked with made considerable progress according to the markers; they demonstrated an understanding of cryptosporidiosis, established or maintained clean and well drained cattle sheds, and took conscious effort to reduce possible infection. Farmers who did not participate in the project (non-contact farmers) were found to be less advanced on the progress marker indicators. Non-contact farmers who carried out risk-reducing practices had done so independently of the project team. The administration leaders, as strategic partners, had a positive attitude towards the project and confidence in their ability to support project objectives. The study demonstrates the utility of OM in helping to identify and support behavioural change.  相似文献

The effects of drip irrigation on the yield and crop water productivity responses of four tea (Camellia sinensis (L.) O. Kuntze) clones were studied four consecutive years (2003/2004-2006/2007), in a large (9 ha) field experiment comprising of six drip irrigation treatments (labelled: I1-I6) and four clones (TRFCA PC81, AHP S15/10, BBK35 and BBT207) planted at a spacing of 1.20 m × 0.60 m at Kibena Tea Limited (KTL), Njombe in the Southern Tanzania in a situation of limited water availability. Each clone × drip irrigation treatment combination was replicated six times in a completely randomized design with 144 net plots each with an area of 72 m2. Clone TRFCA PC81 gave the highest yields (range: 5920-6850 kg dried tea ha−1) followed by clones BBT207 (5010-5940 kg dried tea ha−1), AHP S15/10 (4230-5450 kg dried tea ha−1) and BBK35 (3410-4390 kg dried tea ha−1) and drip irrigation treatment I2 gave the highest yields, ranging from 4954 to 6072 kg dried tea ha−1) compared with those from other treatments (4113-5868 kg dried tea ha−1). Most of these yields exceeded those (4200 kg dried tea ha−1) obtained from overhead sprinkler irrigation system in Mufindi also Southern Tanzania, and Kibena Estate itself. Results showed that drip irrigation of tea not only increased yields but also gave water saving benefits of up to 50% from application of 50% less water to remove the cumulative soil water deficit (treatment I2), and with labour saving of 85% for irrigation. The yield of dried tea per mm depth of water applied, i.e., “the crop water productivity” for drip irrigation of clones TRFCA PC81, BBT207 and BBK35, in 2003/2004 for instance, were 9.3, 8.5 and 7.1 kg dried tea [ha mm]−1, respectively. The corresponding values in 2004/2005 were 2.7, 4.5 and 2.0 kg dried tea [ha mm]−1 while the yield responses from clone AHP S15/10 were linear decreasing by 1 and 1.6 kg dried tea [ha mm]−1 in 2003/2004 and 2004/2005, respectively. In 2005/2006 the crop water productivity from clones TRFCA PC81, AHP S15/10, BBK35 and BBT207 were 4.5, 0.4, 5.2 and 6.9 kg dried tea [ha mm]−1, respectively with quadratic yield response functions to drip irrigation depth of water application. The results are presented and recommendations and implications made for technology-transfer scaling-up for increased use by large and smallholder tea growers.  相似文献

The cation exchange capacity (CEC) of three benchmark soils in the rain forest region of Southwestern Nigeria, were measured by three standard CEC methods. Results obtained were compared with a view to selecting the best suited CEC method for the soils. The study sites were the Teaching and Research Farm of Obafemi Awolowo University, Ile-Ife, and Itagunmodi settlement. Two representative soil profile pits each were established in soils developed in coarse-grained granite and gneiss, fine-grained biotite gneiss and schist, and the amphibolite. The three methods gave different CEC values in the order of CEC-pH 8.2 > CEC-pH 7 > ECEC. There was a significant correlation between soil organic matter (SOM) and the CEC obtained by each of the three methods (P ≤ 0.05), meanwhile, total clay showed no significant correlation. ECEC provided the best estimate of the CEC for the soils and adequate SOM management is crucial to enhance sustainable productivity of the soils.  相似文献
